Project description:Mycotoxin citrinin (CTN) is a secondary metabolite of fungi, becoming a contaminant widely found in foods, feeds, and fermented health supplements. CTN is known to disrupt microtubule and chromosome arrangement at high dose (50 - 150 μM), but the toxicological effect of CTN long-term exposure has not been clearly studied. To investigate the molecular mechanisms of genotoxic, clastogenic, and carcinogenic effects of CTN, RNA-seq was performed on HEK293 cells exposed to chronic 20 μM CTN treatment (3 days for short-term and 30 days for long-term). The transcriptomic profile may reveal some underlying mechanisms regarding chronic carcinogenic potential of CTN, providing information for risk assessment of CTN-contaminated grains and its commercial food products.

Project description:Mycotoxin citrinin (CTN) is widely found in multiple types of grains in foods and feeds globally. CTN also contaminates Monascus-derived health supplements such as red yeast rice and red yeast extracts during the fermentation process, which are originally used for preventing cardiovascular diseases. A previous study has reported that CTN is cardiotoxic to zebrafish embryos during their development by interfering some cardiogenic genes and pathways, showing a potential risk of consuming CTN-contaminated foods and products. However, the cardiotoxic effects and the underlying mechanisms of CTN on mammalian cardiomyocytes remain unclear and need to be ellucidated. In this study, we performed RNA-seq experiments in order to investigate the transcriptomic alterations induced by CTN-exposed rat H9c2 cardiomyocytes. The transcriptome profiling we obtained may reveal some evidence regarding the toxic effects of CTN on cardiac phenotypes, chromosome segregation, tubulin arrangement, mitochondrial functioning, and stress responses, etc.

However, the enzymes which is responsible for the degradation process was unknown. Transcriptome change in response to mycotoxin citrinin was analyzed. The molecular mechanism of Cryptococcus podzolicus Y3 withstand citrinin was revealed.

This natural chemicals is one of the well characterized mycotoxin but the information especially mechanism of toxic action is limited. We used two types of microarrays, one have ORF (Open reading Frame) fragment on the surface of the glass as probes and the other have probes as oligonucleotide probes on the microarray with 3 dimensions. We compared data properties and studied the toxicity of citrinin to yeast cells. Project description:We report the transcriptome and miRNAome of human embryonic kidney 293 cells upon the administration of ochratoxin A (OTA) and citrinin (CTN), to compare the individual and combined toxicity of these two mycotoxins. We identified largely differentially expressed miRNAs and corresponding genes, and the correlations between miRNAs and their target genes associated with apoptotic signaling were furthermore analyzed by pmi-RB-REPORT luciferase assay system. We concluded that hsa-miR-1-3p plays an essential role in promoting OTA and CTN-induced renal cytotoxicity.

Project description:From a long time ago, supplementation of fermented enzyme foods could have worked health effects on the body in the east nevertheless, only a few studies reported functions of them such as weight loss and metabolic syndrome. Thus, it is necessary to be verified whether supplementation of fermented enzyme foods can act in the body as a functional material. Therefore, we investigated the anti-obesity effects of fermented mixed grain with digestive enzymes (FMG) in high-fat diet induced mice. Sixty C57BL/6J mice were divided into six dietary groups and fed a normal diet (ND), a high-fat diet (HFD), Bacilus Coagulans group, steamed grain group, low-dose fermented mixed grain group(L-FMG), high-dose fermented mixed grain group (H-FMG) supplement for 12 weeks. After sacrificing, body weight and body fat mass in H-FMG group were significantly decreased compared to HFD group with a simultaneous decrease in plasma lipids. Also, H-FMG significantly decreased the blood glucose and improved the glucose tolerance compared to HFD group. Moreover high-dose FMG supplementation dramatically decreased the levels of inflammatory cytokines which secreted from adipocyte. Taken together, our findings suggest that H-FMG ameliorate high fat-diet induced obesity and its complication and could be used as a potential preventive agent for obesity.
