Transplanted pluripotent stem cell derived-islets engraft superior to human islets

ABSTRACT: To investigate gene expression differences between human stem cell-derived islets and human islets before transplantation. We performed gene expression profiling analysis using data obtained from RNA-seq of 5 different batches of stem cell-derived islets and human islets from 7 donors.

Project description:Human pluripotent stem cell-derived islets (hPSC-islets) are a promising cell resource for diabetes treatment. Here, we demonstrate that transplantation of human pluripotent stem cell-derived islets into diabetic nonhuman primates effectively restored endogenous insulin secretion and improved glycemic control. Single-cell RNA sequencing analysis of S6D2 clusters confirmed the existence of the three major pancreatic endocrine cell populations (β cells, α-like cells and δ-like cells) and their proportions, which altogether accounted for 80%. Importantly, hierarchical clustering of S6D2 hCiPSC-islets, 10 wpt kidney grafts and primary human islets showed that the hCiPSC differentiated pancreatic endocrine cells shared similar global gene expression profiles to their native counterparts in primary human islets. Single-cell RNA sequencing analysis on PBMCs revealed the potential immune response of recipient macaque to hCiPSC-islets.

Scale Up Manufacturing Approach for Production of Human Induced Pluripotent Stem Cell-Derived Islets Using Vertical Wheel®Bioreactors

Project description:We utilized the 10x Genomics Chromium Single Cell 3’ RNAseq platform to examine the genomic makeup of human induced pluripotent stem cell-derived islets following their differentiation under full suspension conditions using PBS vertical wheel bioreactors. Our investigation aimed to assess the efficacy of the SC-islet differentiation process in suspension, transitioning from a 0.1L to a scaled-up 0.5L volume. Our primary objective was to showcase the effectiveness of a modified protocol for pancreatic endocrine differentiation, with a specific focus on the final stage of maturation during scale-up. To gain a comprehensive understanding of these developmental changes, we integrated our transcriptomic analyses post the final maturation process with transcript data from human donor islets and existing SC-islet datasets.
