Identifying signatures of EV secretion in metastatic breast cancer through functional single-cell profiling

ABSTRACT: Extracellular vesicles (EVs) regulate the tumor microenvironment by facilitating transport of biomolecular cargo including RNA, protein, and metabolites. We developed an integrated method to identify individual cells with differences in EV secretion and linked single-cell RNA-sequencing on cloned single cells from MDAMB231 cell line. Additionally, We profiled parental MDAMB231, MCF7 and HCC70 cell lines.

Variability of bovine milk EV measurements in health and subclinical mastitis

Project description:Mastitis, the inflammation of the mammary gland, is one of the most prevalent diseases in dairy farming worldwide. Unfortunately, the disease is most often present in a subclinical type with no clear symptoms. The sooner the infection is detected, the less opportunities for the disease to progress and the more treatment options remain available. Milk microRNA (miRNA) encapsulated in extracellular vesicles (EV) have been proposed as potential biomarkers of different mammary gland conditions, including subclinical mastitis. However, little is known about the robustness of EV analysis regarding sampling time-point or natural infections. In order to estimate the reliability of EV measurements in raw bovine milk, we first evaluated the changes in EV size, concentration and miRNA cargo during three consecutive days. Then, we compared milk EV differences from natural infected quarters with high somatic cell count (SCC) with their healthy adjacent quarters with low SCC and quarters from uninfected udders. We found that milk EV miRNA cargo is very stable along three days and that infected quarters do not induce relevant changes in milk EV of adjacent healthy quarters, making them suitable controls. We observed cow-individual changes in immunoregulatory miRNA in quarters with chronic subclinical mastitis, pointing towards infection-specific alterations. Finally, we proposed bta-miR-223 as a potential indicator of subclinical mastitis prognosis in raw milk.

Project description:Extracellular vesicles (EVs) are nanosized cell-derived vesicles found in all bodily fluids which provide a route of intercellular communication by transmitting biological cargo. While EVs offer promise as therapeutic agents, the molecular mechanisms of EV biogenesis are not yet fully elucidated, in part due to the concurrence of numerous interwoven pathways which give rise to heterogenous EV populations in vitro. The effects of conditions in the cellular environment on EV production are of particular interest. In this study, we utilize a quantifiable EV-engineering approach to investigate how various cell media conditions alter EV production. The presence of serum, exogenous EVs, and other signaling factors in cell media alters EV production on physical, molecular, and transcriptional levels. Further, we demonstrate that exosome biogenesis pathways are the major factors contributing to EV production under optimized conditions. Our findings suggest a novel understanding to the mechanisms underlying EV production in cell culture which can be applied to develop advanced EV production methods.

Deciphering the ciliary extracellular vesicle (EV) proteome

Project description:Cilia are conserved EV shedding organelles that release ciliary EVs for transmitting 13 signals. However, the content of ciliary EVs produced by living animals has not been explored. 14 Here we used evolutionarily conserved PKD-2::GFP EV cargo of Caenorhabditis elegans as a 15 marker of ciliary EVs to identify animal EV proteome. Top candidates were experimentally 16 confirmed using fluorescent reporter tracking and live animal imaging. Our findings indicate that 17 cilia produce multiple EVs with distinct protein content and that EVs from different neuronal 18 types carry different EV cargo. We discover that ciliary EVs carry nucleic acid binding proteins, 19 implying the role of cilia in epigenetic regulation and transfer of genetic information in trans. 20 Our dataset also contains non-ciliary EV cargo candidates from other tissues, such as cuticle, 21 somatic gonad, and germline, that can be explored using our MyEVome Web App.
