Project description:Understanding the molecular mechanisms driving lineage decisions and differentiation during development is especially difficult in systems with a diverse progenitor pool, such as the mammalian cerebellum. Here, we used single-cell RNA-sequencing (scRNA-seq) to characterise the developmental trajectories of Nestin-expressing progenitors (NEPs) in the neonatal mouse cerebellum. We identified FOXO1 as a key regulator of NEP-to-inhibitory neuron differentiation, acting directly downstream of ASCL1. Genome occupancy and functional genomic experiments using primary NEPs showed that both ASCL1 and FOXO1 regulate neurogenesis genes during in vitro differentiation, while independently regulating proliferation and survival, respectively. Furthermore, we demonstrate that WNT is a regulator of the transition from an ASCL1+ to FOXO1+ state. Finally, we show that the ASCL1-FOXO1 axis and role of WNT signalling is conserved in primary human NEPs. Through understanding how cerebellar interneurons differentiate, our findings may inform therapeutic strategies for cerebellar disorders such as spinocerebellar ataxia, where cerebellar interneurons are overproduced.

Project description:Bergmann glial cells of the vertebrate cerebellum play essential roles in the development and maintenance of cerebellar structure and function. During development, Bergmann glia provide structural support to the expanding cerebellar anlage and also serve as guides for migrating neurons (granule cells). As the cerebellum matures, Bergmann glia become important in dendritic arborization, synapse maintenance and synaptic function. The molecular mechanisms underlying these diverse and important functions of Bergmann glia remain largely unknown. We used microarray analysis to examine global gene expression in individual Bergmann glial cells derived at P6 (a time of extensive neuronal migration and cerebellar growth) and at P30 (when cerebellar development is complete and Bergmann glia play important roles at synapses). After gentle dissociation of cerebellar tissue derived from mice expressing GFP under the GFAP gene promoter (GFAP-GFP mice) (Zhuo, L., et al., 1997, Developmental biology), single GFP-positive Bergmann glial cells were aspirated into microcapillary tubes. Amplified cDNAs were prepared from single cells using RT-PCR and hybridized to Affymetrix GeneChip Mouse Genome 430 2.0 expression arrays (one array per cell). Five P6 cells and five P30 cells were used to generate the data presented in this study.

Here, we present a comprehensive atlas profiling 1,904 transcription regulators across organs, uncovering TOX3 as a potential co-activator of Atoh1 in cerebellar granule neuron progenitors. Tox3-deficient mice display severe ataxia and cerebellar hypoplasia, driven by depletion of granule neuron progenitors, diminished Atoh1 expression, and impaired primary cilia. Single-nucleus RNA-seq analyses reveals compromised maintenance of the progenitor pool. TOX3 is also highly expressed in subsets of medulloblastoma, and its deletion reduces cerebellar neoplasia and prolongs survival in a mouse model. Mechanistically, Tox3 and Atoh1 co-occupy and synergistically activate E-boxes in shared target genes by up to 120-fold, including an ultra-conserved E-box downstream of Atoh1. These findings establish Tox3 as a critical Atoh1 co-activator in cerebellar development, tumorigenesis and evolution, and provides a resource for exploring novel transcriptional regulators in progenitor maintenance and organogenesis.

All cell types of the cerebellum are affected, but the main sign of hypothyroidism in mice is the persistence of the external granular layer, composed of mitotic neuronal precursors at P21. To make the genetic link between thyroid hormone and cerebellar development, we sought to identify new thyroid hormone target genes, in particular in granule cells which represent the vast majority of cerebellar cells.

Project description:It is generally believed that cerebellar granule neurons originate exclusively from granule neuron precursors (GNPs) in the external germinal layer (EGL). Here we identify a rare population of neuronal progenitors in the developing cerebellum that expresses Nestin. Although Nestin is widely considered a marker for multipotent stem cells, these Nestin-expressing progenitors (NEPs) are committed to the granule neuron lineage. Unlike conventional GNPs, which reside in the outer EGL and proliferate extensively, NEPs reside in the deep part of the EGL and are quiescent. Expression profiling reveals that NEPs are distinct from GNPs, and in particular, express markedly reduced levels of genes associated with DNA repair. Consistent with this, upon aberrant activation of Sonic hedgehog (Shh) signaling, NEPs exhibit more severe genomic instability and give rise to tumors more efficiently than GNPs. These studies identify a novel progenitor for cerebellar granule neurons and a novel cell of origin for medulloblastoma.

Primary cultures of cerebellar neurons were made by dissociation of cerebella from newborn wild-type mice. These cells were plated 48 hours in serum-free medium to avoid invasion of the culture by glial cells. In order to include a kinetic and a maximum number of target genes, several cultures were either treated or left untreated as controls for 6 hours (T1), 16 hours (T2), 24 hours (T3) or 48 hours (T4) and results were pairwise compared for each time point.
