Identification of TEAD1-Regulated MicroRNAs

ABSTRACT: The influence of TEADs on tumorigenesis, cancer immunity, drug resistance, metastasis and metabolism of various cancers have been well-studied. In recent years, microRNAs have been confirmed to regulate TEADs in several cancer types. However, it is still unclear how miRNAs orchestrate their regulation and function in TEAD1 network in hepatocellular carcinoma (HCC). Here, we investigated that TEAD1 is up-regulated in liver cancer and highly correlated with the effector YAP1, Smad2/3 of the Hippo, TGFβ pathway. Next, we used small RNA sequencing to characterize the differentially expressed miRNAs after the knockdown of TEAD1 in a time-dependent manner (0, 24, 36 and 48h) by using two siRNAs in HepG2. The most robustly TEAD1-induced 10 miRNAs with consistent trend over time were verified.
