In situ tumor arrays reveal early environmental control of cancer immunity [STAMP D8]

ABSTRACT: KPP-GFP (C57Bl/6) cells were implanted into the laser-induced micropores ears of B6.Cg-Foxn1nu/J (C57Bl/6J Jackson background) that were adoptively transferred with 4x10^6 naive CD4tdTomato-positive Tcells (B6 N/J background) the same day. The STAMP microtumors were imaged daily starting at day 3 after tumor cell implantation. On day 8 after tumor cell implantation tumors were classified as either (1) Inflamed, (2) Excluded, (3) Ignored/Desert or (4) Resolved.

Phosphoproteomics of mouse cardiomyocytes reveals signalling pathways triggered by a vasodilator cryptide

Project description:Animal venoms are natural sources of bioactive molecules such as cryptides that consist in biological active peptides released after proteolytic cleavage of a peptide or protein precursor. The KPP is a cryptided derived from the c-terminal of an hypotensin (TsHpI) a peptide isolated from the venom of Tityus serrulatus scorpion. Previous studies of our group in rodent model demonstrated that KPP maintained the cardioprotective properties of its precursor. Nonetheless, the molecular machinery involved in the cellular response is poorly described to date. Thus, in order to characterize the signalling pathways triggered by KPP we incubate mouse cardiomyocytes with KPP during 5 or 30 min and explore the cell proteome and phosphoproteome by dimethyl labelling quantitation.

Project description:C57BL/6 mice were orthotopically implanted with PK5L1940 cells. 8 Gy RT was administered 7 days post-implantation. PD1-IL2v and aCD25 dosed once per week beginning day 7 post-implantation. Spleens and lymph nodes were harvested 17 days post implantation and the sorted T-cells cultured in the presence of 13C6 glucose for 6h. This study contains longitudinal sampling of the cell culture supernatants.

Modeling Human Cancer-induced Cachexia (Mouse)

Project description:Cachexia is a wasting syndrome characterized by pronounced skeletal muscle loss. In cancer, cachexia associates with increased morbidity and mortality and decreased treatment tolerance. Although advances have been made in understanding the mechanisms of cachexia, translating these advances to the clinic has been challenging. One reason for this shortcoming may be the current animal models that fail to fully recapitulate the etiology of human cancer-induced tissue wasting. Because pancreatic ductal adenocarcinoma (PDA) presents with a high incidence of cachexia, we engineered a mouse model of PDA, that we named KPP. KPP mice, similar to PDA patients, progressively lose skeletal and adipose mass as a consequence of their tumors. In addition, KPP muscles exhibit a similar gene ontology to cachectic patients. We envision the KPP model will be a useful resource for advancing our mechanistic understanding and ability to treat cancer cachexia.

Project description:Proteins from post-nuclear cell lysates from C57BL/6J WT, CD38-deficient and B6.C-H2bm12/KHEg (bm12) recipient mice at 2 weeks after the adoptive transfer of co-isogenic spleen cells were identified using a proteomic approach. The relative abundances of the proteins identified were calculated using emPAIs scores.
