Dual-transcriptome of post-germinating Arabidopsis thaliana mutant lines infected by Alternaria brassicicola

ABSTRACT: We analyzed transcriptome changes from germinating cyp79B2 cyp79B3, qko, and pad3 mutant Arabidopsis seeds. Here were used mutant lines in Col-0 genetic background: a cyp79B2 cyp79B3 double mutant defective in indolic glucosinolate (cyp79B2/B3), a cyp79B2 cyp79B3 myb28 myb29 quadruple mutant defective in indolic and aliphatic glucosinolates (qko), and a camalexin deficient mutant (pad3). Differentially expressed genes were identified at 3, 6 and 10 days after sowing from both the mutant line seeds and from Alternaria brassicicola when inoculated seeds were analyzed.

Alternaria brassicicola infection regulates miRNAs in Arabidopsis thaliana [sRNA]

Project description:Plant microRNAs (miRNAs) have been implicated in plant immunity. These mainly focusing Arabidopsis thaliana threatened by (hemi-)biotrophic pathogens such as the bacterial pathogen Pseudomonas syringae. Here, we show that the Arabidopsis miRNA pathway is important for defense responses against the necrotrophic fungus Alternaria brassicicola. The miRNA pathway mutant ago1 exhibits an exaggerated response when treated with A. brassicicola, proposing that AGO1 is positive regulator. We found a subset of Arabidopsis miRNAs that quickly change their expression and their abundance in AGO1 complexes in plants exposed to A. brassicicola. The miRNAs responding to pathogen treatment are mainly targeting genes encoding metabolic enzymes, proteins involved protein degradation or transposons. In case of miR163, A. brassicicola infection results in increased levels of miRNA precursors and preferential accumulation of an unspliced form of pri-miR163, suggesting that A. brassicicola infection changes the transcriptional and post-regulation of pri-miRNAs. miR163 acts as a negative regulator of plant defense because mir163 mutants are more resistant when treated with A. brassicicola. Taken together, our results reveal the existence of positively and negatively acting Arabidopsis miRNA modulating the defense responses against A. brassicicola and highlight the importance of host miRNAs in the interaction between plants and necrotrophic pathogens.

Indolic metabolites are required for antifungal defense of the Arabidopsis mlo2 mutant

Project description:Arabidopsis thaliana genes MLO2 (Mildew resistance locus-O 2), MLO6 and MLO12 exhibit unequal genetic redundancy with respect to the modulation of defense responses against powdery mildew fungi and the control of developmental phenotypes such as premature leaf decay. We show that early chlorosis and necrosis of rosette leaves in mlo2 mlo6 mlo12 mutants reflects an authentic but untimely leaf senescence program. Comparative transcriptional profiling revealed that transcripts of several genes encoding tryptophan/indole biosynthetic enzymes hyper-accumulate during vegetative development in the mlo2 mlo6 mlo12 mutant. Elevated expression levels of these genes correlate with altered steady-state levels of several indolic metabolites, including the phytoalexin camalexin and indolic glucosinolates, during development in the mlo2 single and the mlo2 mlo6 mlo12 triple mutant. Results of genetic epistasis analysis suggest a decisive role for indolic metabolites in mlo2-conditioned antifungal defense against both biotrophic powdery mildews and a camalexin-sensitive strain of the necrotrophic fungus, Botrytis cinerea. The wound- and pathogen-responsive callose synthase Powdery mildew resistance 4/Glucan-synthase-like 5 (PMR4/GSL5) was found to be responsible for the spontaneous callose deposits in mlo2 mutant plants but dispensable for mlo2-conditioned penetration resistance. Our data strengthen the notion that powdery mildew resistance of mlo2 genotypes is based on the same defense execution machinery as innate antifungal immune responses that restrict invasion of non-adapted fungal pathogens. Mature rosette leaves from WT and mlo2 mlo6 mlo12 triple mutant plants were harvested at 5, 6 and 7 weeks after sowing. 6 arrays.

Gene expression profiles in Arabidopsis plants during the treatment of AgNO3.

Project description:Camalexin, an indolic secondary metabolite, is the magor phytoalexin produced by Arabidopsis thaliana. Camalexin biosynthesis is induced by abiotic stresses such as heavy metal treatment (e.g AgNO3). With the aim of identifying positive regulators of camalexin biosynthesis in Arabidopsis, we listed putative transcription factor genes strongly induced during the time course of AgNO3 treatment from a transcriptome analysis. AgNO3 induced gene expression in Arabidopsis was measured at 48 and 72 hours after the 0.1 mM AgNO3 treatment. Arabidopsis seedlings were germinated under sterile conditions on GM-agar plates. Seedlings (one-week after germination) were then transferred to GM liquid culture and shaken for two weeks. Plants were then treated with 0.1 mM AgNO3 and sampled at 0 h, 48 h and 72 h after the treatment. Three independent experiments were performed at each time (0, 48, or 72 h) using different plants for each experiment.
