Gene and microRNA transcriptome analysis of Parkinson's related LRRK2 mouse models

Project description:Mutations in leucine-rich repeat kinase 2 (LRRK2) are the most frequent cause of familial and sporadic ParkinsonM-bM-^@M-^Ys disease (PD). Here, we investigated in parallel gene and microRNA transcriptome profiles of three different LRRK2 mouse models. Striatal tissue was isolated from adult LRRK2 knockout mice, as well as mice expressinghuman LRRK2 wildtype (hLRRK2-WT) or PD-associated R1441G mutation (hLRRK2-R1441G). We used microarraya to measure gene and microRNA expression levels in various LRRK2 mouse models. LRRK2 wildtype and knockout mice were bred on a C57BL/6 background. LRRK2 non-transgenic and hLRRK2 transgenic mice were bred on a FVB/N background.

Project description:The goal of this observational study is to compare anesthetic modalities (intravenous propofol anesthesia with sevoflurane gas anesthesia) in patients who underwent colorectal cancer resection surgery regarding the outcome of acute kidney injury. The main questions it aims to answer are: * is there a difference in acute kidney injury incidence in the two anesthetic modalities? * is there a difference in plasma creatinine between the two anesthetic modalities? * are there any patient characteristics or intraoperative factors that effect the incidence of acute kidney injury in either anesthetic modality? Inhibition of methylthioadenosine phosphorylase provides protection from experimental acute kidney injury

Project description:This dataset presents transcriptomic profiling of mouse kidney cortex in a model of ischemia-reperfusion injury (IRI) with and without methylthioadenosine phosphorylase (MTAP) inhibition (MTAPi). Male C57BL/6J mice were assigned to sham (n=6), IRI with vehicle (n=6), or IRI with MTAP inhibitor (MTAPi; n=5) groups. MTAPi (MT-DADMe-ImmA, AdooQ Bioscience, Cat# A13651) was administered intraperitoneally 4.5 hours before ischemia. Total RNA was extracted from kidney cortices, depleted of rRNA, and processed using a directional library prep kit. Libraries were sequenced on the Illumina NovaSeq 6000 platform (100 bp paired-end). Transcriptomic analysis revealed that MTAP inhibition reduced injury-associated gene expression, suppressed inflammatory and stress pathways, and preserved epithelial and metabolic gene programs. These data provide insight into the protective role of MTAP inhibition in acute kidney injury.
