WhiB1 transcriptone in response to NO stress

ABSTRACT: Mycobacterium tuberculosis is the etiological agent of tuberculosis. Mtb is an efficient pathogen partially due to its ability to adapt to the disparate conditions encountered during the course of infection. NO is one of the potent antimicrobial chemicals produced in hosts to tackle pathogens. Mtb is known to respond swiftly to NO-stress. Since, WhiB1 is [4Fe-4S] cluster containing transcription factor with high sensitivity to NO, we evaluate the role of WhiB1 in regulating the cellular transcriptional profile of Mtb in presence or absence of NO.

Project description:“Viable but non-culturable” (VBNC) states pose challenges for environmental and clinical microbiology, but their biological mechanisms remain obscure. Mycobacterium tuberculosis (Mtb), the leading cause of death from infection until COVID-19, affords a striking example. Mtb can enter into a “differentially detectable” (DD) state associated with phenotypic antimicrobial resistance in which Mtb cells are viable but undetectable as colony-forming units. We found that Mtb cells enter the DD state when they undergo sublethal oxidative stress that damages their DNA, proteins, and lipids, and in addition, their replication is delayed, allowing repair. Mycobacterium bovis and BCG fail to enter the DD state under similar conditions. These findings have implications for TB latency, detection, relapse, treatment monitoring, and development of regimens that overcome phenotypic antimicrobial resistance.

Genome-wide screening of Mycobacterium tuberculosis-infected macrophages identified the GID/CTLH complex as a determinant of intracellular bacterial growth

Project description:The eukaryotic GID/CTLH complex is a highly conserved E3 ubiquitin ligase involved in a broad range of biological processes. However, a role of this complex in host antimicrobial defenses has not been described. We exploited Mycobacterium tuberculosis (Mtb) induced cytotoxicity in macrophages in a FACS based CRISPR genetic screen to identify host determinants of intracellular Mtb growth restriction. Our screen identified 5 (GID8, YPEL5, WDR26, UBE2H, MAEA) of the 10 predicted members of the GID/CTLH complex as determinants of intracellular growth of both Mtb and Salmonella serovar Typhimurium. We show that the antimicrobial properties of the GID/CTLH complex knockdown macrophages are mediated by enhanced GABAergic signaling, activated AMPK, increased autophagic flux and resistance to cell death. Meanwhile, Mtb isolated from GID/CTLH knockdown macrophages are nutritionally starved and oxidatively stressed. Our study identifies the GID/CTLH complex activity as broadly suppressive of host antimicrobial responses against intracellular bacterial infections.

Proteomic and epi/proteomic response of Mycobacterium tuberculosis to nitric oxide stress

Project description:Reactive nitrogen species (RNS) are the major antimicrobial molecules secreted by host cells to tackle Mycobacterium tuberculosis (Mtb) infection. Mtb respond and adapt to adverse environmental stressors by changes in gene expression, ensuring their survival. This adaptation process is mediated by a combination of transcriptional regulatory networks that result in altered gene expression and enzyme (protein) activities. Previous studies emphasized primarily on transcriptomic response of Mtb to different environmental stresses, including RNS. In this study, the proteomic response of Mtb to sublethal doses of RNS stress after 30 minutes, 2 hours, 6 hours and 20 hours was explored. We used liquid chromatography coupled with electrospray ionization mass spectrometry (LC-MS/MS) to elucidate the proteomic response of Mtb to RNS. Quantitative proteomic analysis revealed identification of 2911 proteins and 6460 acylations in Mtb. A total of 581 unique proteins were found to be affected by RNS at different time points. Multiple-sample test revealed 297 differentially regulated proteins across the four time points. Furthermore, multiple-sample test identified 127 differentially acylated sites on 94 proteins. Marked changes in proteomic response was observed at later time points, most of which were compensatory responses involved in SOS response, iron homeostasis, energy metabolism and other stress responses. This study provides an improved understanding of the dynamic proteomic response of Mtb to RNS and identification of key molecular switches, which is the basis of controlling bacterial growth in different environmental setting.

