Project description:The change in gene expression on the 8th day of gestation was investigated using DNA microarrays. Uterine gene expression of interimplanted sites was analyzed in female mice. Female mice, C57BL/6jXOla129 genetic background were used for experiments. Mice, housed in sterile filter-top cages (3-4 per cage), were acclimatized in a room maintained at 24ºC with a 12-h light-dark fr four months. Two groups were established: a) The control group and b) homozygous Cbs deficient mice and sacrificed at the eight day post-coitum.

Neonatal Genistein Exposure Causes Implantation Failure by Disrupting Uterine Developmental Patterning and Endometrial Gland Formation

Project description:Female mice exposed neonatally to the phytoestrogen genistein (GEN) at doses similar to those in infants consuming soy-based infant formulas are infertile due in part to uterine implantation defects. To determine the mechanisms by which neonatal GEN exposure causes implantation defects, female mice were exposed to vehicle (corn oil) or GEN in corn oil (50 mg/kg/day) on postnatal days (PND) 1-5. Uterine tissues were collected during early gestation for microarray analysis; gestation day 1.5 (GD1.5), GD3.5, GD4.5 and GD5.5. We conclude that neonatal GEN exposure disrupts expression of genes important for uterine development, causing posteriorization and diminished gland function during pregnancy that together cause implantation failure.

Transcriptomic of early embryonic invasion at implantation sites in a murine model.

Project description:Trophoblast Invasion is a complex mechanism that involves several genes and processes that are exquisitely regulated by the mother. Functional genomics has shown that immunomodulatory and proliferation are two essential key processes involved. Adult virgin B6CBA F1/J female mice were mated with CD1 fertile males to induce pregnancy (day 0=vaginal plug). Mice were sacrificed by cervical dislocation. Implantation and inter-implantation sites were divided by sharp dissection 6.5 days post-coitum (dpc) (n=3 mice). Uterine segments included uterine myometrium, stroma, and epithelium. Inter-implantation decidua (ID) were collected and Implantation sites also were divided by sharp dissection to separate extra-embryonic tissue (ET) from surrounding decidua (SD). From the embryo, only the invasive trophoblast formed by the ectoplacental cone was studied too.
