ABSTRACT: We subjected old (21-22 month) and young (3-4 month) female C57BL/6 mice to sham-operation as a control for 45 min transient oclusion of the middle cerebral artery and obtained the brain four days later. We obtained microglia from the ipsilateral brain hemisphere by FACS.

Project description:Intracerebral haemorrhage (ICH) is a major area of unmet clinical need. Inflammation is part of a secondary injury phase linked to both brain damage and repair. Here, we looked to identify the major molecular pathways promoting innate immune activation and recruitment in the first day of injury. To do this, we induced ICH in six month old C57BL/6 male mice (n=4) and isolated RNA from the right (ipsilateral) hemisphere 24 hours later. We used RNA isolated from right hemispheres of six month old naive male C57BL/6 mice (n=3) as controls.

RNA-Seq transcriptome comparison of microglia and CD11c+ cells sorted from the ischemic brain tissue of parabiotic (female) mice

Project description:RNA-Seq transcriptome comparison of the following cell populations (n=3-4 independent samples per cell population): a) CD11c-eYFP+ cells FACS sorted from brain of female adult mice 4 days after cerebral ischemia, b) CD11c-eYFP+ cells FACS sorted from brain of female parabiotic mice 4 days after cerebral ischemia c) CX3CR1+ microglia sorted from the ischemic brain of female CX3CR1CreERT2-ROSA26 tdTomato mice. Purpose: The goal of this study is to compare the transcriptome profile (RNA-Seq) of infiltrating cD11c-eYFP+ cells and microglia, both collected from ischemic brains of mice. Methods: RNA samples were obtained from FACS sorted eYFP+ cells of the ipsilateral brain hemisphere of CD11c-eYFP mice 4 days post-ischemia, the ipsilateral brain hemisphere of CD11c-eYFP/WT parabiotic mice 4 days post-ischemia, and from microglial cells sorted from the ipsilateral brain hemisphere of Cx3cr1CreERT2:ROSA26dTomato mice 4 days post-ischemia. NGS was performed (RNA-Seq) to compare the transcriptome of these populations. Results: the populations we compared clearly separated the differentially expressed genes in an unsupervised cluster analysis. 1509 genes were overrepresented in microglia and 1183 genes were overrepresented in CD11c-eYFP+ cells in the ischemic brain. Conclusions: Our study is the first comparative analysis of the transcriptomes of microglia and the infiltrating CD11c-eYFP+ cells derived from the ischemic brain of mice. The results show that the infiltrating CD11c-eYFP cell population in the ischemic brain tissue of parabiotic mice displays overrepresentation of genes typical of dendritic cells, immune functions, and ClassII antigen presentation, amongst others, that are not found represented in microglia.

mRNA microarray in MCAo rat model with IV-hUMSC

Project description:To evaluate the change of gene expression at cerebral infarction by the treatment of IV-human umblical cord derived mesenchymal stem cell. RNA was isolated from the ipsilateral hemisphere to MCAo in rats. At 72 h post-MCAo, the ipsilateral hemisphere subjected to MCAo was used for mRNA microarray. RNA was isolated from the ipsilateral hemisphere to MCAo in rats without MCAo (control group, n=5), rats treated with 1x106 IV-hUMSC (hUMSC group, n=6) and saline (saline group, n=5) at 24h post-MCAo.

Comprehensive gene expression analysis of cerebral cortices from mature rats after neonatal hypoxic-ischemic brain injury

Project description:Neonatal hypoxic-ischemic (HI) encephalopathy can lead to severe brain damage and is a common cause of neurological handicaps in adulthood. To elucidate the molecular events occurring in cerebral cortices of mature rats (8 weeks old) after neonatal HI brain insult, we performed comprehensive gene expression and gene network analyses using a DNA microarray system (Agilent 4x44K). A rat model of neonatal HI encephalopathy (Rice model) was obtained by unilateral ligation of the common carotid artery of 7-day-old rats with hypoxia (exposure to 8% oxygen). Due to the HI insult-related breakdown of the ipsilateral hemisphere in the brain, RNAs were prepared from the contralateral cerebral cortices of 8-week-old rats and analyzed by DNA microarray. Biofunctional analysis of differentially regulated genes revealed that many upregulated genes were related to cell death signaling, such as the arachidonic acid cascade. In contrast, many downregulated genes were related to gene expression, reflecting progressive damage by the HI insult, even within the contralateral cerebral hemisphere.

Transcriptome Analysis of the Mouse Medial Prefrontal Cortex in Chronic Constriction Injury Model

Project description:The medial prefrontal cortex (mPFC) is critical for both the sensory and emotional /cognitive components of pain. However, the underlying mechanism still remains largely unknown. Here, we examined changes in the transciptomic profiles in the mPFC of mice with chronic pain using RNA sequencing (RNA-seq) technology. We then performed gene expression profiling analysis using data obtained from RNA-seq of 4 different group of mPFC from mice, including ipsilateral part (Left side, the same to our operation part) of mPFC, contralateral part (Right side, opposite to our operation part) of mPFC after CCI (Chronic Constriction Injury of Sciatic Nerve) operation. Combining with naïve mice, at 28 days after operation.
