A549-derived exosomes transform fibroblasts into cancer-associated fibroblasts (CUT&RUN)

ABSTRACT: With the in-depth study of exosomes, it has been discovered that tumor-derived exosomes could transform normal fibroblasts into cancer associated fibroblasts (CAFs) and further affect metastasis and drug resistance of CAFs. FOXN3 was the target of tumor-derived exosomes. The understanding of downstream genes regulated by FOXN3 is nessesary. An alternative ChIP-seq method, CUT&RUN was applied to explored the downstream genes directly regulated by FOXN3

Project description:The communication between tumor-derived exosomes and stroma plays a critical role in facilitating cancer progression. We detected the mRNA expression levels after MRC5 was transfected with shRNAs targeting FOXN3 or incubated with A549 cells-derived exosomes. We reported that in lung cancer microenvironment, tumor cells exhibit a great capacity to convert normal fibroblasts (NFs) to cancer-associated fibroblasts (CAFs). We showed that tumor cells secrete a subset of exosomal microRNAs directly targeting FOXN3, which leads to the activation of fibroblasts. Activated CAFs further promote cancer growth by entering tumor tissues. These results demonstrate that intercellular crosstalk between tumor cells and fibroblasts is mediated by tumor-derived exosomes, providing potential targets for treatment of lung cancer.

Project description:Exosomal miRNAs secreted by cancer-associated fibroblasts (CAFs) play a critical role in facilitating head and neck cancer (HNC) progression. A few studies in other cancers have confirmed some CAF-specific miRNAs could be delivered to tumor cells via exosome, thus contributing to chemoresistance. To identify the specific miRNAs responsible for the capability of CAF-derived exosomes to promote cisplatin resistance in recipient HNC cells, a miRNA array was conducted to identify miRNAs involved in exosomes derived from CAFs and the paired normal fibroblasts (NFs).

microRNA sequencing in rat cardiac fibroblasts and fibroblast-derived exosomes

Project description:We found that cardiac fibroblasts produce and secrete exosomes. miRNA profiling and TaqMan qRT-PCR experiments identified miR-21 expression to be higher in cardiac fibroblasts compared to those of miR-21*, whereas in exosomes miR-21* expression was higher compared to miR-21. The purpose of the study was to validate these findings by miRNA sequencing in cardiac fibroblasts and fibroblasts-derived exosomes. Neonatal rat cardiac fibroblasts were cultured in DMEM + 1% exosome-depleted FBS for 48h. Conditioned medium was collected and exosomes were purified by several centrifugation and filtration steps, following ultracentrifugation. Afterwards total RNA from cardiac fibroblasts and exosomes was isolated for miRNA sequencing.

Next generation sequencing showing miRNA profiles of exosomes derived from DHT-treated CAFs and matched ETOH-treated CAFs

Project description:Background: Androgen deprivation therapy (ADT) is the backbone of therapy for advanced prostate cancer (PCa). Despite the good initial response, castration resistance and metastatic progression will inevitably occur. Cancer-associated fibroblasts (CAFs) may be implicated in promoting metastasis of PCa after ADT. Our aim is to investigate the role and mechanism of CAF-derived exosomes involving in metastasis of PCa after ADT. Methods: PCa cells were co-cultured with exosomes derived from DHT-treated or ETOH-treated CAFs, and their migration and invasion differences under castration condition were examined both in vitro and in vivo. The miRNA profiles of exosomes derived from DHT-treated CAFs and matched ETOH-treated CAFs were analysed via next generation sequencing. The transfer of exosomal miR-146a-5p from CAFs to PCa cells was identified by fluorescent microscopy. The function and direct target gene of exosomal miR-146a-5p in PCa cells were confirmed through Transwell assays, luciferase reporter, and western blot. Findings: Compared with DHT-treated CAFs, exosomes derived from ETOH-treated CAFs dramatically increase migration and invasion of PCa cells under castration condition. MiR-146a-5p level in exosomes from ETOH-treated CAFs was significantly reduced. The loss of miR-146a-5p may strengthen the epithelial-mesenchymal transition (EMT) to accelerate cancer cells metastasis by modulating epidermal growth factor receptor (EGFR)/ERK pathway. Interpretation: CAFs-derived exosomal miR-146a-5p confers metastasis in PCa cells under ADT through the EGFR/ERK pathway and it may present a new treatment for PCa.

Proteomics Identifies Differences in Fibrotic Potential of Exosomes from Human Tendon and Muscle Fibroblasts

Project description:Fibroblasts are the powerhouses responsible for the production and assembly of extracellular matrix. Their activity needs to be tightly controlled especially within the musculoskeletal system, where changes to extracellular matrix composition affect force transmission and mechanical loading that are required for effective movement of the body. Extracellular vesicles, including exosomes, are a mode of cell-cell communication within and between tissues, which has been largely characterised in cancer. However, it is unclear what the role of healthy fibroblast-derived extracellular vesicles is during tissue homeostasis. Here, we performed proteomic analysis of exosomes derived from primary human muscle and tendon cells to identify the potential functions of healthy fibroblast-derived exosomes. Mass spectrometry-based proteomics revealed comprehensive profiles for exosomes released from healthy human fibroblasts from different tissues. We found that fibroblast-derived exosomes were more similar than exosomes from differentiating myoblasts, but there were significant differences between tendon fibroblasts and muscle fibroblasts exosomes. Exosomes from tendon fibroblasts contain higher levels of proteins that support extracellular matrix synthesis, including TGFβ1, and muscle fibroblast exosomes contain a higher abundance of MMP2. Our data demonstrates a marked heterogeneity among healthy fibroblast-derived exosomes, indicating shared tasks between exosomes of skeletal muscle myoblasts and fibroblast, whereas tendon fibroblast exosomes has a marked fibrotic potential in human tendon tissue. These findings suggest an important role for exosomes in tissue homeostasis of both tendon and skeletal muscle in humans.
