Meteorin-like enhances aged muscle regeneration

ABSTRACT: Pathologies associated with sarcopenia include decline in muscular strength, lean mass and regenerative capacity. Despite the substantial impact on quality of life, no pharmacological therapeutics are available to counteract the decline of functional capacity, loss of independence and resilience. Evidence suggests immune-secreted cytokines can improve muscle regeneration; a strategy which we leverage in this study by rescuing the age-related deficiency in Meteorin-like (Metrnl) through several in vivo add-back models. Notably, a direct, intramuscular peptide injection of recombinant METRNL was sufficient to improve muscle regeneration in aging. This RNAseq dataset substantiates that rMETRNL treatment via intramuscular injections is sufficient to rejuvenate the aged muscle regenerating transcriptome to that of young controls.

Project description:Skeletal muscle aging results in a gradual loss of skeletal muscle mass, skeletal muscle function and decreased regenerative capacity, which can lead to sarcopenia and increased mortality. While the mechanisms underlying sarcopenia remain unclear, the skeletal muscle stem cell, or satellite cell, is required for muscle regeneration. Therefore, identification of signaling pathways affecting satellite cell function during aging may provide insights into therapeutic targets for combating sarcopenia. Here, we show that a cell-autonomous loss in self-renewal occurs via novel alterations in FGF and p38αβ MAPK signaling in old satellite cells. We further demonstrate that pharmacological manipulation of these pathways can ameliorate age-associated self-renewal defects. Thus, our data highlight an age-associated deregulation of a satellite cell homeostatic network and reveals potential therapeutic opportunities for the treatment of progressive muscle wasting. Satellite cells were isolated from young (3-6mo) and aged (20-25mo) adult mice; individual date files represent 2 independent pools of RNA from 4-8 mice at each timepoint.

Transcription profiling of rat sarcopenic skeletal muscle after treatment with extract of Ginko biloba Egb 761 shows that a juvenile gene expression profile is reactivated

Project description:The sarcopenia-associated decline of the muscular performances in older people is improved by exercise and caloric restriction but is still lacking a more suitable pharmacological treatment. We investigated the effects of EGb 761, a Gingko biloba extract used to treat chronic age-dependent neurological disorders, on the soleus muscle of aged rats. DNA microarray analysis shows that the EGb 761 treatment is associated with the transcriptional reprogramming of genes related to regeneration and energy production functions. EGb 761 up-regulated genes implicated in myogenesis though the TGF? signaling pathway. Also, EGb 761, by modulating genes implicated in fatty acids and glucose oxidation, reversed the agerelated shift from lipids to glucose and improved energy production in mitochondria. These modifications restore a more juvenile gene expression pattern that is concomitant with the improvement of the muscular performances, as assessed by electrophysiological and biochemical parameters. Thus, EGb 761 may represent a novel treatment for sarcopenia

Profiling of glycerol-induced models of muscle regeneration upon melatonin treatment for 14 days in mice

Project description:Intramuscular injection of glycerol (Gly) has been reported to lead to skeletal muscle regeneration and ectopic fat deposition. We utilized glycerol injections in the tibialis anterior muscles to investigate the effects of melatonin on skeletal muscle regeneration and intramuscular fat infiltration. We characterized genome-wide expression profiles of tibialis anterior muscles from wild-type mice injured by glycerol injection following melatonin intervention. Mice that were treated with melatonin exhibited improved muscle regeneration and reduced intramuscular fat deposition, which were associated with enhanced myogenesis, remodeled lipid metabolism and reduced immune cell infiltration.

Loss of NAD homeostasis leads to progressive and reversible degeneration of skeletal muscle

Project description:NAD is an obligate co-factor for the catabolism of metabolic fuels in all cell types. However, the availability of NAD in several tissues can become limited during genotoxic stress and the course of natural aging. The point at which NAD restriction imposes functional limitations on tissue physiology remains unknown. We examined this question in murine skeletal muscle by specifically depleting Nampt, an essential enzyme in the NAD salvage pathway. Knockout mice exhibited a dramatic 85% decline in intramuscular NAD content, accompanied by fiber degeneration and progressive loss of both muscle strength and treadmill endurance. Administration of the NAD precursor nicotinamide riboside rapidly ameliorated functional deficits and restored muscle mass, despite having only a modest effect on the intramuscular NAD pool. Additionally, lifelong overexpression of Nampt preserved muscle NAD levels and exercise capacity in aged mice, supporting a critical role for tissue-autonomous NAD homeostasis in maintaining muscle mass and function. Messenger RNA was isolated from quadriceps muscle of mice from three different age groups and three different genotypes. Wildtype mice were aged 4, 7, and 24 months. Mice deficient for Nampt in skeletal muscle (mNKO) were aged 7 months. Mice overexpressing Nampt in skeletal muscle were aged 4 and 24 months.

Restoration of CPEB4 Prevents Muscle Stem Cell Senescence during Aging

Project description:Muscle stem cells (MuSCs) are required for muscle regeneration. In resting muscles, MuSCs are kept in quiescence. After injury, MuSCs undergo rapid activation, proliferation and differentiation to repair damaged muscles. Age-associated impairments in stem cell functions correlate with a decline in somatic tissue regeneration capacity during aging. However, the mechanisms underlying the molecular regulation of adult stem cell aging remain elusive. Here, we obtained quisecent MuSCs from young, old, geriatric mice for high resolution mass spectrometry Bruker timsTOF Pro. By comparison of young proteome to old MuSCs proteome or geriatric MuSC proteome, we identified the pathways that are differentially during aging.

Profiling of mouse muscle stem cells aging and activation upon skeletal muscle injury

Project description:Utilizing glycerol intramuscular injections in M. musculus provide a models of skeletal muscle damage followed by skeletal muscle regeneration. In particular, glycerol-induced muscle injury triggers accute activation of skeletal muscle stem cells, called satellite cells. However, aging dramatically impairs the regenerative capacity of satellite cells. We characterized genome-wide expression profiles of young and old satellite cells in the non-proliferative and activated state, freshly isolated to non-injured or damaged muscles, respectively. Our goal was to uncover new regulatory signaling specific to satellite cells entry into the activation and myogenic program that are affected with age.

Aging disrupts gene expression timing during muscle regeneration

Project description:Skeletal muscle function and regenerative capacity decline during aging, yet factors driving these changes are incompletely understood. Muscle regeneration requires temporally coordinated transcriptional programs to drive myogenic stem cells to activate, proliferate, fuse to form myofibers, and to mature myonuclei, restoring muscle function after injury. We assessed global changes in myogenic transcription programs distinguishing muscle regeneration in aged mice from young mice by comparing pseudotime trajectories from single-nucleus RNA sequencing of myogenic nuclei. Aging-specific differences in coordinating myogenic transcription programs necessary for restoring muscle function occur following muscle injury, likely contributing to compromised regeneration in aged mice. Differences in pseudotime alignment of myogenic nuclei when comparing aged to young mice via Dynamic Time-Warping revealed pseudotemporal differences becoming progressively more severe as regeneration proceeds. Disruptions in timing of myogenic gene expression programs may contribute to incomplete skeletal muscle regeneration and declines in muscle function as organisms age.
