Project description:Many lung diseases remain understudied due to a lack of experimental models. Lung organoids, which consist of self-organizing epithelial cells, provide versatile in vitro models for normal and abnormal biology, drug screening, gene editing, and personalized therapeutics. However, human organoids are generally derived from lung tissue, which is not commonly obtained and represents only a small fraction of lung pathologies. Induced pluripotent stem cells have provided an important alternative but require complex manipulation. Recently, one study reported airway organoids from bronchoalveolar lavage (BAL) fluid, though sample sizes and characterization were limited. Here, we demonstrate robust establishment of airway organoids from a variety of human BAL samples and show that these organoids consist predominantly of basal cells plus differentiated airway cell types including secretory, ciliated, KRT13+ “hillock,” and ionocyte cells. Furthermore, we report the development of BAL-derived alveolar organoids comprised of alveolar type 2 (AT2) cells. These techniques significantly expand the scope of lung diseases that can be studied using safely accessible primary human cells.

Project description:This observational study catalogues the overlap in metabolites between matched bronchoalveolar lavage fluid (BALF) and plasma, identifies the degree of congruence between these metabolomes in human and mouse, and determines how molecules may change in response to cigarette smoke (CS) exposure. Matched BALF and plasma was collected from mice (ambient air or CS-exposed) and humans (current or former smokers), and analyzed using mass spectrometry. There were 1155 compounds in common in all 4 sample types; fatty acyls and glycerophospholipids strongly overlapped between groups. In humans and mice, more than half of the metabolites present in BALF were also present in plasma. Mouse BALF and human BALF had a strong positive correlation with 2040 metabolites in common, suggesting that mouse models can be used to interrogate human lung metabolome changes. While power was affected by small sample size in the mouse study, the BALF metabolome appeared to be more affected by CS than plasma. CS-exposed mice showed increased plasma and BALF glycerolipids and glycerophospholipids. This is the first report cataloguing the metabolites present across mouse and human, BALF and plasma. Findings are relevant to translational studies where mouse models are used to examine human disease, and where plasma may be interrogated in lieu of BALF or lung tissue.

Project description:<p>The purpose of this longitudinal study protocol is to define age-related prevalence of phenotypic characteristics and the progression of key features of lung disease in participants with one of these disorders, Primary Ciliary Dyskinesia (PCD). In PCD, the abnormal structure and function of cilia results in impaired clearance of secretions and consequent obstruction and chronic recurrent infection in the airways, sinuses and middle ears. Although it seems likely that the onset of PCD airway disease occurs early in childhood, as has been reported for cystic fibrosis (CF), the clinical course of PCD lung disease is not well defined, nor, is the time course of emergence of specific microbial pathogens, or the age of onset and rate of progression of airway disease and bronchiectasis. This longitudinal study is designed to define the rate of progression of PCD lung function in participants between 5-18 years of age using spirometry which tracks well with lung impairment and prognosis in other disorders of the airways such as cystic fibrosis. This longitudinal protocol will also systematically track other specific outcomes, including pathogens infecting the airways (assessed by respiratory cultures), and age at onset and progression of airway damage and bronchiectasis (assessed by high-resolution computerized tomography, HRCT, of the chest).</p> <p>Blood will be collected for DNA to test for genetic mutations. The blood samples will be processed to establish a cell line (lymphocyte transformation) for a source of DNA for future genetic studies, in participants with disease. For participants unable to provide a blood sample, a buccal sample will be obtained for DNA.</p>

Nineteen subjects, with and without asthma, were exposed to clean air (0 ppb), low (100ppb), and high (200ppb) ambient levels of ozone for 4 hours on three separate occasions in a climate-controlled chamber followed by bronchoscopy with bronchoalveolar lavage (BAL) 20 hours after the end of exposure in a repeated measure design. The BAL cells mRNA expression was examined using Affymetrix GeneChip Microarray.

Project description:Upon returning from deployment to Afghanistan, increasing numbers of United States service personnel are reporting symptoms consistent with an asthma-like airways disease, thought to be caused by prolonged inhalation of toxic desert particulate matter. Deployment Related Lung Disease (DRLD) symptoms persist in the post-deployment setting, where soldiers are then exposed to common household allergens. The goal of this study was to define the transcriptomic responses of leukocytes found in the airway lumen and airspace in a post-deployment setting, mimicked by inhalation of Afghanistan desert particulate matter (APM) and subsequent challenge with house dust mite (HDM) in mice. C57BL/6 mice (n=15/group) were exposed to filtered air or aerosolized APM for 6 hours per day for 12 consecutive days using a whole body exposure system, followed by intranasal PBS or HDM allergen challenges. One day after the last HDM challenge, mice were euthanized to collect bronchoalveolar lavage (BAL) cells for single cell RNA sequencing (scRNAseq) using the 10x Genomics Platform and assessment of inflammation and airway hyperresponsiveness (AHR). Unsupervised clustering of BAL cell scRNAseq data revealed a unique monocyte population induced only by both APM and allergen treatments. This population of monocytes is characterized by expression of genes involved in allergic asthma, including ALOX15, which is involved in airway obstruction and promoting the inflammatory response. We validated ALOX15 expression in monocytes via immunohistochemistry and immunofluorescence staining of lung tissue. APM pre-exposure, followed by HDM challenge lead to significantly increased total respiratory system resistance compared to filtered air controls. Using this post-deployment mimic in mice, we demonstrated that inhalation of desert PM while on deployment to Afghanistan may prime airways to be more responsive to allergen exposure after returning home. DRLD symptoms in post deployment setting may involve induction of a unique lung monocyte population that has the ability to regulate AHR and inflammation, possibly through increased expression of ALOX15.

Project description:Chronic beryllium disease (CBD) is a metal-induced hypersensitivity disorder characterized by CD4+ T cell alveolitis. Single-cell gene expression profiling of bronchoalveolar lavage (BAL) cells focused on T cells may provide insight into unique subsets of CD4+ T cells, functional pathways, and/or molecules that could potentially be developed into novel therapeutics for treating CBD and other granulomatous lung diseases. To understand the unbiased cellular composition of airway associated CD4+ T cells in subjects with beryllium exposure, we used BAL cells from one beryllium-exposed (BeS) and one CBD subject diagnosed using previously defined criteria, and these two samples comprised the disease cohort.
