Project description:We have used SmartSeq2 to sequence single phenotypic skeletal stem cell (SSCs) populations purified via FACS from adult mouse long bones. Two putative SSC populations were isolated based on their previously reported surface marker profiles. An osteochondrogenic SSC (ocSSC, CD45-CD31-Tie2-Ter119-CD51+6C3-Thy1-CD105-) and a perivascular SSC (pvSSC, CD45-CD31-Pdgfra+Sca1+CD24+) were investigated.

Expression data of skeletal stem cells from young mouse spines and femurs

Project description:The spine is a remarkably specialized skeletal structure, exhibiting distinct anatomical phenotypes across different spinal regions. With this understanding in mind, we hypothesized that skeletal stem cells (mSSCs) originating from various spinal regions (cervical, thoracic, and lumbar) might display varying genotypic signatures that may dictate their skeletal tissue formation and shape. We also analyzed femur SSCs as an additional control.

Articular cartilage regeneration by activated skeletal stem cells

Project description:Osteoarthritis (OA) is a degenerative disease resulting in irreversible, progressive destruction of articular cartilage1. The etiology of OA is complex and involves a variety of factors, including genetic predisposition, acute injury and chronic inflammation2-4. Here we investigate the ability of resident skeletal stem-cell (SSC) populations to regenerate cartilage in relation to age, a possible contributor to the development of osteoarthritis. We demonstrate that aging is associated with progressive loss of SSCs and diminished chondrogenesis in the joints of both mice and humans. However, a local expansion of SSCs could still be triggered in the chondral surface of adult limb joints in mice by stimulating a regenerative response using microfracture (MF) surgery. Although MF-activated SSCs tended to form fibrous tissues, localized co-delivery of BMP2 and soluble VEGFR1 (sVEGFR1), a VEGF receptor antagonist, in a hydrogel skewed differentiation of MF-activated SSCs toward articular cartilage. These data indicate that following MF, a resident stem-cell population can be induced to generate cartilage for treatment of localized chondral disease in OA.

Investigation of TOF patient heart specimens

Project description:The protein BBLN with largely unknown function was identified in human right ventricular heart specimens from patients undergoing repair surgery for Tetralogy of Fallot (TOF). Proteins from heart specimens were solubilized and subjected to immuno-affinity enrichment with anti-CAMK2D-specific antibodies. Co-enriched proteins were separated by SDS-PAGE, and the protein band in the range of 8-14 kDa was cut and subjected to identification by nano-LC-ESI-MS/MS.

SmartSeq2 analysis of chemically-defined MiPs with and without valproic acid treatment

Project description:We have optimized a protocol to generate chemically defined induced pluripotent stem cell-derived mesodermal progenitors. We demonstrate that these cells contribute to myotube formation that can be improved by the addition of valproic acid. Thanks to SmartSeq2 single cell RNAseq, we show that valproic acid treatment increase the number of myogenic progenitors. We also used gene set enrichment analysis to annotate the clusters further with correlations to the ARCHS4 Tissues database. In particular, we found that the gene expression profile of the VPA-treated single cell cluster correlated more to myoblasts compared to the untreated cells. Some key myogenic genes found to be differentially expressed were DESMIN and CD82, which were upregulated upon VPA supplementation.

Spinal Analgesia for Colonic Resection Using an Enhanced Recovery After Surgery (ERAS) Program

Project description:This is a blinded randomized controlled trial in patients undergoing laparoscopic colon surgery. The aim of this study is to assess whether spinal analgesia with a mixture of bupivacaine and morphine provides better pain relief than systemic morphine in a group of patients undergoing colonic resection and using the Enhanced Recovery After Surgery (ERAS) program. Twenty patients will receive spinal analgesia and twenty patients will receive only Patient Control Analgesia (PCA).

Skeletal Muscle Fibrosis in Pancreatic Cancer Patients with Respect to Survival

Project description:Skeletal muscle wasting is a devastating consequence of cancer that may be responsible for nearly 30% of cancer-related deaths. In addition to muscle atrophy, we have identified significant muscle fiber damage and replacement of muscle with fibrotic tissue in rectus abdominis muscle biopsies from cachectic pancreatic ductal adenocarcinoma (PDAC) patients that associates with poor survival. Transcriptional profiling of muscle harvested from these same patients supported these findings by identifying gene clusters related to wounding, inflammation and cellular response to TGF-B upregulated in cachectic PDAC patients compared with non-cancer controls. In this dataset, we include the expression data obtained from rectus abdominis muscle biopsies fron non-cancer controls patients undergoing abdominal surgery for benign reasons and from PDAC patients undergoing tumor-resection surgery. PDAC patients were further classified as non-cachectic or cachectic. Cachexia was defined as a body weight loss of >5% during the 6 months prior to surgery. The purpose of this study was to identify the broader transcriptional networks changed in cachectic PDAC patients versus non-cancer controls, that may be associated with the histological changes observed in muscle biopsies harvested from these same patients.

Ovarian Tumor Samples: mixed reference of 106 pooled ovarian samples vs. individual patient ovarian sample

Project description:The goal of the study was to identify molecular subgroups that might predict clinical outcomes in serous epithelial ovarian cancer (EOC) patients. A second objective was to identify potential therapeutic targets for serous EOC based on improved understanding of the molecular diversity of the disease. Ovarian tissues and matched peripheral blood samples were prospectively obtained from sequential patients undergoing planned gynecologic surgery at Cedars-Sinai Medical Center between 1989 and 2005. All patients underwent surgery and received adjuvant chemotherapy with a contemporaneous standard-of-care regimen.
