Project description:Diversity of cortical radial glia cells (RGCs) and their complex relationships to generate neurons in species with expanded germinal zones and a folded cortex, remains unclear. We used single-cell RNA sequencing (scRNA-seq) of microdissected cortical germinal layers (ventricular zone (VZ) and outer subventricular zone (OSVZ)), from two cortical regions (splenial gyrus (SG) and neighboring lateral sulcus (LS)) at two critical time points for ferret cortex development (embryonic day (E) 34 and postnatal day (P) 1) to distinguish the molecular diversity of progenitors and newborn neurons, and study their transcriptomic trajectories.

Human-specific gene ARHGAP11B promotes basal progenitor amplification and neocortex expansion

Project description:The evolutionary expansion of the human neocortex reflects increased amplification of basal progenitors in the subventricular zone, producing more neurons during fetal corticogenesis. Here, we analyze the transcriptomes of distinct progenitor subpopulations isolated by a novel approach from developing mouse and human neocortex. We identify 56 genes preferentially expressed in human apical and basal radial glia that lack mouse orthologs. Among these, ARHGAP11B has the highest degree of radial glia-specific expression. ARHGAP11B arose from partial duplication of the Rho GTPase-activating-protein–encoding ARHGAP11A on the human lineage after separation from the chimpanzee lineage. Expression of ARHGAP11B in embryonic mouse neocortex promotes basal progenitor generation and self-renewal, and can increase cortical plate area and induce gyrification. Hence, ARHGAP11B may have contributed to evolutionary expansion of human neocortex. Gene expression profiles of mouse and human purified neocortical progenitor types and neurons were generated by RNA-seq and analyzed including inter- and intra-species comparison.

PI3K-Yap activity drives cortical gyrification and hydrocephalus in mice

Project description:Mammalian brain evolved through several transitions between gyrencephaly and lissencephaly. Mechanisms generating gyrified or smooth brain are incompletely understood. Here we demonstrate that a short embryonic pulse of activating mutations in Pik3ca, the catalytic subunit of PI3K enzyme is sufficient to cause mouse cortical gyrification. We demonstrate that this gyrification phenotype is initiated by subtle focal regulation of apical cell adhesion and proliferation via PI3K-dependent localization of Yap protein. Treatment with verteporfin (Drug) a nuclear Yap inhibitor, attenuated over-proliferation and adhesion abnormalities, and subsequently gyrification. The purpose of this study was to compare the control and mutant mouse transcriptome, and their respective effect upon verteprofin treatment.

LGALS3BP modulates local gyrification in the human brain

Project description:Basal radial glial cells (bRGs) are neural progenitors enriched in primates and humans and were proposed to contribute to the expansion of neurons during cortical development in gyrencephalic species. Shortly after their generation, bRGs delaminate towards the outer subventricular zone, where they divide multiple times before differentiation. Thus, the regulation of bRGs generation could be essential for the establishment of correct gyrification within the human cortex. Here, we study the role of LGALS3BP, a secreted protein whose RNA expression is enriched in bRGs. By using cerebral organoids, human fetal tissues and mice, we show that manipulation of LGALS3BP regulated bRG generation. Additionally, individuals with unique de novo variants in LGALS3BP demonstrate abnormal gyrification and thickness at multiple sites over their cortex. Single-cell-RNA-sequencing and proteomics reveal the extracellular matrix involvement in the LGALS3BP mediated mechanisms. We find that LGALS3BP is required for bRGs delamination and influences corticogenesis and gyrification in humans.

Genetic and microstructural differences in the cortical plate of gyri and sulci during gyrification in fetal sheep

Project description:Gyrification of the cerebral cortex is a developmentally important process, however the mechanisms that drive cortical folding remain unknown. Theories propose that changes within the cortical plate (CP) cause gyrification, yet differences between the CP below gyri and sulci have not been investigated. Here we report genetic and microstructural differences in the CP below gyri and sulci assessed before (gestational age (GA) 70), during (GA 90) and after (GA 110) gyrification in fetal sheep. The areal density of BDNF-, CDK5-, and NeuroD6-immunopositive cells were increased, and HDAC5 and MeCP2 mRNA levels were decreased, in the CP below gyri compared to sulci during gyrification, but not before. Only the areal density of BDNF-immunopositive cells remained increased after gyrification. MAP2-immunoreactivity was also increased in the CP below gyri compared to sulci at GA 90, and this was associated with microstructural changes assessed via neurite orientation dispersion and density imaging (NODDI) at GA 98. Differential neurite outgrowth may therefore be a strong candidate to explain local changes in CP architecture during gyrification.

Development of molecular genetic analysis tools for the domestic ferret (Mustela putorius furo)

Project description:The domestic ferret (Mustela putorius furo) has been used as animal model for decades, largely because its susceptibility to infection with a large number of pathogens such as influenza virus, SARS Corona virus and Canine distemper virus. Despite its importance for biomedical research, little is known about the genome of the M. Furo. The number of reagents for molecular and immunological analysis is thus restricted. To circumvent this, we present here a parallel sequencing effort to produce an extensive EST dataset derived from a normalized ferret cDNA library made from mRNA from ferret blood, liver, lung, spleen and brain. We produced more than 500000 sequence reads that were assembled into over 15000 partial ferret transcripts. These ESTs were combined with the available ferret sequences in the GenBank to develop a ferret specific microarray platform. Using this array, we detected tissue specific expression patterns which were confirmed by quantitative real time PCR assays and comparison to orthologous transcription profiles of mouse and human. We also present a set of 41 ferret transcript with even transcription profile across the tested tissues, indicating their usefulness as housekeeping genes. This study paves way for development of additional reagents for analysis of the ferret model. Three biological replicates of blood, lung, spleen, liver and brain was hybridized to the ferret specific microarray.
