Single-nucleus sequencing data from developing and adult mouse olivocochlear neurons

ABSTRACT: Transcriptional diversity of mouse olivocochlear neurons (OCNs) was examined using single-nucleus sequencing of brainstem cholinergic neurons at P1, P5, and P26-P28. This dataset includes multiple brainstem cell types, including OCNs and several other populations of cranial motor neurons.

Single cell RNA sequencing of lateral and medial olivocochlear efferent neurons using PatchSeq

Project description:Olivocochlear (OC) efferent neurons are the final component of the descending auditory system. The lateral olivocochlear (LOC) neurons innervate type I spiral ganglion neurons (SGN). Their role in hearing is unknown, but they may have roles in protection from noise trauma or changing the threshold of SGN activation. The medial olivocochlear (MOC) efferent neurons innervate outer hair cells (OHC), inhibiting OHC function and thus dampening cochlear vibrations. LOC and MOC function is poorly understood at the single cell level, but recent use of transgenic mice (ChAT-IRES-Cre; tdTomato) expressing fluorescent reporters in cholinergic neurons, including LOC and MOC neurons, has allowed patch-clamp recordings from the neurons in mouse brainstem slices to begin to characterize their intrinsic electrical properties and synaptic activity. Recent single nucleus sequencing experiments from LOC and MOC neurons has given insight into the genetic programs that generate their function, including changes to expression profiles following noise trauma. Here we utilize the PatchSeq technique to combine patch-clamp electrophysiological recordings with deep single-cell transcriptome analysis from identified LOC and MOC neurons in transgenic ChAT-IRES-Cre; tdTomato mice. In this preliminary dataset, we generated patch-clamp electrophysiology data and single cell transcriptome data from 18 identified LOC and 13 identified MOC neurons.

Lateral olivocochlear neurons modulate cochlear responses to noise exposure

Project description:The sense of hearing originates in the cochlea, which detects sounds across dynamic sensory environments. Like other peripheral organs, the cochlea is subjected to environmental insults, including loud, damage-inducing sounds. In response to internal and external stimuli, the central nervous system directly modulates cochlear function through olivocochlear neurons (OCNs), which are located in the brainstem and innervate the cochlear sensory epithelium. One population of OCNs, the lateral olivocochlear (LOC) neurons, target spiral ganglion neurons (SGNs), the primary sensory neurons of the ear. LOCs alter their transmitter expression for days to weeks in response to noise exposure (NE), suggesting that they are well-positioned to tune SGN excitability over long time periods in response to auditory experience. To examine how LOCs affect auditory function after NE, we characterized the transcriptional profiles of OCNs and found that LOCs exhibit transient changes in gene expression after NE, including upregulation of multiple neuropeptide-encoding genes.

Brainstem organoids from human pluripotent stem cells contain neural crest population

Project description:Brainstem controls heartbeat, blood pressure and respiration which are vital for life, therefore, damage to brainstem can be lethal. Brain organoids derived from human pluripotent stem cells recapitulate the course of human brain development and are expected to be useful for medical research on central nervous system disorders. However, existing organoid models have limitations, hampering the elucidation of diseases affecting specific components of the brain. Here, we developed a method to generate human brainstem organoids (hBSOs), containing neural crest stem cells as well as midbrain/hindbrain progenitors, noradrenergic and cholinergic neurons, and dopaminergic neurons, demonstrated by specific electrophysiological signatures. Single-cell RNA-sequence analysis, together with proteomics and electrophysiology further revealed that the cellular population in the organoids is similar to that of the human brainstem and neural crest, which makes it possible to make use of the hBSOs for modeling and grafting for transplantation.

Molecular and functional profiling of cell diversity and identity in the lateral superior olive, an auditory brainstem center with ascending and descending projections

Project description:The lateral superior olive (LSO), a conspicuous integration center in the auditory brainstem, contains a remarkably heterogeneous neuron population. Ascending neurons, predominantly principal neurons (pLSOs), process interaural level differences for sound localization. Descending neurons (lateral olivocochlear neurons, LOCs) provide feedback into the cochlea and likely protect from acoustic overexposure. The molecular determinants of the neuronal diversity in the LSO are largely unknown. Here, we employed patch-seq analysis in juvenile mice to classify LSO neurons by their functional and molecular profiles, including developmental aspects. Across the sample (n=86), genes involved in ATPsynthesis were particularly highly expressed, confirming the energy expenditure of auditory neurons. Two clusters were identified, pLSOs and LOCs. They were distinguishable by 353 differentially expressed genes (DEGs), most being novel for the LSO. Electrophysiological analysis corroborated the transcriptomic clustering. We focused on genes impacting neuronal input-output properties and validated some by immunohistochemistry, electrophysiology, and pharmacology. These genes encode proteins like osteopontin, Kv11.3, and Kvb3 (pLSO-specific), calcitonin-gene-related peptide (LOC-specific), or Kv7.2 and Kv7.3 (no DEGs). We identified 12 ‘Super DEGs’ and 12 genes demonstrating ‘Cluster similarity’. Collectively, we provide fundamental and comprehensive insights into the molecular composition of individual ascending and descending neurons in the juvenile auditory brainstem and how this may relate to their specific functions, including developmental aspects.
