Transcriptome profiling of bladders of CYP-treated C57BL6/J mice (male and female)

ABSTRACT: In the present study, we aimed to identify a complete transcriptome profile of the bladders of CYP-treated mice, a commonly used animal model of IC/BPS disease. Our study represents a reference database for the understanding of the mouse model, which is crucial for the more successful translation of the data obtained on this model to human patients.

Project description:To analyze the gene expression of non-bacterial bladdder inflammation, mouse cyclophosphamide(CYP)-induced model of cystitis was adapted. Forty-eight hour after CYP or vehicle treatment, total RNA was extracted from CYP-treated group (CYP150) and control group (CYP0). We used microarrays to detail the gene expression underlying bladder inflammation. Five-week-old female Balbc/A mice were injected intraperitoneally with 150mg/kg body weight of CYP diluted in 15ml/kg body weight of saline. Mice injected with saline alone served as controls. Treated mice were killed 48 hours after administaration and the whole bladders harvested. Total RNA was extracted and microarray analysis was performed.

Peptidomics analysis of urine from IC/BPS patients and healthy controls

Project description:Interstitial cystitis/bladder pain syndrome (IC/BPS) is a chronic and debilitating pain disorder of the bladder and urinary tract with poorly understood etiology. A definitive diagnosis of IC/BPS can be challenging because many symptoms of IC/BPS are shared with other urological disorders. An analysis of urine presents an attractive and non-invasive resource for monitoring and diagnosing IC/BPS. Here, a non-targeted LC-MS and LC-MS/MS-based peptidomics analysis of urine samples collected from IC/BPS patients were compared to urine samples from asymptomatic controls.

Transcriptome profiling of TNFa-stimulated urothelial cells

Project description:In this study, we aimed to determine the overall effect of TNFα on transcriptomic profile of urothelial cells using RNA sequencing. Our study represents a reference database that could lead to a better understanding of the role of urothelial cells in IC/BPS pathology and more importantly, sets grounds for future studies exploring potential biomarkers and therapeutic targets for IC/PBS diagnosis and treatment.

Expression data from the bladders of carcinogen-treated mice with genetic Hh pathway manipulation

Project description:Attenuation of Hedgehog (Hh) pathway activity leads to accelerated tumor progression in a mouse model of N-butyl-N-4-hydroxybutyl nitrosamine (BBN) – induced bladder carcinoma. In order to identify genes regulated by the Hh pathway that might be involved in bladder cancer progression, we performed transcriptional profiling of bladders harvested from mice after BBN-exposure, comparing Gli1CreER/WT; Smoflox/WT mice to Gli1CreER/WT; Smoflox/flox mice, which express CreER under control of the Gli1 promoter and carry one or two floxed alleles of the essential Hh pathway transductory component Smoothened (Smo) respectively. Administration of Tamoxifen to these mice results in attenuation of Hh pathway activty to a greater extent in the Gli1CreER/WT;Smoflox/flox mice as compared to Gli1CreER/WT;Smoflox/WT mice, allowing identification of Hh-pathway regulated genes. 6 total samples were analyzed. 3 bladders from Gli1CreER/WT; Smoflox/WT mice and 3 bladders from Gli1CreER/WT; Smoflox/flox mice were analyzed.

Gene expression of unperturbed young and aged mouse bladders

Project description:Aging has multifaceted effects on the immune system, but how aging affects tissue-specific immunity is not well-defined. Bladder diseases characterized by chronic inflammation are highly prevalent in older women, but mechanisms by which aging promotes these pathologies remain unknown. Tissue transcriptomics of unperturbed, young, and aged bladders identified a highly altered immune landscape as a fundamental feature of the aging female bladder. Detailed mapping of immune cells using single cell RNA- sequencing revealed novel subsets of macrophages and dendritic cells and unique changes to the immune repertoire in the aged bladder. B and T cells are highly enriched in aged bladders and spontaneously form organized bladder tertiary lymphoid tissues (bTLTs).
