Chemokines profiling in tumors treated with UNC1999

ABSTRACT: Increased expression of chemokines in UNC1999-treated tumor tissues were successfully checked out through chemokines profiles screening of a total 25 different mouse chemokines using control and UNC1999-treated tumor tissues.

Project description:C57BL/6 mice were subjected to sham surgery or orthotopic Hepa1-6 hepatic tumor inoculation. Spleen tissues from mice subjected to sham surgery (control mice), mice bearing a 2-week orthotopic hepatoma, and mice bearing a 4-week othotopic hepatoma were collected. Total RNA was extracted from the spleen tissues,and we used the RT² Profiler PCR array-mouse chemokines & receptors (SABioscience) array panel to quantitate gene expression of chemokine and receptor genes from the spleens.

The effects of chemokines CXCL1/2/8 on mouse lung fibroblasts

Project description:Our preliminary data show that the primary triple-negative breast tumor-derived chemokines CXCL1/2/8 promote migration and tube formation of mouse lung endothelial cells, which is mediated by lung fibroblasts. To explore the molecular events underlying this mediator function of lung fibroblasts, we treated freshly-isolated lung fibroblasts with chemokines CXCL1/2/8 and performed RNA-Seq assay.

p38α activation-mediated IFNAR1 downregulation by tumor derived factors induced neutrophil-attracting chemokines production

Project description:Tumor derived factors induced type I interferon receptor (IFNAR1) downregulation both in vivo and in vitro. IFNAR1 was shown to be phosphorylated, ubiquitinated and downregulated in response to soluble or vesicular tumor derived factors in a p38-dependent manner. Upon administration of B16F10 tumor derived factors, lungs of Ifnar1S526A knock-in mice (termed "SA"), where mutation in a phospho-acceptor site within the IFNAR1 phospho-degron renders IFNAR1 insensitive to p38-driven phosphorylation, ubiquitination and degradation, retained IFNAR1 and sustained the expression of the IFN stimulated genes. Futuremore, careful examination and validation of gene expression profiles revealed an increased expression of several chemokines known to attract neutrophils (including Cxcl1, Cxcl3 and Cxcl5) in the lungs of WT mice treated with tumor derived factors. This increase was notably less pronounced in the SA mice, suggesting that IFNAR1 downregulation by tumor derived factors induced neutrophil-attracting chemokines production.

Cooperation between Constitutive and Inducible Chemokines Enables T-cell Engraftment and Immune Attack in Solid Tumors

Project description:We investigated the role of chemokines in regulating T-cell accumulation in solid tumors. CCL5 and CXCL9 overexpression was associated with CD8+ T-cell infiltration in common solid tumors. T-cell infiltration required tumor cell-derived CCL5 and was amplified by IFN-dependent, myeloid cell-secreted CXCL9. Accordingly, CCL5-CXCL9 co-expression revealed immunoreactive tumors with prolonged survival and response to checkpoint blockade. Loss of CCL5 expression in human tumors was associated with epigenetic silencing through DNA methylation. Reduction of CCL5 expression caused TIL desertification whereas forced CCL5 expression prevented Cxcl9 and TIL loss and attenuated tumor growth in mice through IFN. The cooperation between tumor-derived CCL5 and IFN-inducible CXCR3 ligands secreted by myeloid cells is key for orchestrating T-cell infiltration in immunoreactive and immunoresponsive tumors.