Lung epithelial signaling mediates early vaccine-induced CD4+ T cell activation and Mtb control

Project description:Tuberculosis (TB) is the leading cause of death due to a single infectious agent. The development of a TB vaccine that induces durable and effective immunity to Mycobacterium tuberculosis (Mtb) infection is urgently needed. Complete and early Mtb control can be induced in M. bovis Bacillus Calmette–Guérin (BCG) vaccinated hosts when the innate immune response is targeted to generate effective vaccine-induced immunity. In the present study, we show that the superior and early Mtb control that is mediated by innate activation of DCs is associated with mucosal localization of clonal activated vaccine-induced CD4+ T cells in the lung. Our studies also show that the lung epithelial cell signaling through the activation of nuclear factor kappa-light-chain-enhancer of activated B cells (NFKB pathway is critical for the mucosal localization of activated vaccine-induced CD4+ T cells for rapid Mtb control. Thus, our study provides novel insights into the immune mechanisms that can overcome TB vaccine bottlenecks and provide early rapid Mtb control.

Novel Role for Macrophage Galactose-type Lectin-1 to Regulate Innate Immunity against Mycobacterium tuberculosis

Project description:Tuberculosis (TB) caused by infection with Mycobacterium tuberculosis (Mtb) is characterized by inflammatory pathology and poorly understood mechanisms of innate immunity. Pattern recognition receptors (PRR), expressed on the surface of macrophages, determine the balance of inflammatory and antimicrobial functions that influence disease outcome. Carbohydrate moieties displayed by mycobacteria can serve as PRR ligands for some members of the C-type lectin receptor (CLR) family; interactions that mediate a variety of incompletely understood immune outcomes. This work identifies a novel role for the CLR Macrophage Galactose-type Lectin-1 (MGL-1) in a mouse model of experimental tuberculosis. Murine macrophages upregulated MGL-1 following in vitro exposure to Mtb, while MGL+ cells accumulated at sites of mycobacteria-driven inflammation in the lung. Pulmonary macrophages from MGL-1-deficient mice displayed increased production of pro-inflammatory cytokines (IL-1b, IL-6 and IFN-g) that was associated with greater lipid accumulation, following Mtb infection. Surprisingly for a CLR, we also observed MGL-1-dependent anti-mycobacterial activity as evidenced by greater Mtb proliferation in BMDM, and the lung, of MGL-1 deficient mice. These results identify MGL-1 signaling as an important mechanism that regulates innate immunity against Mtb and indicate potential for the MGL pathway as a novel therapeutic target for anti-TB immunity.

IL-36/LXR axis modulates cholesterol metabolism and immune defense to Mycobacterium tuberculosis

Project description:Mycobacterium tuberculosis (Mtb) is a life-threatening pathogen in humans. Bacterial infection of macrophages usually triggers strong innate immune mechanisms, including IL-1 cytokine secretion. The newer member of the IL-1 family, IL-36, was recently shown to be involved in cellular defense against Mtb. To unveil the underlying mechanism of IL-36 induced antibacterial activity, we analyzed its role in the regulation of cholesterol metabolism, together with the involvement of Liver X Receptor (LXR) in this process. Here we report that, in Mtb-infected macrophages, IL-36 signaling modulates cholesterol biosynthesis and efflux via LXR. Moreover, IL-36 induces the expression of cholesterol-converting enzymes and the accumulation of LXR ligands, such as oxysterols. Ultimately, both IL-36 and LXR signaling play a role in the regulation of antimicrobial peptides expression and in Mtb growth restriction. These data provide novel evidence for the importance of IL‑36 and cholesterol metabolism mediated by LXR in cellular host defense against Mtb.

Bulk RNA-seq analysis of Mycobacterium tuberculosis infected Sp140-sufficient and -deficient mouse lungs with and without pDC depletion

Project description:Mycobacterium tuberculosis (Mtb) causes 1.5 million deaths annually. Active tuberculosis correlates with a neutrophil-driven type I interferon (IFN) signature, but the underlying cellular mechanisms remain poorly understood. We found that interstitial macrophages (IMs) and plasmacytoid dendritic cells (pDCs) are dominant producers of type I IFN during Mtb infection in mice and non-human primates, and pDCs localize near human Mtb granulomas. Depletion of pDCs reduces Mtb burdens, implicating pDCs in tuberculosis pathogenesis. During IFN-driven disease, we observe abundant DNA-containing neutrophil extracellular traps (NETs) known to activate pDCs. Single cell RNA-seq indicates that type I IFNs act on IMs to impair their responses to IFNg, a cytokine critical for Mtb control. Cell type-specific disruption of the type I IFN receptor suggests IFNs act on IMs to inhibit Mtb control. We propose pDC-derived type I IFNs, driven by NETs, act on IMs to drive bacterial replication, further neutrophil recruitment, and active tuberculosis disease.
