Project description:Injuries to the skin can result in non-healing wounds, characterized by prolonged inflammation, failure to close, and chronic pain. Basal keratinocytes in the epidermis respond to signals activated following injury by proliferating, migrating, and differentiating to restore the epidermal barrier. The skin is densely innervated by peripheral sensory nerves, which contribute to the wound repair response. Although it is known that nerves are important for successful wound healing, the underlying cellular mechanisms of this phenomenon, and particularly the role of nerves in directing keratinocyte re-epithelialization, are poorly understood. To explore the relationship between sensory nerves and keratinocyte function in vitro, we cultured keratinocytes with conditioned media collected from dorsal root ganglia (DRG) in both homeostatic and post-wounding conditions and found that keratinocyte migration and proliferation, functions essential for re-epithelialization, were modulated by DRG conditioned media. Using a proteomic approach, we characterized the secretome of cultured DRG and identified key factors essential for wound healing, including extracellular matrix proteins, growth factors, and metabolic factors involved with ATP production, which was correlated with an increase in ATP rates of keratinocytes cultured in DRG conditioned medium. Our results advance our understanding of the microenvironmental cues that direct keratinocyte function during key events of cutaneous wound healing in vitro to drive the development of therapeutics that target dysregulated re-epithelialization in non-healing wounds.

Project description:The cell transition from an inflammatory phase to a subsequent proliferative phase is crucial for wound healing, yet the driving mechanism remains unclear. By profiling lncRNA expression changes during human skin wound healing and screening lncRNA functions, we identified SNHG26 as a pivotal regulator in keratinocyte progenitors underpinning this phase transition. To study the proteins interact with SNHG26, we performed RNA pull-down assay in human keratinocyte progenitors. The mass spectrometry (MS) analysis was performed to identify the proteins interacted with SNHG26.

Project description:The epidermal stem cell transition from inflammation to proliferation is a crucial process in tissue repair. However, the molecular mechanism underlying this process is poorly understood. Combined with lncRNA expression profiling of human acute wounds and functional screening, we identified SNHG26 as a pivotal regulator of inflammatory to proliferative state transition of keratinocyte progenitor cells. Snhg26-deficient mice showed impaired wound healing characterized by increased inflammatory response and decreased reepithelization in the wound. Consistently, single-cell transcriptomic analysis of the wound-edge tissue identified proliferative, migratory, and proinflammatory basal progenitors, which were dramatically changed in Snhg26 deficient mice. Mechanically, SNHG26 binds with ILF2 to relocate this transcription factor from the inflammatory gene loci (such as JUN) to the LAMB3 genomic loci. Collectively, this work identified a conserved lncRNA SNHG26 as a master regulator of epidermal stem cell state transition from inflammation to proliferation, highlighting the importance of lncRNAs in tissue repair and regeneration.

Project description:Epidermal keratinocytes form cornified skin appendages such as scutate scales on the legs of birds. Here, we investigated the molecular pathways of keratinocyte differentiation in chicken scutate scales by single cell transcriptomics. We identified two distinct populations of differentiated keratinocytes. The first type of differentiated keratinocytes is characterized by mRNAs encoding scale-type corneous beta-proteins (CBPs), also known as beta-keratins and cysteine-rich keratins, indicating that these cells form hard scales. The second type of differentiated keratinocytes contains mRNAs encoding keratinocyte-type CBPs and cysteine-poor keratins, indicating that these cells form the soft interscale epidermis. Immunostaining with a newly raised antibody confirmed that keratin 9-like cysteine-rich 2 (KRT9LC2) or Hard Acid Sauropsid-specific 2 (HAS2) keratin, which is a marker of the first type of keratinocytes, is expressed in the suprabasal epidermal layers of scutate scales but not in interscale epidermis. Furthermore, mRNA of CTNN1B, previously implicated in scale placode formation, was enriched in differentiated scale keratinocytes, whereas genes involved in lipid metabolism, such as ELOVL4 and FADS1 were enriched in keratinocytes of the interscale epidermis. In conclusion, this study defines the gene expression programs that build the scutate scales and interscale epidermis of birds.

Project description:In the present study, we demonstrate that hMSCs migrate toward human keratinocytes as well as toward conditioned medium from cultured human keratinocytes (KCM) indicating that the hMSCs can respond to signals from keratinocytes. Incubation of hMSCs with KCM induced dermal myofibroblast like differentiation characterized by expression of cytoskeletal markers vinculin and F-actin filaments with increased expression of alpha smooth muscle actin. We then examined the therapeutic efficacy of hMSCs in wound healing in two animal models representing normal and chronic wound healing. Accelerated wound healing, as determined by quantitative measurements of wound area, was observed when hMSCs and KCM exposed hMSCs (KCMSCs) were injected near the site of incisional/excisional wounds in nondiabetic athymic and NOD/SCID mice as compared with normal human fetal lung fibroblast WI38 cells or saline control induced wound healing. Project description:Impaired diabetic wound healing compromises patient quality of life, with keratinocytes playing a pivotal role in the wound repair process. This in vitro study investigated the role of ETS variant transcription factor 4 (ETV4) in keratinocyte migration/proliferation in high glucose (HG) environments to assess whether ETV4-mediated signaling is involved in dysfunctional wound repair. Western blotting showed that ETV4 expression was downregulated in human keratinocytes cultured in HG. ETV4 knockdown using small interfering RNA suppressed keratinocyte migration/proliferation. Intervention with the reactive oxygen species (ROS) scavenger tempol or the Akt activator SC79 restored ETV4 expression and rescued impaired migration/proliferation. Tempol also restored Akt phosphorylation levels. Matrix metalloproteinase-14 (MMP14) was identified in transcriptome sequencing/bioinformatic analyses as a downstream target of ETV4. HG-suppressed MMP14 expression was reversed by tempol or SC79. Specific inhibition of MMP14 attenuated keratinocyte migration/proliferation in HG. This study reveals a critical role for the ROS/Akt/ETV4/MMP14 signaling axis in HG-induced keratinocyte dysfunction, suggesting potential therapeutic targets for diabetic wound healing.
