Project description:We used microarrays to detail the global programme of gene expression underlying the effect of sleep deprivation in the mouse hippocampus and identified distinct classes of regulated genes during this process. Hippocampal tissue was taken from sleep deprived mice and time-matched non-sleep-deprived control animals that were left undisturbed in their home cages during the sleep deprivation period for a total of 8 and 9 replicates per group. RNA was isolated and cDNA was synthesized from hippocampal tissue, and the sample from each animal was hybridized to a separate Affymetrix Mouse 430_2 microarray

Project description:We used microarrays to detail the global programme of gene expression after 4 months of TFEB overexpression in the brain. Groups of Tg4510 and Wild-type mice were injected at P0 in the brain. Those groups of mice and the respectively controls were sacrificed and analyzed after 4 months from the injection. RNA extraction from the hippocampus and hybridization on Affymetrix microarrays was done.

Project description:Activity-dependent gene expression is central for sculpting neuronal connectivity in the brain. Despite the importance for synaptic plasticity, a comprehensive analysis of the temporal changes in the transcriptomic response to neuronal activity is lacking. In a genome wide survey we identified genes that were induced at 1, 4, 8, or 24 hours following neuronal activity in the hippocampus. 3 month old male mice were injected with kainic acid or isotonic saline solution. Animals were sacrificed by cervical luxation 1, 2, 4, 8, or 24 h after onset of the first seizure, and RNA was extracted from the hippocampal region.

Project description:Purpose: The goals of this study are to analyze the mRNAs expression in hippocampal CA3 after Adora1 knockout by High-throughput sequencing. Methods: The brain was dissected and followed by 20 μm cryosection onto 20 glass slides. The hippocampus CA3 was isolated from brain slides by using laser capture microdissection. Then the samples were stored on dry ice until RNA extraction by Trizol. The RNAseq was conducted by using Illumina HiSeq2000 platform. Results: By performing RNA sequencing on RNAs isolated from the hippocampus CA3 neurons of AKO and C57 mice at 3-month-old. We found that 51 mRNAs were upregulated, whereas 77 mRNAs were downregulated in AKO mice. Conclusion:We deciphered the underlying neuroprotective mechanisms of A1R knockdown in AD mouse models. By employing the RNA-seq technique, we compared the alterations in gene expression in the hippocampus between AKO and wild-type mice.

Project description:Alzheimer’s disease (AD) is an unremitting neurodegenerative disorder characterized by cerebral amyloid-β (Aβ) accumulation and gradual decline in cognitive function. Changes in brain energy metabolism arise in the preclinical phase of AD, suggesting an important metabolic component of early AD pathology. Neurons and astrocytes function in close metabolic collaboration, which is essential for the recycling of neurotransmitters in the synapse. However, how this metabolic interplay may be affected during the early stages of AD development has not been sufficiently investigated. Here, we provide an integrative analysis of cellular metabolism during the early stages of Aβ accumulation in the cerebral cortex and hippocampus of the 5xFAD mouse model of AD. Our electrophysiological examination revealed an increase in spontaneous excitatory signaling in hippocampal brain slices of 5xFAD mice. This hyperactive neuronal phenotype coincided with decreased hippocampal TCA cycle metabolism mapped by stable 13C isotope tracing. Particularly, reduced astrocyte TCA cycle activity led to decreased glutamine synthesis, in turn hampering neuronal GABA synthesis in the 5xFAD hippocampus. In contrast, cerebral cortical slices of 5xFAD mice displayed an elevated capacity for oxidative glucose metabolism suggesting a metabolic compensation. When we explored the brain proteome and metabolome of the 5xFAD mice, we found limited changes, supporting that the functional metabolic disturbances between neurons and astrocytes are early events in AD pathology. In addition, we show that synaptic mitochondrial and glycolytic function was impaired selectively in the 5xFAD hippocampus, whereas non-synaptic mitochondrial function was maintained. These findings were supported by ultrastructural analyses demonstrating disruptions in mitochondrial morphology, particularly in the 5xFAD hippocampus. Collectively, our study reveals complex region and cell specific metabolic adaptations, in the early stages of amyloid pathology, which may be fundamental for the progressing synaptic dysfunction in AD.

Project description:Fos induction is the most commonly used marker of neuronal activation. Here, we investigate the binding profile of Fos in the adult mouse hippocampus using antibodies specific against this transcription factor and the chromatin Immunoprecipitation coupled to DNA-sequencing (ChIP-seq) method. We generated profiles in basal conditions and 1 hour after neuronal stimulation. To trigger the syncronous activation of hippocampal exciatory neurons we treated the mice with kainic acid (KA) through peritoneal administration. This model of status epilepticus has been characterized in detail at the epigenomic level (Fernandez-Albert et al., 2019).

Project description:Elucidating the developmental patterns of gene expression across brain regions is a critical step toward understanding brain development and disease. We previously studied the transcriptome of multiple brain regions and peripheral tissues across development in the vervet monkey, and identified transcripts related to hippocampal size in 59 animals across 10 developmental timepoints, ranging between 7 days to 9 years of age. Here, we extended this analysis with a focus on transcriptomic changes in hippocampus across early development in vervet neonates and infants under 1 year of age. We combined this dataset with the previously analyzed data. We identified groups of transcripts with age-related patterns, which correlated well with existing human resources. Additionally, expression quantitative trait locus (eQTL) analysis identified two further genes, RAB31 and CHMP1B, associated with hippocampal volume, refining the previously identified locus. This dataset provides unique developmental expression data in the hippocampus of a non-human primate, and expands our catalog of local and distant eQTL in this brain region.

Project description:The hippocampus is important for memory formation and is severely affected in the brain with Alzheimer's disease (AD). Since AD brain tissue is available postmortem, our understanding of early pathogenic processes occurring in hippocampi remains speculative. Here, an MS-based proteomic approach was used to analyze free-floating hippocampal spheroids (HSs) enriched in PROX1-positive granule neurons, from induced pluripotent stem cells (iPSCs) of healthy individuals and AD patients. HSs generated from two AD patients carrying variations in amyloid precursor protein (APP) or presenilin 1 (PS1) genes, and their age and gender matched controls.
