RNA seq DATA （miR-361-3p mimics transfected Huh7 cells VS. mimics-NC transfected Huh7 cells ）

ABSTRACT: Studies have shown that miR-361-3p is associated the progression of HCC cells and has potential to be used as a biomarker for cancer therapy. However, the mechanism of how miR-361-3p regulates the progression of HCC cells is not clear. To explore this mechanism, we transfected miR-361-3p mimics into Huh 7 cells , and verified the transfection efficiency with qRT-PCR. Then, we performed transcriptomic analysis using data obtained from RNA-seq of 3 pairs of these transfected cells.

Transcriptomic analysis of hepatocellular carcinoma cell lines HepG2 treated with Sorafenib and transfected with miR-200c-3p inhibitor, and miR-222-5p and miR-512-3p mimics

Project description:Hepatocellular carcinoma (HCC) is one of the most common causes of death worldwide and the fourth most prevalent type of cancer. Whereas curative treatments such as liver transplantation, ablation or surgery are optimal for early stages, only paliative treatments are given to intermediate and advanced stages of the disease. Despite the introduction of immune regulators as first-line treatments for advanced stages, Sorafenib is still the standard of care in the clinical practice. In cell lysates, anti-tumoral properties of Sorafenib were related to upregulation of miR-200c-3p (anti-tumoral miRNA) at 6 hours of treatment and downregulation of miR-222-5p and miR-512-3p (pro-tumoral miRNAs) at 24 hours. We have identified these miRNA biomarkers of Sorafenib treatment response in plasma of patients with advanced HCC treated with Sorafenib. In particular, miR-200c-3p has been related to increased survival benefit whereas miR-222-5p and miR-512-3p have been related to worse prognosis. Our study has sequenced HepG2 cells treated with Sorafenib and miR-200c-3p inhibitor, and transfected with miR-222-5p and miR-512-3p mimics to unravel the molecular pathways governing Sorafenib response

Project description:To clarify the gene expression profile in MLE-12 cells transfected with microRNA mimics upon influenza virus infection, we transfected microRNA mimics (mmu-miR-483-3p or Negative control miRNA) into MLE-12 cells and infected A/Puerto Rico/8/1934 (PR8) strain at an MOI of 2 at 24 hours post transfection. RNA was isolated from cells at 12 hours post infection. We found that miR-483-3p transfection down-regulated the genes involved in the innate immunity regulation upon influenza virus infection.

Gene expression data from HEK-293T cells transfected with miR-194 mimics

Project description:Nuclear factor κB (NF-κB) pathway plays an important role in hepatocellular carcinoma (HCC) progression. miR-194 was previously shown to reduce the induction of NF-κB activity upon addition of tumor necrosis factor α (TNFα). To clarify the molecular mechanism responsible for the effect of miR-194 on NF-κB pathway, mRNA microarray assays were performed to identify the genes that were suppressed by miR-194. HEK-293T cells transfected with miR-194 mimics were cultured for RNA extraction and hybridization on Affymetrix mRNA microarrays. These were compared against the control, which were HEK-293T cells transfected with negative control mimics.
