Apobec1 and acute kidney injury

ABSTRACT: Kidney tissues from a cisplatin-induced acute kidney injury mouse model of global Apobec1 knockout mice compared with wild type mice. 6 months old male mice examined. We used microarrays to identify genes that are differentially expressed during acute kidney injury in the absence of apobec1 compared with wild type mice.

Proteomic analysis of cisplatin exposure on the kidney cortex proteome and the effect of preconditioning

Project description:Acute kidney injury and nephrotoxicity are important clinical side effects of cisplatin. Thus, the mechanisms of this disease, and potential treatment options are important to understand in their entity. Here, we analyzed the proteome of cisplatin induced acute kidney injury in a mouse model. Functionally we found that calorie restriction was able to completely blunt Cisplatin induced AKI, and hypoxia ameliorated cCisplatin induced AKI. To investigate the mechanism for this in high throughput, we performed label-free single-shot proteomic analyses of these kidneys.Acute kidney injury and nephrotoxicity are important clinical side effects of cisplatin. Kidney toxicity in miR-155-/- mice

Project description:The development of nephrotoxicity limits the maximum achievable dosage and treatment intervals for cisplatin chemotherapy. Therefore, identifying mechanisms that regulate this toxicity could offer novel methods to optimize cisplatin delivery. MicroRNAs are capable of regulating many different genes, and can influence diverse cellular processes, including cell death and apoptosis. We previously observed miR-155 to be highly increased following ischemic or toxic injury to the kidneys and, therefore, sought to determine whether mice deficient in miR-155 would respond differently to kidney injury. miR-155-/- mice (B6.Cg-Mir155tm1.1Rsky/J originally purchased from The Jackson Laboratory) and age- and weight-matched C57BL/6 (wild type) mice (Charles River Laboratories) were used in this study. Mice were injected with one single 20mg/kg dose of cisplatin. Kidney tissues were collected at 0, 24, 48, and 72 hours i.p. with saline-injected mice as control. Total RNA extracted from the kidney tissues were used for gene expression analysis with Agilent Mouse whole genome 4x44k v2 array.

RNA-seq analysis of proximal tubule specific Fan1 knockout transgenic kidneys subjected to cisplatin injury

Project description:FAN1 is a DNA endonuclease that we have previously identified as the underlying genetic cause of karyomegalic interstitial nephritis (OMIM: 614817) in humans. In order to deduce the molecular function of FAN1 in the setting of acute kidney injury and progression to chronic kidney disease, we generated a proximal tubule specific Fan1 knockout mouse model and subjected these mice to cisplatin injury. RNA-seq of the transgenic mice kidneys revealed dysregulation of the DNA damage repair pathway as well as cell cycle associated genes. Together, the data supports for a role of Fan1 in DNA damage repair, and that in its absence results in dysfunctional proximal tubule repair and regeneration upon cisplatin injury.
