Gene expression study in skin fibroblasts

ABSTRACT: To investigate the in vivo developmental potential, we generated dedifferentiated-skin fibroblast from adult skin fibroblasts with a FVB background, a different genetic strain from the protein-donor embryonic stem (ES) cell. Primary cardiac fibroblast (cFB) was cultered from an adult C57/BL mouse to compare with ES cells(E14-mES, C57-mES). We also introduced C57 background ES cell-derived extract proteins into cFB by reversible permeabilisation. The ES-like cells (colonies) were expanded with subculture every 3 to 5 days using the standard ES cell culture protocol.

Induction of pluripotent stem cells from adult somatic cells by protein-based dedifferentiation

Project description:The concept of dedifferentiation of somatic cells into pluripotent stem cells has opened a new era in regenerative medicine. Viral transduction of defined factors has successfully achieved pluripotency derived from somatic cells. However, during the generation process of induced pluripotent stem (iPS) cells, genetic integration of certain factors may cause mutagenesis or tumorigenicity, which limits further application. Therefore, there is currently ongoing an extensive search for new methods such as transient gene delivery and oocyte-free and non-viral inducers like small molecules. Here we show that the transient delivery of embryonic stem (ES) cell-derived soluble proteins enables dedifferentiation of mouse adult somatic cells converting them into pluripotent stem cells without the introduction of certain transcription factors or genetic manipulation. During the dedifferentiation, global gene expression patterns and epigenetic status were converted from the somatic to the ES-equivalent status. Dedifferentiated somatic cells were morphologically, biologically and functionally indistinguishable from ES cells. Furthermore, the dedifferentiated cells possessed in vivo differentiation and development potential. Our results provide an alternative and safe strategy for dedifferentiation of somatic cells that can be used to facilitate pluripotent stem cell-based cell therapy. Total RNA from mES cell (triplicate), adult fibroblast (triplicate), or dedifferentiated adult fibroblast was isolated. Samples were hybridized to a Affymetrix Mouse Gene 1.0 ST Array according to the manufacturer's protocol. After hybridization, the chips were stained and washed in a Genechip Fluidics Station 450(Affymetrix) and scanned by using a Genechip Array scanner 3000 7G (Affymetrix).

The proteomic study of serially passaged human skin fibroblast cells uncovers down-regulation of SMC4 involved in replicative senescence

Project description:In this study, a label-free quantitative proteomic approach was employed to analyze the serial passaged human skin fibroblast （CCD-1079Sk） cells, within 3305 proteins quantified. Of which, 372 proteins were significantly changed in early passage(P6), middle passage (P12) and later passage (P21), with a time-dependent decrease or increase tendency. Then, of which, SMC4 was selected for further biological validation. The results confirmed that the expression of SMC4 was significantly down-regulated in a time-dependent manner in the subculture of human skin fibroblasts (HSFb) with Western Blot experiment.

Genome wide analysis of gene expression of rat ES cells, rat embryonic fibroblast cells and mouse ES cells

Project description:Rat ES cells were derived using 3I medium from E4.5 blastocysts. Rat embryonic fibroblast cells were derived form E14.5 embryos. To analyze the mechanism under the selfrenewal of rat ES cells, microarrays were used for the genome wide analysis of gene expressoin profiles in rat ES cells. Rat embryonic fibroblast cells and mouse ES cells were tested at same time as control. Our results from clustering analysis demonstrated that the gene expression profile of rat ES cells resembles mouse ES cells, but not REFs. Hnrnpll promotes multiple ES cell preferred-exon skipping during ES cells differentiation, to reduce the ES cells specific isoform and facilitate ES cells exit from pluripotency towards differentiation.

Deficiency of CFB attenuates renal tubulointerstitial damage via inhibiting ceramide synthesis in diabetic kidney disease

Project description:Accumulating evidence suggests the pathogenic role of immunity and metabolism in diabetic kidney disease (DKD). Herein, we aimed to investigate the effect of complement factor B (CFB) on lipid metabolism in the development of DKD. We found that in patients with diabetic nephropathy (DN), the staining of Bb, CFB, C3a, C5a, and C5b-9 were significantly elevated in renal tubulointerstitium. Cfb knockout diabetic mice had significantly milder tubulointerstitial injury and less ceramide biosynthesis. The in vitro study demonstrated that cytokine secretion, endoplasmic reticulum stress, oxidative stress, and cell apoptosis were ameliorated in siCFB HK-2 cells under high glucose conditions. Exogenous ceramide supplementation attenuated the protective effect of CFB knockdown in HK-2 cells, while inhibiting ceramide synthases (CERS) with fumonisin B1 in CFB-overexpressing cells rescued the cell injury. CFB knockdown could downregulate the expression of NF-κB p65, which initiates the transcription of CERS3. Furthermore, C3 knockdown abolished CFB-mediated cytokine secretion, NF-κB signaling activation, and subsequently ceramide biosynthesis. Thus, CFB deficiency inhibited activation of the complement alternative pathway and attenuated kidney damage in DKD, especially tubulointerstitial injury, via inhibiting the NF-κB signaling pathway, further blocking the transcription of CERS, which regulates the biosynthesis of ceramide. CFB may be a promising therapeutic target of DKD.

Gene expression profiles of Tet1 knockout (T1KO), Tet2 knockout (T2KO) and Tet1/Tet2 double knockout (DKO) mouse embryonic stem cells.

Project description:Global gene expression profile of single and double mutant mouse ES cells were compared to wt ES cells. Two male Tet1 KO, one male Tet2 KO, two male double KO, two female double KO, two male WT and two female WT mouse ES cells were compared. Global gene expression profile of single and double knockout mouse embryonic stem cells were compared to that of wild type mouse ES cells. All used ES lines were derived from C57/BL/6 mixed background mice. RNA from feeder free mutant mouse ES cells was competetively hybridized against RNA from WT ES cells. Same sex lines were compared. Two independent ES line of each genotype were used, with the exception of Tet2 KO ES cells where only one male line was used.

CGH analysis of haploid ES cells

Project description:We performed microRNA microarray analysis to determine microRNAs that are differentially expressed between the differentiated hfRPE monolayer (pigmented, hexagonally packed) and dedifferentiated hfRPE cells (non-pigmented, fibroblast-like). Experiment was performed using the Agilent-019118 Human miRNA Microarray 2.0 G4470B. 2 -condition experiment: differentiated and dedifferentiated hfRPE cells. 3 biological replicates for each condition. Also includes a sample that has characteristics of both cell types (representing a transitional state).
