Effect of TDB and PEG-TGL treatment on gene expression in BMDC from C57BL/6 mice

ABSTRACT: There is much demand for more water-soluble Mincle ligands which is promissing adjuvants. We synthesized water-soluble Mincle ligad, PEG-TGL, and assessed its potential on adjuvanticity. Comparing to TDB, known ligand for Mincle, PEG-TGL induced less IL-1beta secretion. To elucidate the gene expression differenses leads to IL-1beta secretion, we performed RNA-seq analysis using bone marrow-derived dendritic cells, and revealed that PEG-TGL were less able to active phagocytic pathways.

Project description:In order to better understand the context of root water stress and its relationship to other stresses, I undertook an evaluation of osmotic water stress by transcriptome analyses. Whole transcriptome MARSeq (Diego Adhemar Jaitin, 2017) analysis can enable us to study the changes in gene expression (changes in transcript prevalence) in depth under different treatments. It is a cost effective method to develop transcriptional activity. It uses relatively small amounts of RNA and reads sequences from the 3’-polyA containing end. This method can reveal underlying trends that can help us understand the less obvious processes happening in our system. To carry this out I processed whole transcriptome data separately from WT seedling roots exposure to PEG, and from roots under 30% PEG + 5mM HIS, 30% PEG + 1mM SHAM for 0, 1, 2, 3 h.

Expression profiling of PEG induced water stress responsive genes in Eucalyptus grandis

Project description:The study was conducted to identify differentially expressed polyethylene glycol (PEG) induced water stress responsive genes in E. grandis. Forty day old rooted cutting of E. grandis was subjected to -0.225 MPa PEG treatment and total RNA was isolated from leaves of water treated control and PEG treated samples after three hours of treatment. The differential expression of water stress responsive genes was analyzed using microarray technique.

Gene expression in chicken embryo liver

Project description:We have profiled the gene expression in eight chicken embryo livers in the 11th day of incubation. Four eggs (labelled as WAT1-4) were injected with 0.15 ml/egg deionized water in days 9, 10 an 11 post incubation while four eggs (labeled CTR1-4) were kept as environmental controls. We found that water injections had a very small effect, regulating less than 1% of the genes. Both WAT and CTR data sets were used as references in further experiments for eggs expossed to various water-soluble carcinogenic compounds.

Mycobacterial cord factor reprograms the macrophage response to IFN-gamma towards enhanced inflammation yet impaired antigen presentation and expression of Gbp1

Project description:Mycobacteria can survive and persist in macrophages despite triggering multiple pattern recognition receptors and strong T cell-derived IFN-gamma production. The mycobacterial cord factor trehalose-6,6-dimycolate binds the C-type lectin receptor Mincle and induces inflammatory gene expression through the Syk-Card9 pathway. It is well established that TDM and IFN-gamma synergize to induce expression of iNOS. However, the cord factor may also act to down-regulate IFN-gamma-induced responses and contribute to mycobacterial immune evasion. Here, we have investigated the cross-regulation of the macrophage transcriptome by IFN-gamma and by TDM, as well as its synthetic analog TDB. For genome-wide expression profiling, bone marrow derived macrophages (BMMs) from female C57BL/6 mice aged 6 to 8 weeks were used for stimulation.
