Gene expression profiles of the egg and synergid cell in rice

ABSTRACT: To reveal comprehensive gene expression profiles and their characteristics of the rice egg and synergid cell, we performed cell type-specific microarray analysis using nonenzymatic cell isolation technique. 40 and 50 genes expressed specifically in the egg cell and synergid cell were identified by cluster analysis using microarray results including that of control samples. Furthermore, we found that the differences in the gene expression profiles of the egg cell and the synergid cell were more extensive than previously realized, indicating that these reproductive cells are specified during the short period encompassing the three sequential mitotic nuclear divisions after generation of the megaspore.

Genes dominantly expressed in germinating pollen

Project description:In sexual reproduction, a proper communication and cooperation between male and female organs and tissue are essential for male and female gametes to unite. In flowering plants, female sporophytic tissues and gametophyte direct a male pollen tube towards an egg apparatus, which consists of an egg cell and two synergid cells. The cell-cell communication between the pollen tube and the egg apparatus, such as the reception of a signal from the egg apparatus at the pollen tube, makes the tip of pollen tube rapture to release the sperm cell. To isolate male factors involved in the interaction between a pollen tube and an egg apparatus, we focused on receptor-like kinases (RLKs), which are extensively diversified in the flowering plant lineage to comprise a large monophyletic gene family. Approximately 620 members were found in the Arabidopsis thaliana genome. Expression patterns of 558 RLKs were analyzed using an Affymetrix ATH1 microarray of A. thaliana. We focused on two RLKs, ANXUR1 (ANX1) and ANXUR2 (ANX2), and characterized their function. Here we report that pollen tubes of anx1/anx2 ruptured before arriving at the egg apparatus, suggesting that ANX1 and ANX2 are male factors controlling pollen tube behavior with directing rupture at proper timing. Furthermore, ANX1 and ANX2 were the most closely related paralogs to a female factor FERONIA/SIRENE controlling pollen tube behavior expressed in synergid cells. Our finding shows that the coordinated behaviors of female and male reproductive apparatuses are regulated by the sister genes, whose duplication might play a role in the evolution of fertilization system in flowering plants.

Transcriptomic profiling of egg quality in seabass (Dicentrarchus labrax)

Project description:The aim of the project was to identify differently expressed genes in eggs of European seabass (Dicentrarchus labrax) characterized by different quality. In this way it was expected to identify genes possibly being a molecular indicator of egg quality in this species, which was never studied to date. For the study microarray analysis of over 26 thousand genes in 16 egg batches was performed. Additionally, for each egg batch biological quality was determined, what allowed to compare the gene expression profile with overall egg quality (divided into two groups representing ‘high’ and ‘low’ egg quality). The analysis allowed to identify 39 differently expressed genes between the two groups representing ‘high’ and ‘low’ egg quality. From those genes, expression level of 7 were verified by real-time qPCR which confirmed significant difference in expression in 5 of them.

Comparative gene expression in synergid cells and antipodal cells at anthesis

Project description:Synergid cells represent the receptor cell type of angiosperm gametophytes that attract and receive the male pollen tube and are implicated in the transfer of the sperm cells to the target egg and central cells prior to double fertilization. The antipodals represent an accessory cell type that typically degenerates after fertilization; in rice it may initially have a short-lived nutritional role prior to endosperm establishment. We examined the transcriptome of these two cell types using Oryza sativa ssp. japonica (cv Katy) using the Affymetrix 57K rice genome GeneChip to provide an overview of genes activated at receptivity in the synergid and antipodal cells and transcript up- and down-regulated at female gametophyte maturity just prior to fertilization. This is part of a larger microarray paroject to examine the global program of gene expression during sexual reproduction in Oryza sativa which serves as a model angiosperm in which male and female gametophytic and embryonic cells may be readily isolated and characterized.

Cell-type specific transcriptome analysis to study germline development and reproduction in apomictic Boechera gunnisoniana- ATH1 arrays

Project description:Apomixis differs from sexual reproduction only in three major aspects: While the sexual megaspore mother cell undergoes meiosis, the apomictic initial cell omits or aborts meiosis (apomeiosis); the unreduced egg cell of apomicts forms an embryo without fertilization (parthenogenesis), and formation of functional endosperm requires specific developmental adaptations. Currently, our knowledge about the gene regulatory programs underlying apomixis is scarce. We used the apomict Boechera gunnisoniana, a close relative of Arabidopsis thaliana, to investigate the transcriptional basis underlying apomeiosis and parthenogenesis. Here, we present the first comprehensive reference transcriptome for reproductive development in an apomictic species. To compare sexual and apomictic development at the cellular level, we then used a combination of laser-assisted microdissection with microarray and RNA-Seq analysis. Our study yields important new insights into the transcriptional basis underlying apomixis. Cell-type specific transcriptome profiles were generated from the apomictic initial cell (2 biological replicates), surrounding sporophytic tissues (sporo_nucellus; 2 biological replicates), egg cell, central cell and synergid cells (one sample each) from the triploid pseudogamous obligate apomict Boechera gunnisoniana by heterologous hybridization on the Affymetrix ATH1 arrays.

Egg quality after natural and controlled ovulation

Project description:The present study aimed at studying the rainbow trout egg transcriptome using 9152-cDNA microarrays after natural or controlled ovulation. The analysis of egg transcriptome after natural or controlled ovulation led to the identification of 26 genes. We observed that both hormonal induction and photoperiod control of ovulation induced significant changes in the egg mRNA abundance of specific genes. We demonstrate that hormonal induction of ovulation has an impact on the egg mRNA abundance of specific genes even though the resulting effects on the developmental potential of the egg is so far unknown. Project description:We used single cell laser-assisted microdissection (LAM) to perform egg cell transcriptome analysis for comparison with available expression profiles of vegetative tissues and male reproductive organs. We observed an overlap in gene activity between bryophyte and angiosperm egg cells, but also clear differences. Strikingly, several processes that are male germline-specific in Arabidopsis are active in the P. patens egg cell. Among those were the moss PbBNB genes and using reporter lines and knock-out mutants we could show that they control proliferation and identity of both female and male germlines.
