Project description:Focal adhesion kinase (FAK) is a non-receptor tyrosine kinase that plays an important role in proliferation, motility, adhesion, invasion, angiogenesis, and survival signaling. Focal adhesion kinase has been shown to be overexpressed in many types of tumors, including breast cancer at early stages of tumorigenesis. To study the biological role of FAK in breast tumorigenesis, we used FAKsiRNA to down-regulate FAK in MCF-7 cell lines. Gene expression changes in human K562 cells by a synthetic transcription factor (peptide), that mimics the Homeobox protein DLX4 or its isoforms, DLX7 or BP1.

Project description:Homeobox proteins are known as master regulators of developmental morphogenesis. DLX genes encode transcription factors that significantly regulate the hematopoitic system. DLX4 and its isoform BP1 are expressed in different stages of development in human hematopoitic cells and regulate further differentiation. BP1 is the repressor of adult beta and delta globin genes found in fetal and embryonic tissues and rarely observed in adult tissues. In K562, a human chronic myelogenous leukemia cell line, BP1 is found to be over-expressed. In this study we have designed and synthesized a peptide-based artificial transcription factor that mimics the DNA binding domain of DLX4 homeodomain and can antagonise its binding. For the micro-array analysis we have taken peptide treated K562 cells and as control untreated K562 cells. Our study provides gene expression profile upon treatment of the homeodomain mimic.

Genome-wide mapping of HIST1H2ac binding sites in MCF-7 cells

Project description:Previously, we have shown that HIST1H2ac is overexpressed in MCF-7 breast cancer cell line. It acts as a master regulator of estrogen receptor alpha-dependent gene expression in ER+ breast cancer cells. In the present study, we investigate the genome-wide protein DNA-binding events of HIST1H2ac protein in MCF-7 breast cancer cell line by over-expressing hemagglutinin (HA)-tagged HIST1H2ac and compared with MCF-7 cells over-expressing HA. The protein-bound DNA was recovered by immunoprecipitation using anti-HA antibody. The ChIP DNA and input DNA were sequenced with an Illumina HiSeq 2000 sequencer.
