Synergistic targeting of TWIST1 and HOXA9 in MLL-AF9+ AML via combination of SD70 and MI-503

ABSTRACT: To identify the downstream targets of TWIST1 or Hoxa9, we carried out RNA sequencing to characterize transcriptional changes upon Twist1 deletion or Hoxa9 knockdown in LSCs from MLL-AF9+ AML mice.

Combination of SD70 and MI-503 exerts a synergistic effect against MLL:: AF9-driven AML

Project description:Acute myeloid leukemia (AML), a heterogeneous hematologic malignancy, is the most common form of acute leukemia in adults. MLL rearrangements (MLL-r) are observed in approximately 10% of AML and are associated with a relatively poor prognosis, highlighting the need for new treatment regimens. MLL fusions recruit KDM4C to mediate epigenetic reprogramming, which is required for maintenance of MLL-r leukemia. In this study, we used a combinatorial drug screen to selectively identify synergistic treatment partners for the KDM4C inhibitor SD70. The results showed that the drug combination of SD70 and MI-503, a potent Menin-MLL inhibitor, induced synergistically enhanced apoptosis in MLL-r leukemia cells without affecting normal CD34+ cells. In vivo treatment with SD70 and MI-503 significantly prolongs survival in AML xenograft models. Differential Gene expression analysis by RNA-seq following combined pharmacological inhibition of SD70 and MI-503 revealed changes in numerous genes, with MYC target genes being the most significantly downregulated. Taken together, these data provide preclinical evidence that the combination of SD70 and MI-503 is a potential dual-target therapy for MLL-r AML.

The transcription factor HOXA9 induces expression of the chromatin modifier SMYD3 to drive leukemogenesis

Project description:Fusion oncogene MLL-AF9 initiates AML via downstream targets such as HOXA9. Drivers in the complicated settings of advanced AML, however, remain to be incompletely elucidated. Any factors to incur upregulation of the effector HOXA9 predictably aggravate the effect of DOT1L-mediated H3K79 methylation on HOXA9 expression in MLL-AF9-driven AML. In the present study, we identified that SET and MYND domain-containing protein 3 (SMYD3) was overexpressed in AML and predicted a poor prognosis for AML patients. Given that H3K4me3 typically activates the transcription of oncogenes, we hypothesized that SMYD3-catalyzed H3K4me3 may directly increase HOXA9 transcription, offering an additional regulation layer to HOXA9 gene transcription activation in MLL-AF9 AML. We tested this hypothesis and unveiled that SMYD3 is responsible for mediating H3K4me3 enrichment and for independently activating HOXA9 transcription. Transcription factor HOXA9 in turn bound to the promoter region of SMYD3 and enhanced its transcription. The resultant vicious circle of SMYD3-H3K4me3-HOXA9 exacerbated proliferation and blocked differentiation in both AML cell lines and primary cells fractionated from patients with AML. Combinational disruption of this loop and DOT1L inhibition led to enhanced anti-leukemia activity against MLL-AF9 AML in vitro and in vivo. In conclusion, our findings may advocate the current understanding regarding the underlying mechanism and offer SMYD3 as a promising intervention target to override the complicated settings in advanced AML.

Project description:How the stemness of adult stem cells and cancer stem cells is regulated by environmental cues through surface receptors is poorly understood. In this gene expression analysis, we found that, in the mouse MLL-AF9 acute myeloid leukemia (AML) model, a deficiency in intracellular signaling of inhibitory receptor PIR-B resulted in increased differentiation and decreased stemness of leukemia stem cells, revealing that PIR-B supports leukemia development. Our study indicates unexpected functional significance of a classical immune inhibitory receptor in the maintenance of stemness of cancer stem cells.
