Serological autoantibody profiling of emmetropic controls and highly myopic patiens

ABSTRACT: High myopia is a leading cause of blindness worldwide, among which pathologic myopia, characterized by typical myopic macular degeneration, is the most detrimental. However, its pathogenesis remains largely unknown. By using a protein microarray assaying more than 20,000 human proteins, we determined in this study the autoantibody profiles of highly myopic patiens and emmetropic controls.

microRNA profiling in the aqueous humor of highly myopic eyes by next generation sequencing

Project description:Purpose: To characterize microRNAs (miRNAs) and their possible roles in high myopia by using next generation sequencing Methods: Aqueous humor samples were obtained from 15 highly myopic eyes and 15 cataract eyes at the onset of surgery. miRNA next generation sequencing and bioinformatics analyses were performed using RNA extracted from aqueous humor samples. Results: A total of 341 miRNAs were detected in the aqueous humor samples of highly myopic eyes; 201 miRNAs were detected in the aqueous humor samples of cataractous control eyes. A total of 249 mature miRNAs and 17 novel miRNAs were differentially expressed during myopia. Possible pathways regulated by these aberrantly expressed miRNAs included the TNF, MAPK, PI3K-Akt, and HIF-1 signaling pathways. Conclusions: The current study provided an overall view of miRNA profiling in the aqueous humor of highly myopic eyes. These profiles may be associated with myopia pathogenesis, and are potential biomarkers.

Project description:Glaucoma is the leading cause of irreversible blindness worldwide. An early diagnosis of the disease is crucial for treatment success. Mechanisms behind glaucoma pathogenesis are not completely understood, but disease related alterations in the serological autoantibody profile indicate an immunologic component. These changes in immunoreactivity may serve as potential biomarkers. Therefore, a serological proteome analysis (SERPA) approach was used to identify novel autoantibody biomarker candidates in primary open angle glaucoma. Autoantibodies in individual sera samples of glaucoma patients and control were analysed by serological proteome analysis (SERPA) using porcine trabecular meshwork (TM) tissue lysates as antigen source. Immunoblot spot intensities were computed by “Delta2D” (Decodon) and spots of interest were matched with preparative 2DE gels and identified by mass spectrometry to identify the potential autoantigens.

Insights into myopic choroidal neovascularization based on quantitative proteomics analysis of the aqueous humor

Project description:Previous studies on the biomarkers of pathologic myopia choroidal neovascularization (pmCNV) development merely detected limited types of proteins and provide a meagre illustration of the underlying pathways. Hence, an intact picture of molecular changes in the aqueous humor (AH) of pmCNV patients is lacking. Here, to explore the potential mechanisms and biomarkers of pmCNV, we analyzed the correlation between atrophic (A) lesions and neovascular (N) lesions in a myopic cohort based on the ATN grading system for myopic maculopathy (MM). In a quantitative proteomics test of AH profile of our patients, a significant correlation was found between A and N lesions (R = 0.5753, P < 0.0001). Accordingly, groups were divided into patients without MM, patients with myopic atrophic maculopathy (MAM), and patients with pmCNV (N2a lesion). In proteomics analysis, the existence of GFAP and complement-associated molecules in AH samples of the 3 groups also supported that MAM and pmCNV shared similar characteristics. Analyses of the differentially expressed proteins between the pmCNV group and two controls were performed. We identified several potential biomarkers for pmCNV, including FCN3, GFAP, EGFR, SFRP3, PPP2R1A, SLIT2, and CD248. Taken together, our study provides new insights for further research on pmCNV development.

Although the pathogenesis of myopia remains controversial, proteomics studies suggest that dysregulation of retinal metabolism is potentially involved in the pathology of myopia. Lysine acetylation of proteins plays a key role in regulating cellular metabolism, but little is known about its role in the form-deprived myopic retina. In this study, we performed acetylation proteomic analysis of the retinas of form-deprived myopic guinea pigs and found downregulated levels of acetylation of metabolism-critical enzymes in the retina. As the first report on retinal acetylation in myopic eyes, this study provides a reliable basis for further studies on retinal acetylation in myopic eyes

Evaluation of MicroRNA Expression Profiles for Form-Deprivation Myopia in Mouse

Project description:The study aim was to determine microRNA (miRNA) expression profiles of ocular tissues in form-deprivation induced myopia (FDM) mice. Form-deprivation myopia was induced in C57BL/6Jmice over the right eye; the contralateral left eyes were used as controls. Whole genome microRNA expression profiles in myopic whole eye, retina, and sclera were determined using the Agilent mouse miRNA microarray. The normalized microarray data were performed ANOVA test to identify differences in miRNA expression between myopic and control eyes. The differential expression for selected miRNAs was validated by quantitative real time PCR (qRT-PCR).

Large-scale microRNA expression profiling identifies retinal miRNA-mRNA signaling pathways underlying form-deprivation myopia in mice

Project description:Development of myopia is associated with large-scale changes in ocular tissue gene expression. Although differential expression of coding genes underlying development of myopia has been a subject of intense investigation, the role of non-coding genes such as microRNAs in the development of myopia is largely unknown. In this study, we explored myopia-associated miRNA expression profiles in the retina and sclera of C57Bl/6J mice with experimentally induced myopia using microarray technology. We found a total of 53 differentially expressed miRNAs in the retina and no differences in miRNA expression in the sclera of C57BL/6J mice after 10 days of visual form deprivation, which induced -6.93 ± 2.44 D (p < 0.000001, n = 12) of myopia. We also identified their putative mRNA targets among mRNAs found to be differentially expressed in myopic retina and potential signaling pathways involved in the development of form-deprivation myopia using miRNA-mRNA interaction network analysis. Analysis of myopia-associated signaling pathways revealed that myopic response to visual form deprivation in the retina is regulated by a small number of highly integrated signaling pathways. Our findings highlight substantial involvement of miRNAs in the regulation of refractive eye development, and in the development of myopia through the retinal gene regulation.

Etiologically, COPD is mediated by toxic gases and particles, e.g. cigarette smoke, while the pathogenesis of the disease is largely unknown. Several lines of evidence indicate a link between COPD and autoimmunity but comprehensive studies are lacking. By using a protein microarray assaying more than 19,000 human proteins we determined in this study the autoantibody profiles of COPD and non-COPD smokers.

Comparing lens epithelium gene expression profiles between highly myopic and emmetropic eyes

Project description:Clinical observations suggest increased size of lens in highly myopic subjects compared with emmetropic subjects. The layer of anterior lens epithelial cell membrane, being the most metabolically active part of the lens, can be easily acquired during standard lens surgery and is an ideal sample to study the gene expression difference between lens of different populations. In this study, to investigate the possible molecular mechanisms underlying different features of lens between highly myopic and emmetropic subjects, gene expression profiling of lens epithelia of corresponding population was conducted.
