Differential gene expression in the grf10Δ mutant of Candida albicans in high copper containing growth medium

ABSTRACT: To identify genes in that are misregulated in the grf10Δ mutant of Candida albicans in response to growth in high copper, we compared expression patterns of the grf10-null mutant relative to the isogenic WT in YPD medium supplemented with and without 12 mM CuSO4.

Project description:When using YPM media, the carotenoid yield was increased 10-fold compared to using the YPD media. To elucidate the hidden mechanism, the transcriptome analysis was performed and showed that 464 genes changed significantly in YPM media. Furthermore, inspired by the differential gene expression analysis which indicated that ADY2, HES1, and CUP1 showed the most remarkable changes, we found that the improvement of carotenoid accumulation in YPM media was mainly due to the copper, since supplementation of 80 µM CuSO4 in YPD media could increase carotenoid yield 9.2-fold

To gain insight into the cellular processes that are modulated by copper abundance in C. albicans, we monitored the global gene expression dynamic under both copper depletion and excess using RNA-seq.

The Two-component Signal Transduction System CopRS of Corynebacterium glutamicum is Required for Adaptation to Copper-excess Stress

Project description:Copper is an essential cofactor for many enzymes but at high concentrations it is toxic for the cell. Copper ion concentrations ≥50 µM inhibited growth of Corynebacterium glutamicum. The transcriptional response to 20 µM Cu2+ was studied using DNA microarrays and revealed 26 genes that showed a ≥3-fold increased mRNA level, including cg3280-cg3289. Several genes in this genomic region code for proteins presumably involved in the adaption to copper-induced stress, e. g. a multicopper oxidase and a copper-transport ATPase. In addition, this region includes the copRS genes (previously named cgtRS9) which encode a two-component signal transduction system composed of the histidine kinase CopS and the response regulator CopR. Deletion of the copRS genes increased the sensitivity of C. glutamicum towards copper ions, but not to other heavy metal ions. Using comparative transcriptome analysis of the ΔcopRS mutant and the wild type in combination with electrophoretic mobility shift assays and reporter gene studies the CopR regulon and the DNA-binding motif of CopR were identified. Evidence was obtained that CopR binds only to the intergenic region between cg3285 and cg3286 in the genome of C. glutamicum and activates expression of the divergently oriented gene clusters cg3285-cg3280 and cg3286-cg3289. Altogether, our data suggest that CopRS is the key regulatory system in C. glutamicum for the extracytoplasmic sensing of elevated copper ion concentrations and for induction of a set of genes capable of diminishing copper stress. Four or five biological replicates of each experiment were performed. Experiment 1: Transcriptome comparison of wild type grown with 1.25 µM or with 21.25 µM CuSO4; Exp. 2: WT vs. copRS deletion mutant grown with 21.25 µM CuSO4; For analysis via DNA microarrays, the RNA was isolated from exponentially growing cells cultivated in CgXII medium containing glucose as carbon source and either 1.25 uM CuSO4 or 21.25 uM CuSO4.

The gene expression difference in strain AT01, a mutant of Trichoderma reesei, with high ability of copper accumulation

Project description:The gene expression profile of AT01 was compared with a wild-type T. reesei strain in medium containing 0.7 mM Cu2+. A total of 627 genes were up-regulated and 305 genes were down-regulated under copper stress. The expression analysis showed that amino acid transport- and metabolism-related genes are up-regulated, which is associated with adenine metabolism. Furthermore, inorganic ion transport- and metabolism-associated genes, such as superoxide dismutase and copper transporters, were also up-regulated. Strain AT01 and wild-type strain were inoculated into YPD medium containing 1 mM CuSO4, and cultured at 25 M-BM-0C for 5 d. Each sample was carried out with two repetitions.

Cytosolic copper concentration modulates spore germination and secondary metabolism in Streptomyces coelicolor

Project description:We studied the influence of copper in physiological and morphological differentiation of Streptomyces coelicolor. We demonstrate differences in phenotype (germination, growth rate, antibiotic production) and genetic expression between a strain mutated at copper chaperone CopZ (SCO2730::Tn5062), the wild-type strain and a wild-type strain sporulated in a media with 80µM CuSO4. These differences are correlated with the cytosolic copper. Our results demonstrate a pleiotropic effect of copper modulating S. coelicolor development.
