Project description:Meiosis is a highly complex process that underpins recombination in sexually reproducing organisms. Recent genomics studies suggest that up to several thousand genes/proteins contribute to the meiotic pathway, yet relatively few have been functionally characterised. Our understanding of the physical interactions between meiotic proteins and how the meiotic machinery interacts with other components of the cell is also limited. In this study, we used affinity proteomics targeting the meiotic chromosome axis protein, ASY1, to enrich for axis-associated proteins in Brassica oleracea anthers or meiocytes in prophase I of meiosis. LC-MS/MS analysis identified 540 proteins which co-immunoprecipitated with ASY1 in a sample-specific manner. These correspond to 485 Arabidopsis orthologues, 90% of which form a coherent predicted protein-protein interaction network which includes known and novel meiotic proteins, based on mutant analysis, but also proteins more usually associated with other cellular processes including replication and proteolysis. Our data provided new insights into the role of the chromosome axis in plant meiosis. We identified a novel axis-associated protein and showed that during prophase I, ASY1 and its interacting partner, ASY3, are extensively phosphorylated. Further studies targeting other meiotic proteins may ultimately enable the construction of a comprehensive meiosis protein-protein interaction network for higher plants.

Project description:We report gene expression profiling in the fission yeast Schizosaccharomyces pombe. We performed high-throughput sequencing of RNA isolated from wild-type, erh1∆, and ccr4∆ strains. We find that many meiotic gene containing degradation sequence DSR are expressed in vegetative erh1∆, while these meiotic mRNAs do not increase in ccr4∆, indicating that Erh1 and Ccr4 target different set of genes during vegetative growth.

Project description:The CCR4-NOT complex is a major mRNA deadenylase in eukaryotes and comprise CNOT6/6L and CNOT7/8, both of which are two types of catalytic subunits, as well as CNOT4, which is a regulatory subunit with previously undetermined functions. These subunits are previously hypothesized to play synergistic biochemical functions; however, their in vivo functions have remained poorly investigated. In this study, we constructed viable Cnot6/6l double knockout mice and the males were fertile, whereas Cnot7–/– male mice have been reported to be infertile. These results are against canonical viewpoints and indicate that CNOT7 has CNOT6/6L-independent functions in vivo. We also demonstrate that the mouse testis has an endogenous complex that contains CNOT7 and CNOT4, but not CNOT6/6L. CNOT4 is required for post-implantation embryo development and meiosis progression during spermatogenesis. Selective knockout of Cnot4 in male germ cells leads to defective DNA damage repair and homologous crossover formation between X and Y chromosomes in a cell autonomous manner. CNOT4 functions as a previously unrecognized mRNA adaptor of CCR4-NOT by targeting mRNAs to CNOT7 for deadenylation of the poly(A) tails, thereby mediating the degradation of a subset of transcripts from the zygotene to pachytene stage. Therefore, the mRNA removal promoted by CNOT4-regulated CCR4-NOT complex during the zygotene to pachytene transition is crucial for the selection of DNA repair pathways, subsequent activation of meiosis-related genes, sex chromosome paring, and ultimately, male fertility.

Project description:Telomeres have a prominent role in meiosis – telomere tethering to nuclear envelope proteins is essential for successful meiosis and fertility. This is accomplished through meiosis-specific telomere adaptor complexes that interacts with protein complexes spanning the nuclear envelop to connect with motor proteins. Connecting telomeres to motor proteins through this protein chain drives rapid prophase I movements (RPM) of chromosomes, which results in clustering of telomeres at one side of the nucleus to form the meiotic bouquet. Telomere attachment to the nuclear envelop proteins and RPMs are essential for homologue chromosome pairing, synapsis, and meiotic recombination. Mutations disrupting telomerase reverse transcriptase (tert) result in multiple pathologies including impaired fertility. Here, we investigate effects of loss of tert function on meiosis in zebrafish. We demonstrate that tert mutant meiotic germ cells have shortened telomeres and display defects in meiotic progression with eventual loss of meiotic cells with age, followed by loss of mitotic germ cells several months later. These data indicate that meiotic cells are more sensitive to shortened telomeres than mitotic germ cells. Furthermore, we found that orthologues to the mouse meiotic telomere adaptor complex genes, terb1, terb2, and majin, are expressed primarily in meiotic germ cells in zebrafish. Through mutant analysis we show that zebrafish terb1 is necessary for formation of the meiotic bouquet and completion of meiosis, suggesting that zebrafish Terb1 functions as a meiotic telomere adaptor protein, similar to mouse Terb1. These data point to the importance of telomere maintenance and telomere associated proteins in meiotic prophase I during spermatogenesis.

Project description:Meiotic progression in mammals is governed by tightly coordinated transcriptional programs and surveillance mechanisms that ensure the successful pairing, synapsis, and segregation of homologous chromosomes. In spermatocytes, homolog pairing is safeguarded by the pachytene checkpoint, whose mechanism is critically involves meiotic sex chromosome inactivation (MSCI), a silencing mechanism that targets the largely unsynapsed X and Y chromosomes during pachytene. HORMAD2, a HORMA domain-containing protein, plays a key role in MSCI. Although in vitro studies have implicated SYCP2 in recruiting HORMAD proteins to chromosome axes, the mechanism and physiological relevance of this interaction during meiosis has remained unclear. To gain transcriptome-level insights into how loss of HORMAD2 axial recruitment affects meiotic progression and sex chromosome regulation, we performed single-cell RNA sequencing (scRNA-seq) of testicular cell suspensions of adult wild-type and Hormad2-/-mice as well as mice where SYCP2 lacked the region that is predicted to bind HORMAD2 (Sycp2e16/e16 mutant). These datasets allowed us to resolve 17 distinct germ cell states spanning the full course of spermatogenesis. We observed that, unlike wild-type cells which transcriptionally progressed into mid-pachytene and beyond, neither Sycp2e16/e16 nor Hormad2-/- spermatocytes progressed beyond an early pachytene-like transcriptional state. We also saw clear evidence of impaired meiotic sex chromosome inactivation (MSCI) in the mutants. Specifically, sex-linked gene expression abnormally persisted in the mutants resulting in overexpression of Y-linked genes Zfy1 and Zfy2, whose high expression induces spermatocyte apoptosis in mid pachytene. Project description:An in vitro model of human meiosis would accelerate research into this important reproductive process and development of therapies for infertility. We have developed a method to induce meiosis starting from male or female human pluripotent stem cells. We demonstrate that DNMT1 inhibition, retinoid signaling activation, and overexpression of regulatory factors (anti-apoptotic BCL2, and pro-meiotic HOXB5, BOLL, or MEIOC) rapidly activates meiosis, with leptonema beginning at 6 days, zygonema at 9 days, and pachynema at 12 days. Immunofluorescence microscopy shows key aspects of meiosis, including chromosome synapsis and sex body formation. The meiotic cells express genes similar to meiotic oogonia in vivo, including all synaptonemal complex components and machinery for meiotic recombination. These findings establish an accessible system for inducing human meiosis in vitro.
