Single-nucleus sequencing data from developing and adult mouse olivocochlear neurons

ABSTRACT: Transcriptional diversity of mouse olivocochlear neurons (OCNs) was examined using single-nucleus sequencing of brainstem cholinergic neurons at P1, P5, and P26-P28. This dataset includes multiple brainstem cell types, including OCNs and several other populations of cranial motor neurons.

Single nucleus RNA-sequencing defines unexpected diversity of cholinergic neuron types in the adult mouse spinal cord

Project description:We developed a targeted single nucleus RNA sequencing approach to enrich and transcriptomically characterize cholinergic neurons of the adult mouse spinal cord. Our data expose markers for known classes of cholinergic neurons and their extremely rich diversity. Visceral/preganglionic motor neurons could be divided into more than a dozen transcriptomic classes with anatomically restricted localization along the spinal cord. The complexity of the skeletal motor neurons was also reflected in our analysis with alpha, gamma, and a third subtype clearly distinguished. We further identified previously unrecognized subtypes of cholinergic interneurons.

Single nucleus transcriptomics: Apical resection in newborn pigs extends the time-window of cardiomyocyte proliferation and myocardial regeneration

Project description:Background: In this study, we have extended the myocardial regenerative window to postnatal day 28 (P28) by a double injury pig model (ARP1MIP28) of apical resection at P1 (ARP1) and LAD ligation at P28 (MIP28). However, the molecular mechanism of regenerative window extension is not known. Results: Gene-expression profiles revealed that regenerating ARP1MIP28 hearts contained different subpopulations of cardiomyocytes early after 2nd injury. The six cardiomyocyte clusters were identified (CM1-CM6) by Louvain clustering. The CM6, CM3, and CM2 clusters have mainly consisted of fetal (92.4%), CTL-P1 (95.7%), and CTL-P56 (96.2%) cardiomyocytes. In contrast, the CM1 cluster has consisted of heterogeneous cardiomyocyte groups from all other animal groups, including less than 1% of fetal and CTL-P1 cardiomyocytes. The CM5 and CM4 clusters have mainly consisted of ARP1MIP28-P35 cardiomyocytes, 97.8%, and 68.6%, respectively. Sparse modeling analysis revealed that AR and MI injury, both alone and in combination, appeared to promote the proliferative capacity of cardiomyocytes and perturb cardiomyocyte maturation. Brainstem organoids from human pluripotent stem cells contain neural crest population

Project description:Brainstem controls heartbeat, blood pressure and respiration which are vital for life, therefore, damage to brainstem can be lethal. Brain organoids derived from human pluripotent stem cells recapitulate the course of human brain development and are expected to be useful for medical research on central nervous system disorders. However, existing organoid models have limitations, hampering the elucidation of diseases affecting specific components of the brain. Here, we developed a method to generate human brainstem organoids (hBSOs), containing neural crest stem cells as well as midbrain/hindbrain progenitors, noradrenergic and cholinergic neurons, and dopaminergic neurons, demonstrated by specific electrophysiological signatures. Single-cell RNA-sequence analysis, together with proteomics and electrophysiology further revealed that the cellular population in the organoids is similar to that of the human brainstem and neural crest, which makes it possible to make use of the hBSOs for modeling and grafting for transplantation.

Phenotypic and genotypic stability in a preclinical model of renal cell carcinoma

Project description:Renal cell carcinomas are often resistant to conventional cytotoxic agents. Xenograft models are used for in vivo preclinical studies and drug development. The validity of these studies is highly dependent on the phenotypic and genotypic stability of the models. We evaluated the stability of six kidney tumors xenografted in nude/NMRI mice. We compared the initial sample (P0), 1st (P1) and 5th (P5) passages for the following criteria: histopathological analysis, immunohistochemistry for CK7, CD10, vimentin and p53, and DNA analysis with 10 microsatellites. We also performed CGH-array and we compared the sensitivity to sunitinib with tumor response in patients. The six xenografted tumors showed no difference between the morphology of the primary tumor and that of the xenografted tumor at P1 and P5 but we observed differences for CK7 expression in one case and for p53 expression in another. Out of 44 P0-P1-P5 groups with fully available microsatellite data, 29 (66%) showed no significant difference from P0 to P5 while 15 (34%) showed new or lost alleles. In CGH-array, genomic alterations at P5 were close to those already present in the initial tumor. We also observed similarity between the models and the patients for response to antiangiogenic treatment. We showed that the six xenograft models were phenotypically and genotypically stable across passages and showed similarity with response to treatment in patients.
