Role of exogenous abscisic acid in freezing tolerance of mangrove Kandelia obovata under natural frost condition at near 32oN

ABSTRACT: Mangrove Kandelia obovata, an important coastal shelterbelt and landscape tree, is distributed in tropical and subtropical shores and likely delimited in the latitudinal range by varying sensitivity to cold. Here, we explored the temporal variations in physiological status and transcriptome profiling of K. obovata under natural frost conditions at ~32oN, as well as the positive role of exogenous abscisic acid (ABA) in cold resistance.

Unravelling Chilling-stress Resistance Mechanisms in Endangered Mangrove Plant Lumnitzera littorea (Jack) Voigt

Project description:Mangrove plants are the demonstrated woody plants in tropical and subtropical coastal area with great ecological and economic importance. However, due to the extreme global climate change and increased human activities, mangroves is suffering a dramatic declining worldwide. Lumnitzera littorea (Jack) Voigt is one of the most endangered mangrove species in China. A comprehensive understanding on its resistance mechanisms to low ambient temperature help us to better conserve and restore L. littorea. In the current study, we performed comparative transcriptome analysis to investigate the genome-wide changes of gene expression profile in L. littorea under chilling stress (8°C day/5°C night) over normal condition (25°C day/23°C night). The low temperature suppressed fatty acid elongation and protein phosphorylation in L. littorea, while induced calcium ion binding process and signaling transduction, suggesting an activation of cold-stress sensing and signaling in L. littorea. Combining our analysis with our previous physiological assays, we showed a substantial photoinhibition occurring in the seedlings of L. littorea with the decrease of ambient temperature, and the synthesis of photosystem II reaction center protein D1 and peroxidase-involved scavenging of reactive oxygen species (ROS) were enhanced accordingly to combat the adverse impacts. Finally, we highlighted the biological significance of post-transcriptional regulation, including RNA binding and surveillance, in coping with cold stress in L. littorea. Collectively, our findings provide a global view to the resistance mechanisms of chilling stress in L. littorea, and valuable genetic resources to assist the protection and restoration of L. littorea.

Comparative transcriptome analysis unveiling molecular mechanisms underlying low temperature resistance in the white mangrove Laguncularia racemosa

Project description:Temperature is an important factor that affects the growth and reproduction of mangrove plants. Laguncularia racemosa (L.) C.F. Gaertn is a controversial species in China, in terms of being a pioneer species for mangrove restoration and a putative invasive species occupying the natural habitats. One of its advantages is the strong resistance to low temperature, which makes L. racemose more adaptive to extreme climate change than local mangrove species. However, little is known about the regulatory mechanisms underlying the cold-stress tolerance in L. racemose, which restricted our understanding on its biological features and invasion potential. In this study, In the current study, we investigated the physiological and molecular mechanisms involved in chilling-stress adaptation in L. racemose. Freezing temperature caused damage to cell membrane system and reduced photosynthesis efficiency in L. racemose. To combat the adverse impacts, plasma membrane biosynthesis and antioxidant processes were substantially enhanced. Furthermore, we showed that there was a difference between the responses to freeze-thaw injury in L. racemose from different locations. It may provide new clues to the different genetic background between varieties of L. racemose. These novel findings could provide biochemical and genetic basis for the cultivation and restoration of L. racemose.

Differential gene expression during cold-acclimation in barley, wheat and rye

Project description:The present transcript profiling compares the gene expression during cold-acclimation in different genotypes of barley (Hordeum vulgare L.), wheat (Triticum aestivum L.) and rye (Secale cereale L.) in order to determine factors influencing frost tolerance. Because of its outstanding robustness against adverse environmental conditions rye is considered to be a model species for abiotic stress tolerance. Wheat is moderate frost-tolerant and barley is most sensitive species in this study. The aim of this study elucidate conserved, as well as, species-specific gene regulation across the Triticeae. Furthermore, transcript abundances were correlated between the distinct frost tolerances of genotypes within each species in order to find candidate genes for frost tolerance.

Insight into cellular proteome of Lolium multiflorum/Festuca arundinacea introgression forms to decipher crucial mechanisms of cold acclimation in forage grasses

Project description:Frost tolerance is the main component of winter-hardiness. To express this trait, plants have to sense low temperature, and respond by activating the process of cold acclimation. The molecular mechanisms of this acclimation have not been fully understood in the agronomically important group of forage grasses, including Lolium-Festuca species. Herein, the introgression forms of L. multiflorum/F. arundinacea distinct with respect to their frost tolerance, were used as models for the comprehensive, proteomic and physiological, research to recognize the crucial components of cold acclimation in forage grasses. The obtained results stressed the importance of photosynthetic performance under acclimation to low temperature. The stable level of photochemical processes after three weeks of cold acclimation in the introgression form with a higher level of frost tolerance, combined simultaneously with the stable level of CO2 assimilation after that period, despite decreased stomatal conductance, indicated the capacity for that form to acclimate its photosynthetic apparatus to low temperature. This phenomenon was driven by the Calvin cycle efficiency, associated with revealed here accumulation profiles and activities of chloroplastic aldolase. The capacity to acclimate the photosynthetic machinery to cold could be one of the most crucial components of forage grass metabolism to improve frost tolerance.

Impact of heavy metals on cold acclimation of Salix viminalis roots

Project description:Frost is a major abiotic stress limiting plant growth and development. Climate change models predict an increase in the magnitude and frequency of late-frost events, which, together with an observed loss of soil insulation, will significantly damage roots. To withstand freezing stress, plants have evolved an adaptative process known as cold acclimation. While this process is well documented, it is known that the plant response to multiple stresses is unique and cannot be deduced from the response to each stress taken separately. Here, we investigate the impact of long-term metal exposure on the cold acclimation of Salix viminalis roots. To do so, we used physiological, transcriptomic and proteomic approaches. We found that while metal exposure significantly affected plants morphology and physiology, it did not impede cold acclimation. The impact of the simultaneous exposure to metals and cold acclimation on the transcriptome was unique, while at the proteomic level, the cold acclimation component seemed to be dominant. Further analysis revealed that metals strongly and negatively impacted the cellular antioxidant system. While this should have led to a loss of frost tolerance, it was not observed. A group of proteins was identified that could have played a role in compensating the impediment of the antioxidative system in metal-exposed roots.
