Differences in biliopancreatic duct development exist in Type I congenital biliary dilatation based on microarray expression analysis of mRNAs and long noncoding RNAs

ABSTRACT: Congenital biliary dilatation (CBD) is a malformation presenting extra- and/or intrahepatic dilatation bile ducts and often associated with pancreaticobiliary maljunction. Different types of CBD have been noted clinical differences. LncRNA and mRNA sequencing and bioinformatics analysis in CBD can help us to further study the pathogenesis of different types of CBD at the genetic level in the context of clinical data.

Genomic DNA methylation profile in peripheral blood of children with Congenital biliary dilatation

Project description:Congenital biliary dilatation (CBD) is a relatively rare biliary tract disease. It is a prevalent congenital biliary disease in children. CBD mainly affects in infants and children, with a higher prevalence in females. To investigate the DNA methylation patterns in children with CBD and in healthy children, we performed a comparative analysis of the genome‐wide DNA methylation profiles of whole blood samples from patients with CBD child and from healthy children using the Illumina Infinium Human Methylation 850k BeadChip. Bioinformatics statistical analysis revealed that 5,863 genes with differential DNA methylation between the two groups correlated to a total of 24,230 methylated sites.Of these differential methylation sites, 831,3 were hypermethylated and 15,917 were hypomethylated in comparison to the methylation status of those sites in the control samples. Differentially methylated regions (DMRs) between the CBD cases and the control group was significant difference in DMR_1~DMR_54. Furthermore, T cell receptor signaling pathways represented accounted for an important percentage of the differentially methylated genes. Abnormalities in the DNA methylation pattern identified in the peripheral blood of CBD patients may play an important role in the development of CBD and warrant further investigation.

Clinical and Imaging Features of a Focal Intrahepatic Biliary Stricture Visualized Only as Duct Dilatation.

Project description:PurposeWe assessed the proportion of patients with a focal intrahepatic stricture (FIHS) that was a precursor lesion or malignancy and visualized only as a duct dilatation.Materials and methodsThis retrospective study assessed patients who underwent surgery or biopsy for an FIHS on CT or MRI between January 2010 and March 2022. The number and proportion of non-precursor benign lesions, precursors, and malignancies were calculated. Clinical variables and imaging features were compared between non-premalignant benign and premalignant/malignant FIHSs.ResultsTwenty-eight patients with confirmed histopathological diagnoses were identified, including 15 men (54.0%) and 13 women (46.0%). The median age of all patients at the first imaging diagnosis was 65 ± 9.54 (range, 43-78) years. Of the 28 patients with FIHSs, 9 (32%) were diagnosed with cholangiocarcinoma and 7 (25%) were diagnosed with precursor lesions, which included six intraductal papillary neoplasms of the bile duct and one biliary intraepithelial neoplasm. Accordingly, 16 (57%) patients had malignant or precursor lesions, and 12 (43%) were diagnosed with non-precursor benign lesions. None of the clinical variables and imaging features used for analysis showed a statistically significant difference between the non-premalignant benign and premalignant/malignant FIHS groups (p > 0.05).ConclusionFIHSs visualized only as duct dilatation can harbor malignant or precursor lesions.

Circulating microRNAs in patients with aneurysmal dilatation of coronary arteries

Project description:To understand the mechanism of coronary artery aneurysmal dilatation, we identified and compared the expression of circulating miRNAs in three groups of patients. Group 1 enrolled patients with aneurysmal dilatation of coronary arteries (n = 20), Group 2 included patients with angiographically confirmed coronary artery disease (CAD), whereas Group 3 included 20 patients with normal coronary arteries. The miRNAs were isolated from plasma and profiled using PCR arrays miRCURY LNA Serum/Plasma Focus PCR Panels. We demonstrated that the plasma miRNAs levels were significantly different in Group 1 in collation with Group 2 and Group 3 (fold change > 2 and p < 0.05). The comparison of Group 1 with Group 3 identified twenty-one significantly up-regulated and two down-regulated miRNAs in patients with aneurysmal coronary artery dilatation compared to the control groups. Moreover, we identified six up-regulated and two down-regulated miRNAs in patients with CAD compared to the controls. The third comparison revealed four up-regulated and three down-regulated miRNAs in Group 1, when compared to CAD patients. In conclusion, this study demonstrates the specific signature of plasma miRNA, namely up-regulated and down-regulated, in patients with abnormal dilatation of coronary arteries and the comparison between the groups consisting of atherosclerotic and control patients.

Identification of molecular pathways involved in oxaliplatin-associated sinusoidal dilatation

Project description:Oxaliplatin-based chemotherapy for colorectal liver metastases (CRLM) can result in vascular liver lesions such as sinusoidal dilatations. Physiopathology remains unclear and variability between patients suggests that there is individual susceptibility. A better understanding of the molecular mechanisms of oxaliplatin liver toxicity may allow to identification of biomarkers and adaptation of chemotherapy delivery. non-tumor frozen liver samples were obtained from patients operated on for CRLM after an exclusive oxaliplatin-based chemotherapy. Gene-expression profiles were first analyzed by microarray on a selected population of 20 patients; 9 patients with severe sinusoidal dilatation (SD) after a short period of chemotherapy and 10 patients without any sinusoidal dilatation after a long period of chemotherapy. These were compared with a control group of 5 patients without any chemotherapy and lesions. This dataset is part of the TransQST collection.

Liver-restricted deletion of the biliary atresia candidate gene Pkd1l1 causes bile duct dysmorphogenesis and ciliopathy

Project description:A recent multicenter genetic exploration of the biliary atresia splenic malformation (BASM) syndrome identified mutations in the ciliary gene PKD1L1 as candidate etiologic contributors. We hypothesized that deletion of Pkd1l1 in hepatoblasts would provide a mouse model of the developmental cholangiopathy of BA. We then performed gene expression profiling analysis using data obtained from total RNA-seq, isolated from the livers of Pkd1l1Fl/Fl (Fl/Fl) and Pkd1l1Fl/Fl x AFP-CRE mice (Pkd1l1ΔExon8/ΔExon8 Liver knockout ; LKO) at 7 weeks.

Risk Factors associated with Paraurethral Duct Dilatation following Gonococcal Paraurethral Duct Infection in Men.

Project description:No studies have explored the risk factors for paraurethral duct dilatation following paraurethral duct infection by Neisseria gonorrhoeae in men undergoing ceftriaxone therapy. The present study was performed to explore the risk factors for paraurethral duct dilatation following paraurethral duct infection by N. gonorrhoeae in men undergoing ceftriaxone therapy and thus guide clinical interventions. We compared the demographic, behavioral, and clinical data of men with paraurethral duct infection by N. gonorrhoeae with and without dilatation of the paraurethral duct. Univariate analysis showed significant differences in age, disease course of the infected paraurethral duct, Chlamydia trachomatis infection in the paraurethral duct, and a history of paraurethral duct infection by N. gonorrhoeae between the patient and control groups (P<0.05). Multivariate logistic regression analysis showed consistent results (P<0.05). This study that shows delayed treatment may be a major risk factor for paraurethral duct dilatation secondary to paraurethral duct infection by N. gonorrhoeae in men. Age, C. trachomatis infection in the paraurethral duct, and a history of paraurethral duct infection by N. gonorrhoeae are also risk factors. Thus, educating patients to undergo timely therapy and treating the C. trachomatis infection may be effective interventions.
