Defective cuticle-derived signals enhance extracellular ATP response and plant immunity

Project description:Extracellular ATP (eATP) acts as a damage-associated molecular pattern (DAMP) in both animals and plants. In plants, eATP is recognized by the purinergic receptors P2K1 and P2K2, which mediate responses to various environmental stresses. From a p2k1 suppressor screen, we identified two allelic mutants of the cutin biosynthesis gene, BODYGUARD (BDG). The BDG mutations specifically suppress p2k1 and p2k1p2k2 mutant defects in response to eATP. The suppression of p2k1 by bdg modifies plant defense against the necrotrophic fungal pathogen, Botrytis cinerea. In addition, the results show that the defective cuticle in bdg mutants generates cutin breakdown products, such as cutin monomers, which serve as signaling molecules to activate eATP response and plant immunity through an alternative eATP signaling pathway. Our data provide new insights into the signaling role of cutin and reveal the first link between the cuticle and eATP signaling.

L-Glutamic acid negatively regulates extracellular ATP-induced reactive oxygen species signaling

Project description:Extracellular ATP (eATP) and L-Glutamic acid (L-Glu) are important damage associated molecular pattern (DAMP) molecules released from cells following injury. Both molecules trigger wound-associated signal transduction pathways, as well as the enhanced production of reactive oxygen species (ROS) by the RESPIRATORY BURST OXIDASE HOMOLOG D (RBOHD) protein. However, whether eATP and L-Glu trigger overlapping or distinct pathways is mostly unknown. Here we report that Arabidopsis thaliana responses to eATP or L-Glu are distinct from each other in terms of tissue specificity and transcriptomic responses. Thus, although both DAMPs trigger the expression of multiple wounding and hormone response transcripts, systemically induced eATP and L-Glu response transcripts have little overlap between them. We further show that wounding of different tissues may result in ROS responses that are controlled by different DAMP receptors. Thus, activation of ROS production following injury of non-vascular tissues primarily depended on the eATP receptors PURINORECEPTOR 2 KINASE 1 and 2 (P2K1P2K2), while activation of ROS responses in vascular tissues primarily dependent on the L-Glu receptors GLU-LIKE RECEPTORS 3.3 and 3.6 (GLR3.3GLR3.6). Interestingly, we found that in the absence of the GLR3.3GLR3.6 receptors (i.e., in the glr3.3glr3.6 double mutant), the ROS response to eATP application is enhanced. This finding suggests that during wounding the L-Glu pathway may suppress the eATP pathway. Taken together, our findings suggest that the DAMP molecules eATP and L-Glu have complex interactions that appear to be both partly complementary and partly antagonistic, as well as tissue dependent.

Tumor-Associated Microglia Secrete Extracellular ATP to Support Glioblastoma Progression

Project description:Glioblastoma (GBM) is a highly aggressive brain tumor with poor prognosis and high recurrence rates. The complex immune microenvironment of GBM is highly infiltrated by tumor-associated microglia and macrophages (TAMs). TAMs are known to be heterogeneous in their functional and metabolic states and can transmit either pro-tumoral or anti-tumoral signals to glioma cells. Here, we performed bulk RNA-seq and single-cell RNA-seq on GBM patient samples, which revealed increased ATP synthase expression and oxidative phosphorylation (OXPHOS) activity in TAMs located in the tumor core relative to the tumor periphery. Both in vitro and in vivo models displayed similar trends of augmented TAM mitochondrial activity, along with elevated mitochondrial fission, glucose uptake, mitochondrial membrane potential, and extracellular ATP (eATP) production by TAMs in the presence of GBM cells. Tumor-secreted factors, including GM-CSF, induced the increase in TAM eATP production. Elevated eATP in the GBM microenvironment promoted glioma growth and invasion by activating the P2X purinoceptor 7 (P2X7R) on glioma cells. Inhibition of the eATP-P2X7R axis attenuated tumor cell viability in vitro and reduced tumor size and prolonged survival in glioma-bearing mouse models. Overall, this study revealed elevated TAM-derived eATP in GBM and provided the basis for targeting the eATP-P2X7R signaling axis as a therapeutic strategy in GBM.

An emerging epithelial to mesenchymal transition inducer: extracellular ATP activates and modulates EMT at levels of transcription, translation, and cell function independent of TGF-b

Project description:EMT is an early process in metastasis that is associated with up to 90% of cancer-related death. ATP has been known to play some roles in EMT. We previously described that internalization of extracellular ATP (eATP) by cancer cells in vitro and in vivo via macropinocytosis increases intracellular ATP levels, cell proliferation and resistance to anticancer drugs. Recently, we reported eATP also induces EMT and other early metastatic activities independent of TGF-b, a well-known inducer of EMT, in human lung cancer cells. However, exactly how eATP induces EMT at gene expression level is far from understood. In this study, we hypothesize that eATP acts as an inducer and regulator of EMT alternative to TGF-b. Transwell assays revealed that eATP treatment, alone, induced human lung cancer A549 and H1299 cells to invade at rates faster than TGF-b. eATP also induced formation of filopodia-like protrusions faster than TGF-b, and it restored viability of cancer cells treated with TGF-b-neutralizing antibodies. eATP significantly elevated intracellular ATP levels while TGF-b did not. RNA sequencing / western blots revealed that eATP induced changes in expression of genes/proteins involved in EMT similar to but with differences in number and induction time compared with those induced by TGF-b. Metabolomics analysis shows that eATP-induced major metabolic changes are consistent/correlated with those identified at the gene expression level. These differences could be accounted for by the multi-functional and multi-localization properties of eATP and macropinocytosis-internalized eATP, strongly suggesting that eATP is a previously unrecognized master inducer and regulator of EMT.

Extracellular ATP is an environmental cue in bacteria

Project description:<p>In animals and plants extracellular ATP (eATP) functions as signal and regulates the immune response. During inflammation intestinal bacteria are exposed to elevated eATP originating from the mucosa. However, whether bacteria respond to eATP is unclear. Here we show that non-pathogenic Escherichia coli responds to eATP by modifying its transcriptional and metabolic landscapes. A genome-scale promoter library showed that the response is time-, concentration-, and medium-dependent and ATP-specific. Second messengers and genes related to metabolism, biofilm formation, and envelope stress were regulated downstream of eATP. Metabolomics confirmed that eATP triggers enrichment of compounds with bioactive properties on the host or bacteria. Combined genome-scale modelling revealed global metabolic and biomass building blocks modifications. Consequently, eATP altered the sensitivity to antibiotics and antimicrobial peptides. Finally, in pathogens, eATP controlled virulence factor expression. Our results indicate that eATP is an environmental cue in prokaryotes which broadly regulates physiology, antimicrobial resistance, and virulence.</p>
