Effects of arbuscular mycorrhizal symibiosis on gene expression in the foxtail millet roots

ABSTRACT: To investigate the involvement of arbuscular mycorrhizal symbiosis in the moleular regulation in foxtail millet roots and the effects of genetic variation on AMS-mediated molecular regulation, we isolated total RNA from the roots of 3 different landraces for comprehensive transcriptomic analysis. We then performed gene expression profiling analysis using data obtained from RNA-seq of 3 different landraces (Hanevalval, TT8, ICE36) after 6-week mock or arbuscular mycorrhizal fungi treatments.

Genotype-Specific Modulation of Drought Tolerance by Arbuscular Mycorrhizal Symbiosis in Foxtail Millet

Project description:Drought stress is a major environmental factor limiting crop productivity. Arbuscular mycorrhizal fungi (AMF), as beneficial soil microbes, can improve plant growth and stress resilience; however, the effectiveness of this symbiosis is often influenced by the host plant's genetic background. In this study, we investigated the interaction between AM symbiosis and drought tolerance in two foxtail millet (Setaria italica) genotypes with contrasting drought responses: the drought-tolerant ISE42 and the drought-sensitive TT8. Following a 14-day drought treatment, both genotypes exhibited wilting, but only AMF-colonized ISE42 plants recovered upon rewatering. Transcriptomic analysis revealed that AM symbiosis significantly enhanced the expression of genes involved in nitrogen transport, assimilation, lignin metabolism, and cellulose biosynthesis in ISE42, but not in TT8. These molecular changes suggest improved nutrient uptake and cell wall reinforcement as key mechanisms underlying enhanced drought tolerance. Additionally, stress hormone signaling pathways were downregulated in colonized ISE42 roots, indicating possible alleviation of drought-induced stress through AM symbiosis. Our results demonstrate genotype-specific effects of AMF on drought tolerance and highlight the importance of considering host genetic variation in the application of AMF for crop improvement.

SiYSL1 may Paticipate in Iron Absorption, Transportation, and Grain Accumulation in Foxtail Millet

Project description:Iron (Fe) deficiency is a globally widespread condition in which the body lacks sufficient Fe to produce hemoglobin. However, major food crops generally have low grain Fe contents. Consequently, enhancing grain Fe concentrations is important for improving the health of populations that rely on grains as staple foods. Here, we isolated a yellow stripe leaf mutant of foxtail millet (Setaria italica), designated yellow stripe-like 1 (ysl1). This mutant exhibited typical Fe deficiency symptoms that were alleviated when grown under Fe-sufficient conditions. Compared with the wild-type, Siysl1 showed lower Fe concentrations in seedling roots, shoots, stems, elongation-stage leaves, panicles, and seeds, but a higher Fe concentration in heading-stage leaves. Using MutMap+, we identified and cloned SiYSL1 and validated its function through CRISPR/Cas9-mediated knockout experiments. SiYSL1 encodes an Fe-phytosiderophore transporter and is highly induced under Fe deficiency conditions. Histochemical staining revealed that SiYSL1 is specifically expressed in vascular bundles of roots and leaves of plants grown under Fe deficiency conditions, and in spikelets, expanding ovaries, basal endosperm, and embryo-surrounding tissues. Thus, SiYSL1 appears to regulate Fe uptake and homeostasis, and plays an essential role in Fe translocation to seeds. The overexpression of SiYSL1 in rice and foxtail millet significantly increased seed Fe contents, suggesting its value in crop breeding. Predicted transcription factor binding sites in the SiYSL1 promoter and a spikelet transcriptome analysis indicated that transcription factors regulate SiYSL1 expression. Our study provides new genetic resources for the Fe bio-enhancement of food crops and insights into the mechanisms responsible for seed Fe accumulation.
