Transcriptome analysis reveals hub genes and Ihh and KLF9 axis in the endometrial restoration in IUA mice

ABSTRACT: To investigate the molecular mechanism during the restoration of mouse endometrium by CRCs transplantation, uterine horns were collected from the mice of CRC cells- transplanted, injury and sham operated groups at 7 days after surgery for RNAsequencing (RNA-seq). We then performed gene expression profiling analysis using data obtained from RNA-seq of three groups mice.

Although differences in gene expression exist between the endometrium of uterine horns ipsilateral and contralateral to the CL in cattle, they do not impact conceptus survival or length between Days 7 and 14.

Endometriosis: normal and ectopic endometrium

Project description:To explore the difference between ectopic and normal endometrium and find out the characteristics and critical changes in ectopic endometrium, we analyzed transcripts of stromal cells from two groups of patients. The ectopic endometrium and normal endometrium had similar tissue structures and compositions, unlike normal ovaries. The similarity of gene expression profile between the two groups is as high as 97.2%, only 2.8% is significantly different. The differentially expressed genes in the ectopic endometrium are mainly enriched in the adhesion process and the adhesion pathway, which has the adhesion characteristics. Among these differentially expressed genes, CNTN1 is the most variable and participates in multiple pathogenic pathways, which is considered as the core gene to enhance stromal cell adhesion. The up-regulated expression of CNTN1 in all stages of ectopic endometrium is confirmed.

Apelin-13 modulates the endometrial transcriptome of the domestic pig during the implantation phase of pregnancy

Project description:Apelin is a hormone secreted primarily by white adipose tissue. Recent studies have shown that adipokine and its receptor, APJ, are expressed in different reproductive tissues, including the ovary and uterus, which suggests that it may act as an important local regulator of reproductive functions. This study aimed to explore the influence of apelin on global gene expression in the porcine endometrium during early gestation, on days 15 to 16 of pregnancy (implantation period). We analysed samples obtained from 4 individuals (n=4). The tissue explants obtained from the uterine horns were divided into controls [without treatment] and apelin-13 (20 ng/mL) treated ones. In vitro tissue cultures were conducted for 24 hours.

Endometrial protein expression and phosphorylation landscape decipher aberrant insulin and mTOR signaling in patients with recurrent pregnancy loss

Project description:Research question: What are the proteomic and phosphoproteomic differences between the endometrium of women with recurrent pregnancy loss (RPL) and healthy control women during the proliferative (P) and secretory (S) phases of the menstrual cycle? Design: The present study collected a total of 54 endometrial samples during either P or S phases from women with RPL (n = 28) or healthy control (n = 26). Comprehensive proteomic and phosphoproteomic analyses were conducted using label-free liquid chromatography-tandem mass spectrometry (n = 44) and verified through western blotting (n = 10). Three comparison groups were established, including the total RPL endometrium compared to the total control (R/C), proliferating endometrium compared to control (RP/CP), and secretory endometrium compared to control (RS/CS). Results: Differentially expressed proteins (DEPs) and differentially phosphorylated proteins (DPPs) were respectively identified in the three comparison groups. Combining pathway enrichment, network analysis, and soft clustering analysis, we identified the insulin/cyclic nucleotide signaling pathway and AMPK/mTOR signaling pathway as the major contributors to the aberration of RPL endometrium. Western blotting verified altered expression of four proteins, including PRKAR1B, ADCY3, PRKAA2, and LPIN2.Conclusions: This exploratory study provides insights into the differentiated protein expression and phosphorylation profiles of RPL endometrium in both P and S phases. The results highlight potential proteins associated with RPL pathogenesis that may serve as potential indicators for RPL. The findings contribute to the identification of potential targets for RPL treatment as well as the pathogenesis of RPL.

Novel stool-based protein biomarkers for improved colorectal cancer screening

Project description:Fecal immunochemical tests (FIT) detecting hemoglobin in stool are widely used for non-invasive colorectal cancer (CRC) screening, but their sensitivity leaves room for improvement. Our aim is to identify novel protein biomarkers in stool that outperform or complement hemoglobin in detecting CRCs and advanced adenomas (AAs). Stool samples (one series of 12 CRCs and 10 controls, and a second series of 81 CRCs, 40 AAs, 43 non-advanced adenomas and 129 controls) were analyzed by mass-spectrometry and searched for human proteins. Classification and regression tree and logistic regression analyses were performed to identify protein combinations that differentiated CRCs and/or AAs from controls. Antibody-based assays for four selected proteins were performed on an independent series of FIT samples (14 CRCs, 16 AAs, 18 non-advanced adenomas and 24 controls) Results: In total, 834 human proteins were identified, of which 29 were significantly enriched in CRCs versus controls in both stool sample series. Combinations of four proteins reached sensitivities of 80% and 45% for detecting CRCs and AAs, at 95% specificity, which was higher than hemoglobin alone.

Beetle horns evolved from wing serial homologs

Project description:Understanding how novel complex traits originate is a foundational challenge in evolutionary biology. Yet how descent with modification in developmental evolution may lead to morphological innovation remains poorly understood. We investigated the origin of thoracic horns in scarabaeine beetles, one of the most dramatic classes of secondary sexual traits in the animal kingdom. We show that thoracic horns derive from bilateral source tissues, that diverse wing genes are functionally required for instructing this process, and that in the absence of Hox-input thoracic horn primordia transform to contribute to ectopic wings. Once induced, however, the transcriptional profile of thoracic horns diverges markedly from that of wings and other wing serial homologs. Our results provide evidence for the serial homology between thoracic horns and insects wings, and suggest that other insect innovations may similarly derive from wing serial homologs and the concomitant recruitment of diverse genes from outside a wing formation context.
