Syngeneic model of carcinogen-induced tumor mimics basal/squamous, stromal-rich, and neuroendocrine molecular and immunological features of muscle-invasive bladder cancer

ABSTRACT: Invasive bladder tumors were induced in C57BL/6 mice when continuously exposed to N-butyl-N-(4-hydroxbutyl nitrosamine) (BBN) in the drinking water. Tumors were excised and serially passed by subcutaneous implantation into sex-matched syngeneic C57BL/6 hosts. Eight lines were named BBN-induced Urothelium Roswell Park (BURP) tumor lines. BURP lines were characterized by applying consensus molecular classification to RNA expression.

Project description:We developed a UPPL (Upk3a-CreERT2;p53f/f;Ptenf/f;Rosa26LSL-Luc) mouse model of bladder cancer and compared it with the existing BBN (N-butyl-N-(4-hydroxybutyl)nitrosamine) mouse model of blader cancer. We cultured UPPL and BBN primary tumor cells as cell lines along with MB49 cancer cell lines and KT immortalized normal urothelial cell lines and implanted them back into mice as cell-line derived tumors.

Expression data from the bladders of carcinogen-treated mice with genetic Hh pathway manipulation

Project description:Attenuation of Hedgehog (Hh) pathway activity leads to accelerated tumor progression in a mouse model of N-butyl-N-4-hydroxybutyl nitrosamine (BBN) – induced bladder carcinoma. In order to identify genes regulated by the Hh pathway that might be involved in bladder cancer progression, we performed transcriptional profiling of bladders harvested from mice after BBN-exposure, comparing Gli1CreER/WT; Smoflox/WT mice to Gli1CreER/WT; Smoflox/flox mice, which express CreER under control of the Gli1 promoter and carry one or two floxed alleles of the essential Hh pathway transductory component Smoothened (Smo) respectively. Administration of Tamoxifen to these mice results in attenuation of Hh pathway activty to a greater extent in the Gli1CreER/WT;Smoflox/flox mice as compared to Gli1CreER/WT;Smoflox/WT mice, allowing identification of Hh-pathway regulated genes. 6 total samples were analyzed. 3 bladders from Gli1CreER/WT; Smoflox/WT mice and 3 bladders from Gli1CreER/WT; Smoflox/flox mice were analyzed.

Elucidating the role of glycogen debranching enzyme AGL in bladder carcinogenesis by generation and characterization of knockout mice

Project description:Amylo-alpha-1-6-glucosidase-4-alpha-glucanotransferase (AGL) is an enzyme primarily responsible for glycogen debranching. Inactivating germline mutations lead to glycogen storage disease III (GSDIII). AGL was found to be a tumor suppressor in xenograft models of bladder cancer (BC) and low levels of expression in BC were associated with poor patient prognosis. However, the impact of AGL levels in normal tissues on susceptibility to carcinogenesis is unknown. We address this gap by developing a full body/germline AGL knock-out (AGL-FBKO) that recapitulates biochemical and histological features of GSDIII. AGL-FBKO mice exposed to N-Butyl-N-(4-hydroxybutyl) nitrosamine (BBN) had higher BC incidence at several time points following exposure, compared to wild type littermates (AGL-FBWT). To determine if this increased BC prevalence was due to decreased expression of AGL in the urothelium, we derived a urothelium specific conditional AGL knock-out (AGL-cKO) mouse driven by a Uroplakin II-Cre/loxP system. BBN experiments with these mice revealed AGL-cKO had higher BC incidence than mice without AGL knock-out (AGL-cWT). RNA-sequencing revealed that tumors from AGL-FBKO had 19 differentially expressed genes compared to AGL-FBWT mice. An “AGL Loss” gene signature was developed and found to successfully stratify normal and tumor samples in two BC patient datasets. These results support the role of AGL as a tumor suppressor. Our work suggests that evaluating AGL expression levels, or AGL KO gene signature scores, in normal urothelium may serve as a predictor for increased risk of BC development in individuals with carcinogenic exposures, such as smoking.

