Project description:Histone lysine lactylation (Klac) is a new posttranslational modification (PTM) that is installed by acetyltransferase to modulate specific immune responses1 and oncogenesis2,3. Akkermansia muciniphila (A. muciniphila) is a beneficial bacterium that blunts ulcerative colitis (UC) in a mouse model by secreting extracellular vesicles (SEVs)4. Although many histone sites are known to contain lysine lactylation, whether this modification is regulated to modulate specific biological functions by exogenous acetyltransferase or intestinal microbiota is not well understood. Here, we discovered that SEV from A. muciniphila, rather than A. muciniphila per se, has anti-Th17 (T helper 17) differentiation activity. We further screened the composition of SEV and found that Amuc_2172 is the key active component. As a GCN5-related acetyltransferase of A. muciniphila, Amuc_2172 is accessible to naïve T cells and functions as a lactylation transferase on Lys4 of histone H3. Accelerated histone H3 lactylation (H3Klac) on Lys4 competitively blocks trimethylation (H3Kme3) on Il17a loci in the process of Th17-cell differentiation. Additionally, intraperitoneal application of recombinant Amuc_2172 also inhibited Th17 and dextran sulfate sodium (DSS)-induced UC phenotypes in vivo; moreover, bioengineered Amuc_2172 showed improved colitis site delivery and higher Th17 inhibition potential compared to Amuc_2172 alone. Our study reveals not only a potential therapeutic strategy for treating colitis but also a model via which lysine lactylation is regulated by the intestinal microbiota, which may be broadly applicable to understand the crosstalk of bacteria and immunity.

Project description:Previous studies have implicated a causal role for the gut bacterium Akkermansia muciniphila in counteracting diet-induced obesity and metabolic dysfunctions. However, a systems level understanding of the molecular mechanisms underlying the anti-obesogenic effect of A. muciniphila is lacking. Using fructose-induced obese mice as a model, we carried out multiomics studies to investigate the molecular cascades mediating the effect of A. muciniphila. We found that A. muciniphila colonization in fructose-induced obese mice triggered significant shifts in gut microbiota composition as well as alterations in numerous gut and plasma metabolites and gene expression in the hypothalamus. Among these, we found that the metabolite oleoyl-ethanolamide in the gut and circulation and hypothalamic oxytocin are the key regulators of gut-brain interactions that underlie the A. muciniphila anti-obesity effect. Our multiomics investigation elucidates the molecular regulators and pathways involved in the communication between A. muciniphila in the gut and hypothalamic neurons that counter fructose-induced obesity .

Project description:The use of Akkermansia muciniphila as potential therapeutic intervention is receiving increasing attention. Health benefits attributed to this bacterium include an improvement of metabolic disorders and exerting anti-inflammatory effects. The abundance of A. muciniphila is associated with a healthy gut in early mid- and later life. However, the effects of A. muciniphila on a decline in intestinal health during the aging process are not investigated yet. We supplemented accelerated aging Ercc1-/Δ7 mice with A. muciniphila for 10 weeks and investigated histological, transcriptional and immunological aspects of intestinal health. The thickness of the colonic mucus layer increased about 3-fold after long-term A. muciniphila supplementation and was even significantly thicker compared to mice supplemented with Lactobacillus plantarum WCFS1. Colonic gene expression profiles pointed towards a decreased expression of genes and pathways related to inflammation and immune function, and suggested a decreased presence of B cells in colon. Total B cell frequencies in spleen and mesenteric lymph nodes were not altered after A. muciniphila supplementation. Mature and immature B cell frequencies in bone marrow were increased, whereas B cell precursors were unaffected. These findings implicate that B cell migration rather than production was affected by A. muciniphila supplementation. Gene expression profiles in ileum pointed toward a decrease in metabolic- and immune-related processes and antimicrobial peptide production after A. muciniphila supplementation. Besides, A. muciniphila decreased the frequency of activated CD80+CD273- B cells in Peyer’s patches. Additionally, the increased numbers of peritoneal resident macrophages and a decrease in Ly6Cint monocyte frequencies in spleen and mesenteric lymph nodes add evidence for the potentially anti-inflammatory properties of A. muciniphila. Altogether, we show that supplementation with A. muciniphila prevented the age-related decline in thickness of the colonic mucus layer and attenuated inflammation and immune-related processes at old age. This study implies that A. muciniphila supplementation can contribute to a promotion of healthy aging.
