Monolayer cells transcriptome of human embryonic stem cell differentiation

ABSTRACT: We examined the toxic effects of environmental pollutants on embryonic stem cell induced monolayer differentiation by RNA sequencing. RNA extraction was performed on monolayer cells exposed to multiple compounds during monolayer differentiation. The toxic effect of the compound on early embryonic development was obtained by analyzing the difference in RNA expression. Our study provides data closer to the toxic effects of human exposure to neonicotinoids, organophosphate esters and synthetic phenolic antioxidants.

Assessing developmental neurotoxicity of emerging environmental chemicals using multiple in vitro models: A comparative analysis.

Project description:Newly synthesized chemicals are being introduced into the environment without undergoing proper toxicological evaluation, particularly in terms of their effects on the vulnerable neurodevelopment. Thus, it is important to carefully assess the developmental neurotoxicity of these novel environmental contaminants using methods that are closely relevant to human physiology. This study comparatively evaluated the potential developmental neurotoxicity of 19 prevalent environmental chemicals including neonicotinoids (NEOs), organophosphate esters (OPEs), and synthetic phenolic antioxidants (SPAs) at environment-relevant doses (100 nM and 1 μM), using three commonly employed in vitro neurotoxicity models: human neural stem cells (NSCs), as well as the SK-N-SH and PC12 cell lines. Our results showed that NSCs were more sensitive than SK-N-SH and PC12 cell lines. Among all the chemicals tested, the two NEOs imidaclothiz (IMZ) and cycloxaprid (CYC), as well as the OPE tris(1,3dichloro-2-propyl) phosphate (TDCIPP), generated the most noticeable perturbation by impairing NSC maintenance and neuronal differentiation, as well as promoting the epithelial-mesenchymal transition process, likely via activating NF-κB signaling. Our data indicate that novel NEOs and OPEs, particularly IMZ, CYC, and TDCIPP, may not be safe alternatives as they can affect NSC maintenance and differentiation, potentially leading to neural tube defects and neuronal differentiation dysplasia in fetuses.

Effects of PFOA, PFBA, PFHxA, PFOS, PFBS, PFHxS on gene expression in monolayer and non-neuroectoderm cells differentiated from H1

Project description:Per- and polyfluoroalkyl substances (PFAS) are a group of organic chemicals that contain multiple carbon-fluorine bonds, which make them highly stable and widespread in our environment, even in the human body. They have been detected in cord blood and showed correlation with neonatal health abnormalities such as weight loss. Here, we utilized human embryonic stem cell-based model to assess the early developmental toxicity of six PFAS (PFOA, PFHxA, PFBA, PFOS, PFHxS, and PFBS) and their skin developmental toxicity. We employed global monolayer differentiated model to mimic the early embryonic development and performed non-neural ectoderm (NNE) differentiation which can give rise to keratinocytes to imitate early skin development. Transcriptomic analysis revealed that PFOS caused the most differentially expressed genes (DEGs) both in day 16 global monolayer samples and NNE samples to making it the most toxic of the six PFAS. Further validation of gene expression levels showed that the TGF-β signaling pathways were involved in the early global developmental toxicity and in the early skin developmental toxicity of PFOS. Interestingly, our results showed that PFOS inhibited the ciliogenesis of NNE cells, which may interfere with the transduction of TGF-β, Notch, Hedgehog, and Hippo signaling pathways, ultimately inhibiting early embryonic skin development and posing a potential threat to the occurrence of congenital skin diseases. Overall, our study suggests a potential toxicity mechanism by which PFAS inhibit ciliogenesis to interfere with the transduction of important signaling pathways, leading to early global developmental toxicity as well as early embryonic skin developmental toxicity and potential congenital skin diseases.

Gene expression profiles in human duodenal organoid-derived monolayer

Project description:The human small intestine is the key organ for absorption, metabolism, and excretion of orally administered drugs. To preclinically predict these reactions in drug discovery research, a cell model that can precisely recapitulate the in vivo human intestinal monolayer is desired. Here, we developed a monolayer platform using human biopsy-derived duodenal organoids for application to pharmacokinetic studies. The human duodenal organoid-derived monolayer was prepared by a simple method in 3–8 days. It consisted of polarized absorptive cells and had a barrier function. It showed much higher cytochrome P450 (CYP) 3A4 and carboxylesterase (CES) 2 activities than Caco-2 cells. It also showed efflux activity of P-glycoprotein (P-gp) and inducibility of CYP3A4. Finally, its gene expression profile was closer to the adult human duodenum, compared to the profile of Caco-2 cells. Based on these findings, this monolayer assay system using biopsy-derived human intestinal organoids is likely to be widely adopted.