Gene expression study for effectiveness of nicotinamide using chemically-induced mouse bladder cancer model

Project description:The objective of this study was to evaluate the efficacy of nicotinamide in a N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN)-induced urinary bladder cancer model in mice, and to identify through gene expression profiling the molecular signatures of cancer prevention by nicotinamide. We used 20 mice for microarray experiments: five mice with normal bladders (group I), five with nicotinamide-treated bladders (group II), five with BBN-induced mouse bladder tumors (group III), and five with non-tumorigenic bladders treated with BBN and nicotinamide (group IV). Keywords: Gene expression, Mouse bladder cancer, Cancer prevention Total RNA was isolated using TRIzol reagent (Life Technologies, NY), according to the manufacturer's protocol. The quality and integrity of the RNA were confirmed by agarose gel electrophoresis and ethidium bromide staining, followed by visual examination under ultraviolet light. Five-hundred nanograms of total RNA were used for labeling hybridization according to the manufacturerM-bM-^@M-^Ys protocols (Illumina Mouse-6 BeadChips, version 1.0). Arrays were scanned with an Illumina Bead Array Reader confocal scanner (BeadStation 500GXDW; Illumina, Inc., San Diego, CA) according to the manufacturer's instructions. After scanning, the microarray data were normalized using quantile normalization. Measured gene expression values were log2 transformed and median-centered across genes and samples.

Single-Cell Profiling of Murine Bladder Cancer Identifies Sex Specific Transcriptional Signatures with Prognostic Relevance

Project description:Bladder cancer (BLCA) is more common in men but more aggressive in women. A common model used to study sex-based differences in cancer biology is murine BLCA generated by N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN). While tumors in this model have been profiled, these profiles provided limited information on the tumor microenvironment. Hence, we applied single-cell RNA sequencing to characterize cell type specific transcriptional differences between male and female BBN induced tumors and then studied their human relevance. We found proportion and gene expression differences in the epithelial and non-epithelial subpopulations between male and female tumors. Of translational importance, expression of several genes was found to predict sex specific survival in several human BLCA datasets. We also noted a male biased CD8 T cell exhaustion program in this model that correlates highly with androgen receptor activity. Our study identified novel and clinically relevant sex specific transcriptional signatures and validates the relevance of the BBN model for studying sex differences in human BLCA. This work highlights the importance of considering sex as a biological variable in cancer marker development.

The influence of age and sex on the pre-treatment immune microenvironment of a carcinogen induced murine model of bladder cancer

Project description:The incidence of urothelial carcinoma of the bladder is four times higher in males than females; however, females tend to present with a more aggressive disease, a poorer response to immunotherapy and suffer worse clinical outcomes. Recent findings have demonstrated sexual dimorphism in the tumor immune microenvironment of non-muscle invasive bladder cancer and associated clinical outcomes. However, a significant gap in knowledge remains with respect to the current pre-clinical modeling approaches and more precisely recapitulating these differences towards improved therapeutic design. Based on the similarities in mucosal immune physiology between humans and mice, we evaluated the sex and age-related immune alterations in healthy murine bladders. Bulk-RNA sequencing and multiplex immunofluorescence based spatial immune profiling of healthy murine bladders from male and female mice of age groups spanning young to old showed a highly altered immune landscape that exhibited sex and age associated differences, particularly in the context of B cell associated responses. Spatial profiling using markers specific to macrophages, T lymphocytes, B lymphocytes, activated dendritic cells, high endothelial venules, myeloid cells and the PD-L1 immune checkpoint showed sex and age associated differences. Bladders from healthy older female mice showed a higher number of tertiary lymphoid structures (TLSs) compared to both young female and male equivalents. Spatial immune profiling of N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN) carcinogen exposed male and female bladders from young and old mice revealed a similar frequency of TLS formation, sex differences in the bladder immune microenvironment and, age differences in latency of tumor induction. These findings support the incorporation of sex and age as factors in pre-clinical modeling of NMIBC and will potentially advance the field of immunotherapeutic drug development to improve clinical outcomes in NMIBC.
