Project description:Sperms being foreign to the female are to be promptly eliminated by the female local immune defense. However, avoiding the local immune defense sperm can be stored for lenthy period in the oviductal sperm reservoir. It is currently unknown whether oviductal sperm reservoirs changes their gene expression to tolerate the spermatozoa after mating or sperm free seminal plasma infusion. Therefore this was tested using Swedish Landrace pigs in this study using cDNA microarray. We used 12 sows seperated into three groups- either oestrus sows were inseminated with 50 ml BTS (control, n=4) or mated with boars (treatment 1, n=4) or inseminated with sperm-free seminal plasma (treatment 2, n=4). The utero-tubal junction was retrieved within 24 h of treatment by operation.

Project description:EVs are known to contain important cargo of biologically active and regulatory molecules including ncRNAs. EVs in seminal plasma, play an important role in sperm functions , including motility and fertilization. However, less is known about the comprehensive profile of spEV ncRNAs in clinical cohort and if these profiles differ by semen quality status. We assessed sperm-free spEV ncRNA profiles from semen samples of male partners receiving IVF treatment. Men were classified into having normal (n = 59, Status 0) or poor (n = 32, Status 1) semen quality based on WHO semen parameter guidelines.

Project description:Seminal plasma contains a large number of extracellular vesicles (EVs). However, the roles of these EVs and their interactions with sperm are not clear. To identify the important molecules affecting sperm motility in EVs, we performed proteomics analysis of SPEVs from the two groups with different sperm motilities. In total, 1435 exosomal proteins covered a broad range of protein classes, such as enzymes, cytoskeletal proteins, extracellular proteins and transcription factors. We identified 76 DEPs (differentially expressed protein) from the SPEVs of boars with high- and low-sperm motility phenotypes (|FC| ≥1.5, P≤ 0.05). A total of 38 proteins were downregulated in the L group compared with the H group, while 6 proteins were significantly upregulated. In addition, 28 proteins were identified only in the H group, and 4 proteins were identified only in the L group. The GO and KEGG analyses showed that the DEPs were associated primarily with negative regulation of cell motility, extracellular exosomes, the extracellular space, cell-cell adhesion and phagosome biological activities. The present fndings represent a major and novel contribution to the study of boar seminal extracellular vesicles proteins.

Project description:Quantitative proteomic studies are contributing greatly to our understanding of the spermatozoon through the provision of detailed information on the proteins spermatozoa acquire and shed in the acquisition of fertility. Extracellular vesicles (EVs) are thought to aid in the delivery of proteins to spermatozoa in the male reproductive tract. The aim of this study was to identify EV proteins isolated from ram seminal plasma. High-speed centrifugation using Optiprep sucrose gradients was used to purify two EV populations from seminal plasma. Liquid chromatography and tandem mass spectrometry (LC-MS/MS) with quantitative SWATH was used to identify proteins enriched in the extracellular vesicle preparation (e.g. EDIL3) and those that were co-isolated with vesicular preparations due to their abundance or adhesive nature (e.g. BSPs). Ram sperm plasma membrane proteins were also isolated using nitrogen cavitation and identified with LC-MS/MS to better understand the interplay of proteins between the sperm membrane and extracellular environment. The categorisation of proteins enriched in the EV population (P<0.05, 2-fold increase compared to whole seminal plasma) according to their function revealed three main groupings: vesicle biogenesis, metabolism and membrane adhesion and remodelling. The latter group contained many reproduction-specific proteins that show demonstrable links to sperm fertility. Many of these membrane-bound proteins show testicular expression and are shed from the sperm surface during epididymal maturation (e.g. TEX101, LY6K). Their association with seminal EVs suggests that EVs may not only deliver protein cargo to spermatozoa but also assist in the removal of proteins from the sperm membrane.

Project description:Seminal plasma contributes to the safe environment for sperm maturation, sperm viability and fertilization in mammals (Kumar 2013). Moreover, seminal plasma is a promising source for the study of potential reproductive biomarkers, because it is a complex mixture of secretions from testis, epididymis and male accessory sex glands (González-Cadavid et al. 2014). The protein composition of mammalian seminal plasma varies among species, and has important effects on sperm function (Mortarino et al. 1998). In rabbits, seminal plasma has a positive effect in maintaining sperm motility and viability during in vitro storage (Castellini et al. 2000). To study proteins on a large scale, the application of a proteomic approach is mandatory (Dietrich 2014). Rabbit seminal plasma proteome has been proved different between rabbit genetic line A (maternal) and R (growth) (Viudes-de-Castro et al. (2004), Safaa et al. (2008), Casares-Crespo 2016) and between seasons (Casares-Crespo 2016). In these previous studies a traditional 1 dimension polyacrylamide gel was done, identifying only major proteins visible after Coomassie Colloidal Blue staining and obtaining the relative quantity of these protein bands. On the contrary, new approaches such as liquid chromatography in-gel or in-solution digestion coupled with mass spectrometric analysis are a powerful, yet relatively uncomplicated strategy to identify and characterize proteins. Mass spectrometry-based proteomics has become the tool of choice for identifying and quantifying the proteome of an organism (Karpievitch 2010). Therefore, the present study was conducted to describe the seminal plasma proteome of rabbits, compare the differentially expressed seminal plasma proteins between genetic lines A and R and study the effect of season in rabbit seminal plasma proteome.

Project description:We investigated the nutritional effects on gene expression in sperm cells of F0 boars from a three generation Large White pig feeding experiment. A group of experimental (E) F0 boars were fed a standard diet supplemented with high amounts of methylating micronutrients whereas a control (C) group of F0 boars received a standard diet. These differentially fed F0 boars sired F1 boars which then sired 60 F2 pigs which were investigated in a previous study. The aim of this study was to investigate if the nutrition affects gene expression in sperm cells of differentially fed boars and thus carry information in the form of RNA molecules to the next generation. Four RNA samples from sperm cells of these differentially fed boars were analyzed by RNA-Seq methodology. We found no differential RNA expression in sperm cells of the two groups based on the adjusted P-value > 0.05. Nevertheless, we performed a pathway analysis with 105 genes that differed in gene expression on the level of nominal P-value < 0.05 between the two diet groups. We found a significant number of these differentially expressed genes were enriched for the pathway maps of bacterial infections in cystic fibrosis (CF) airways, glycolysis and gluconeogenesis p.3 and cell cycle_Initiation of mitosis. The GO processes including a significant portion of differentially expressed genes were viral transcription and viral genome expression, viral infectious cycle, cellular protein localization, cellular macromolecule localization, nuclear-transcribed mRNA catabolic process and nonsense-mediated decay. In summary, the results of the pathway analysis are also inconclusive and it is concluded that RNA expression in sperm cells is not significantly affected by extensive supplementation of methylating micronutrients. Consequently, RNA molecules could not be established as epigenetic marks in this feeding experiment. Gene expression in sperm cells from differentially fed F0 boars was measured. F0 boars received either a standard diet or a standard diet supplemented with methylating micronutrients. These boars were used to study transgenerational epigenetic inheritance in a three generation pig pedigree. Therefore it was of interest if the diet affects gene expression in sperm cells which could then be transmitted to next generations.
