Ionizing Radiation-induced Acute Damage in the Submandibular Gland

ABSTRACT: To investigate the unique mechanism of radiation submandibular gland injury, we gave 15Gy local one-time irradiation to the submandibular gland area of eight-week-old C57BL/6J female mice. We then collected submandibular glands 24 hours after radiation for transcriptome sequencing.

Proteomic characterization of native and decellularized rat submandibular gland scaffolds prepared using the S3T1 protocol

Project description:Salivary gland hypofunction lacks effective regenerative treatment, and organ-specific decellularized extracellular matrix scaffolds represent a promising biomaterial platform for tissue engineering. In this study, rat submandibular glands were decellularized using a rapid S3T1 workflow consisting of freeze-thaw pretreatment, 1% SDS treatment for 3 h, and 0.25% trypsin-EDTA treatment for 1 h. Native submandibular glands and decellularized scaffolds were compared by data-independent acquisition LC-MS/MS to characterize protein composition and extracellular matrix preservation. The dataset was generated to identify retained matrisome components and to evaluate the proteomic features of the decellularized scaffold in comparison with native tissue. These data support the use of the decellularized submandibular gland scaffold as a bioactive platform for salivary gland tissue engineering.

RNA-Seq reveals heterogeneity in the submandibular glands and submandibular gland draining lymph nodes of SS mice

Project description:In this study, we analyzed the submandibular glands of P40-/-CD25+/- (control mice) and P40-/-CD25-/- (SS mice) after PD-1-targeted CAR-T cells or control T cells treatment using RNA-Seq. We found that the transcription of inflammatory genes was down-regulated and the transcription of tissue repair genes was up-regulated in the submandibular glands of SS mice after PD-1-targeted CAR-T cells treatment. In addition, PD-1+Tigit+CD8+ T cells in the submandibular gland draining lymph nodes of SS mice showed transcription characteristics of tissue resident memory precursors.

Genome-wide expression analysis of submandibular salivary gland tumours of Smgb-Tag mice

Project description:To reveal novel molecular factors behind the development of salivary gland cancer, we performed gene expression analyses from Smgb-Tag mouse salivary gland samples. The overall purpose was to apply these results for clinical use to find new approaches for both possible therapeutic targets and more accurate diagnostic tools in identification of salivary gland cancers. Smgb-Tag mouse strain, in which salivary neoplasms arise through a dysplastic phase in submandibular glands, was investigated using genome-wide microarray expression analysis, Ingenuity pathway analysis, RT-PCR, and immunohistochemistry. 3 normal, 3 dysplastic, and 3 adenocarcinomatous submandibular gland tumours of Smgb-Tag mice.

Project description:The salivary glands often become damaged in individuals receiving radiotherapy for head and neck cancer, resulting in chronic dry mouth. This leads to detrimental effects on their health and quality of life, for which there is no regenerative therapy. Progenitor cells can replenish damaged tissue. We have shown the macrophages interact with KRT14+ basal epithelial progenitor cells; however, their function is unknown. Here, using bulk RNAseq we profile the transcriptomes of KRT14+ cells isolated from adult mouse submandibular salivary gland 14 days after targeted irradiation which is either macrophage-proficient or macrophage-deficient. For each sample equal numbers of cells from 8 mice were pooled.
