Gene expression changes in mouse hippocampus after overnight exposure to corticosterone

ABSTRACT: The aim of the experiment was to test the effect of elevated level of corticosterone on hippocampal transcriptome after 12 hours of treatment during active phase to mimic the effect of a day-long stress. Additionally, we also tested the decay time of transcriptomic response during resting period when the level of glucocorticoids returns to the baseline. Our study shows that transcriptomic responses to glucocorticoids are heterogenous in terms of the decay time with some genes displaying persistent changes in expression during 9 hours of rest.

To clarify the mechanism, we examined the effects of glucocorticoids on cultured neuronal progenitor cells (NPCs) from cerebral cortices from rat embryo. We performed comparative analysis using gene expression profiling of corticosterone- or vehicle-treated NPCs. Results provide insight into the effects of glucocorticoids in regulating genetic programs important for controlling NPCsâ properties. Experiment Overall Design: Three couples of RNA samples from corticosterone- or vehicle-treated neuronal progenitor cells were analyzed.

However, patients often develop adverse metabolic effects, including hyperphagia, obesity, and hyperglycemia.The arcuate nucleus has an established role in the central control of energy balance. Ventral hippocampal transcriptome analysis after acute corticosterone exposure.

Corticosterone induces obesity via promoting intestinal cell proliferation and survival

Project description:A vicious cycle ensues whereby prolonged exposure to social stress causes increased production of glucocorticoids, leading to obesity even further. Understanding the role of glucocorticoids, the key element in the vicious circle, might be helpful to break the vicious circle. However, the mechanism by which glucocorticoids induce obesity remains elusive. Herein we find that administration of corticosterone (CORT) to mice promotes the proliferation and survival of intestinal cells, which might contribute to the longer small intestines and the elongated intestinal villi, thus leading to increased nutrient absorption and obesity in mice. RNA-seq was performed to analyze transcriptional alterations in small intestines and livers after CORT-treatment, indicating differentially expressed genes (DEGs) were enriched in several cell growth/death-associated signaling pathways. This study would provide novel therapeutic clues for stress or glucocorticoids-related obesity.

Corticosterone pattern-dependent glucocorticoid receptor binding and transcriptional regualtion

Project description:Ultradian glucocorticoid rhythms are highly conserved across mammalian species, however, their functional significance is not yet fully understood. Here we demonstrate that pulsatile corticosterone replacement in adrenalectomised rats induces a dynamic pattern of glucocorticoid receptor (GR) binding at ~3,000 genomic sites in liver at the pulse peak, subsequently not found during the pulse nadir. In contrast, constant corticosterone replacement induced prolonged binding at the majority of these sites. Additionally, each pattern further induced markedly different transcriptional responses. During pulsatile treatment, intragenic occupancy by active RNA polymerase II exhibited pulsatile dynamics with transient changes in enrichment, either decreased or increased depending on the gene, which mostly returned to baseline during the inter-pulse interval. In contrast, constant corticosterone exposure induced prolonged effects on RNA polymerase II occupancy at the majority of gene targets, thus acting as a sustained regulatory signal for both transactivation and repression of glucocorticoid target genes. The nett effect of these differences were consequently seen in the liver transcriptome as RNA-seq analysis indicated that despite the same overall amount of corticosterone infused, twice the number of transcripts were regulated by constant corticosterone infusion, when compared to pulsatile. Target genes that were found to be differentially regulated in a pattern-dependent manner were enriched in functional pathways including carbohydrate, cholesterol, glucose and fat metabolism as well as inflammation, suggesting a functional role for dysregulated glucocorticoid rhythms in the development of metabolic dysfunction.

Mice overexpressing MR in cardiomyocytes (MR-Cardio) and their controls (Ctrl) were treated for 7 days with aldosterone or corticosterone and a whole genome microarray analysis was performed.
