Comparison of GNC and GNL Overexpression with ga1 and pif1345

ABSTRACT: We have used Agilent whole genome arrays (v4) to compare the gene expression changes between 5 days-old Arabidopsis ga1 mutants, pif1345 mutants (Leivar et al., 2008) and transgenic lines overexpressing the regulators GNC and GNL/CGA1 to the wild type. All mutants or transgenic lines are in the Columbia-0 background. Three independent biological samples were prepared for each genotype.

Experiment was performed in two rounds: Col-0_1, gnc gnl, quintuple and Col-0_2, triple.

Project description:To investigate the role of LLM-domain B-GATAs in starch metabolism we performed gene expression profiling analysis of RNA-seq data, obtained with Illumina technologies. B-GATA double mutant gnc gnl and the transgenic line overexpressing GNL were grown in dark conditions to avoid interference from photosynthesis or other light-dependent processes affecting sugar and starch biosynthesis.

Project description:GATA transcription factors are involved in multiple processes in plant growth and development. Two GATA factors, nitrate-inducible, carbon metabolism-involved (GNC) and CYTOKININ-RESPONSIVE GATA FACTOR 1 (CGA1, also named GNL) are important regulators in greening, flowering, senescence and hormone signaling. However, their direct target genes in regulating these biological processes are poorly characterized. Here, we report a genome-wide identification of the target genes of Arabidopsis GNC and CGA1.

Expression data from Arabidopsis wt and ga1-3, brm-1, ga1-3/brm-1 mutants

Project description:Arabidopsis brm plants depleted in a SWI/SNF-type ATPase BRM have decreased level of endogenous gibberellins and a phenotype that in many respects resembles the phenotype of mutants with repressed GA signaling or biosynthesis, like ga1-3. ga1-3/brm double mutant showed several additive and synergistic effects. To examine whether the phenotypic traits of brm, ga1-3 and ga1-3/brm lines are reflected at the gene expression level, we compared the expression profiles of brm, ga1-3, ga1-3/brm and wild-type plants using microarray analysis. Microarray analysis was performed on total RNA isolated from shoots of 18-d-old wt, brm, ga1-3, and ga1-3/brm seedlings. Three biological replicates were examined for each genotype. Plants were grown simultaneously under the same conditions.
