ABSTRACT: Tmem127 is a tumor suppressor gene that has been linked to inherited susceptibility to pheochromocytomas. We used single nuclei RNA sequencing (snRNA-seq) to explore the cellular role of Tmem127.

Project description:The human adrenal glands are highly dynamic endocrine organs that are involved in the secretion of various hormones such as steroids and catecholamines. Here we present a single-nuclei and spatial transcriptomic analysis of healthy adult human adrenal glands to provide a complete adrenal gland atlas. With this, we show how such an atlas can be taken advantage when studying adrenocortical diseases, such as adrenocortical adenomas (ACA). Using nornal adrenal as reference, we showed a high intra-tumoural heterogeneity in the single-nuclei transcriptome of ACA, revealing the presence of specific cell populations associated with cortisol secretion and genetic background.

Project description:MicroRNAs are involved in the pathogenesis of several tumors, however, there have been no data on microRNA expression in pheochromocytomas to date. In this study we have performed microRNA expression profiling in sporadic benign, hereditary benign (multiplex endocrine neoplasia type 2-related and von Hippel-Lindau syndrome-related pheochromocytomas), and sporadic recurring adrenomedullary tumors. Due to the rarity of these neoplasms, the retrieval of sufficient quantities of fresh or immediately snap frozen tumor samples is difficult, therefore the applicability of formalin-fixed paraffin-embedded tissue samples for the analysis of microRNA expression in pheochromocytomas was examined.

Extensive patient-to-patient single nuclei transcriptome heterogeneity in pheochromocytomas and paragangliomas

Project description:Pheochromocytomas (PC) and paragangliomas (PG) are rare neuroendocrine tumors of varied genetic makeup, associated with high cardiovascular morbidity and a variable risk of malignancy. We focused on PCPG tumors with germline SDHB and RET mutations, representing distinct prognostic groups with worse or better prognoses, respectively. We applied single-nuclei RNA sequencing (snRNA-seq) on tissue samples from 11 patients.
