RNA sequencing on in vitro differentiated WT and GOT1-/- Th17 cells

ABSTRACT: Naive CD4+ T cells (CD4+CD25-CD44-CD62L+) were FACS sorted from WT or T cell conditional GOT1-/- C57BL/6 mouse spleen and lymph nodes. Sorted cells were differentiated into Th17 cells in the presence of TGFβ, IL1β, IL6, IL23, anti-IFNγ and anti-IL4. RNA sequencing was then performed on day4 differentiated TH17 cells.

Transcription profiling of human CD4 positive T cell subsets

Project description:Peripheral Blood Mononuclear Cells (PBMCs) were isolated from a buffy coat (Australian Blood Bank) using Ficoll methodology. CD4+ T cells were isolated using Dynal Beads kit. Pure CD4+ T cells were then stained using a cocktail of monoclonal antobodies (mAbs), including: anti-CD4PE, CD45RO ECD, CD62L APC-Cy7, CD25 APC, CD127 Pacific Blue. After incubation, cells were washed twice in PBS/FCS (0.2%), and sorted into five different cell subsets: CD4+CD25+CD127low CD62L+CD45RO- (naive regulatory T cells), CD4+CD25+CD127low CD62L+/- CD45RO+ (activated regulatory T cells), CD4+CD25+CD127hi CD62L+/- CD45RO+ (memory T cells), CD4+CD25-CD127low CD62L+/- CD45RO+ (effector T cells) and CD4+CD25-CD127hi CD62L+ CD45RO- (naive T cells).

miRNA-seq study between Th0 and Th17 cells

Project description:To explore the differences in miRNA profiles, naive CD4+CD62L+ helper T cells were sorted by magnetic cell sorting from spleens of female C57BL/6 mice and were induced to differentiate into Th1 or Th17 cells, then total RNA was extracted and miRNA-seq were conducted. The results showed that many microTNAs presented a different expression pattern between Th1 and Th17 cells, which prompted us to further study their roles in Th117 differenciation. miRNA profiles of Th0 and Th17 cells were generated by deep sequencing, in triplicate, using Illumina HiSeq 2500

Project description:Steroid receptor coactivator 3 (SRC3) was reported to regulates pathogenic Th17 cell differentiation in vitro and in vivo.However, the role of SRC2, another member of SRC family, and its relationship with SRC3 in the regulation of Th17 cell differentiation remain unknown. We used RNA-seq to study the global function of SRC2 and SRC3. Naive CD4+ T cells were activated with 5ug/ml plate-bound anti-CD3/28, and cultured with TGF-β+IL-6 or with IL-6+IL-1β+IL23. 4days later, cells were restimulated with plate-bound anti-CD3 for 4hrs for RNA-seq.

Regulation of gene expression by RORgamma in Th17 cells

Project description:We observed significant growth inhibition of melanoma tumor in ROR?-/- mice, suggesting an important role for the T helper 17 cell (TH17) pathway in tumor immunity. Though RORgamma t (ROR?t) plays a critical role in the development of IL-17-secreting Th17 cells, the role of other genes remained an open question. To explore the expression of unknown genes with anti-tumor properties, gene expression analysis was performed. Sorted naïve TH cells (CD4+CD25-CD62Lhigh) from RORc+/+ and RORc-/-mice were differentiated under TH17 polarizing conditions. After 4 days, cells were harvested, RNA was isolated and gene expression analysis was performed.

The kinase DYRK1A reciprocally regulates the differentiation of Th17 and regulatory T cells [array]

Project description:Using an unbiased chemical biology approach, we discover harmine as a novel regulator of Treg/Th17 differentiation. Harmine enhances Treg differentiation (working in conjunction with low levels of exogenous TGFb) and inhibits Th17 differentiation. Analysis of global gene expression of Tregs generated using low TGFb + harmine reveals significant similarity to Tregs generated using high TGFb only and suggests relevance of harmine-engaged mechanisms to IBD. Naïve CD4+CD62L+ T cells were purified from Foxp3GFP mice either by MACS or FACS, then cultured in either low TGFb + harmine or high TGFb conditions. GFP+ Tregs were sorted by FACS at day 4.
