SphK-produced S1P in somatic cells is indispensable for LH-EGFR signaling-induced mouse oocyte maturation

ABSTRACT: To further investigate the mechanism of SphK1/2 on oocyte maturation, we established Sphk1/2gc -/- and WT mice. We then performed transcriptomic analysis on the MGCs and cumulus cells of Sphk1/2gc-/- and WT mice at 2 h post-hCG.

Targets from two biological replicates at each time point (0h, 8h, 16h post-hCG treatment) were generated and the expression profiles were determined using Affymetrix GeneChip Mouse Genome 430 2.0 Arrays. Comparisons between the sample groups allow the identification of genes with temporal expression patterns. Analysis of transcriptomic diversitification of developing cumulus and mural granulosa cells in mouse ovarian follicles

Project description:Cumulus cells and mural granulosa cells (MGCs) are spatially and functionally distinct cell types in antral follicles: cumulus cells contact the oocyte and most MGCs contact the basal lamina. For transcriptomic analyses, both cell types were collected from small and large antral follicles, before and after stimulation of immature mice with eCG, respectively. Both cell types underwent dramatic transcriptomic changes and the differences between them became greater with follicular growth. Although cumulus cells of both stages of follicular development are competent to undergo expansion in vitro, they were otherwise remarkably dissimilar with transcriptomic changes quantitatively equivalent to those of MGCs. Gene Ontology (GO) analysis showed that cumulus cells of small follicles were enriched in transcripts generally associated with catalytic components of metabolic processes while those from large follicles were involved in regulation of metabolism, cell differentiation, and adhesion. Upon contrasting cumulus cells versus MGCs, cumulus cells were enriched in transcripts associated with metabolism and cell proliferation while MGCs were enriched for transcripts involved in cell signaling and differentiation.

Project description:We sought to identify genes that are regulated by the ovulatory signals in mouse cumulus cells. We compared the transcriptomes between cumulus cells colected from mice before (hCG-0h) and after 6 h (hCG-6h) of hCG injection. 3 individual cumulus cell samples of hCG-0h and hCG-6h were collected and subjected to microarray study. Copmarison was done between hCG-0h and hCG-6h groups.

Expression data from progesterone receptor knockout versus heterozygous mouse ovaries: cumulus oocyte complexes

Project description:Ovulation requires sequential molecular events and structural remodeling in the ovarian follicle for the successful release of a mature oocyte capable of being fertilised. Critical to this process is progesterone receptor (PGR), a transcription factor highly yet transiently expressed in granulosa cells of preovulatory follicles. Progesterone receptor knockout (PRKO) mice are anovulatory, with a specific and complete defect in follicle rupture. Therefore, this model was used to examine the critical molecular and biochemical mechanisms necessary for successful ovulation. Although PGR is not expressed in the cumulus cells or oocyte of the preovulatory cumulus oocyte complex (COC), it is well known that the COC responds to the cascade of gene expression changes that occurs in preovulatory granulosa cells. We used microarrays to identify putative ‘ovulation’ genes in preovulatory COCs at a time when PGR expression is maximal in granulosa cells (eCG + 8h hCG) and the preovulatory COC and follicle are undergoing the final changes necessary for successful ovulation.

Rhesus monkey cumulus cells revert to a mural granulosa cell state following an ovulatory stimulus

Project description:Follicular somatic cells (mural granulosa cells and cumulus cells) and the oocyte communicate through paracrine interactions and through direct gap junctions between oocyte and cumulus cells. Considering that mural and cumulus cells arise through a common developmental pathway and that their differentiation is essential to reproductive success, understanding how these cells differ is a key aspect to understanding their critical functions. Changes in global gene expression before and after an ovulatory stimulus were compared between cumulus and mural granulosa cells to test the hypothesis that mural and cumulus cells are highly differentiated at the time of an ovulatory stimulus and further differentiate during the periovulatory interval. The transcriptomes of the two cell types were markedly different (>1500 genes) before an ovulatory hCG bolus but converged after ovulation to become completely overlapping. The predominant transition was for the cumulus cells to become more like mural cells after hCG. This indicates that the differentiated phenotype of the cumulus cell is not stable and irreversibly established but may rather be an ongoing physiological response to the oocyte.

Change in the ability of granulosa cells to elongate transzonal projections during follicle development

Project description:Cumulus cells (CCs) and mural granulosa cells (MGCs) have different phenotypes and different roles in the follicle. However, our previous reports have suggested that CCs and MGCs from bovine early antral follicles become MGC-like cells and CC-like cells, respectively. In this study, we examined whether granulosa cells (GCs) from secondary follicles and MGCs from late antral follicles were able to reconstruct complexes with transzonal projection (TZP)-free denuded oocytes (DOs) and regenerate TZPs similar to MGCs from early antral follicles. Furthermore, to confirm that the regenerated TZPs were functional, the development of reconstructed complexes and oocyte growth in the complexes were determined. After coculture, GCs were able to reconstruct complexes with DOs and regenerate TZPs similar to MGCs from early antral follicles. In addition, the oocytes in integrally reconstructed complexes grew fully and acquired meiotic competence, suggesting that the regenerated TZPs are functional. In contrast, MGCs from late antral follicles had lost the ability to elongate TZPs. Since the ability to regenerate TZPs differed among the cells, the transcriptome was analyzed among GCs, CCs, and MGCs collected from follicles of different sizes. The characteristic of TZP generation coincided with transcriptome changes in two directions from GCs to CCs and MGCs. In conclusion, until the early antral follicle stage, bovine GCs, CCs, and MGCs have common characteristics to elongate TZPs and form antrum-like structures to support oocyte growth in vitro. Furthermore, as the follicle develops, MGCs lose the ability to elongate TZPs.
