Gene expression profile at single cell level of adipocyte progenitors from subcutaneous inguinal adipose depots

ABSTRACT: Stromal vascular fraction (SVF) derived from adipose tissue is enriched for adipocyte progenitor cells, it is composed of aheterogeneous populations of cells and cell types. We use single-cell RNA-Sequencing (scRNA-Seq) to deconvolution of cell types in adipocyte progenitors and a general hierarchical structure is emerging.

CRISPLD2 regulates adipocyte function

Project description:Obesity with accompanying adipose fibrosis is associated with worsening insulin resistance. Weight loss and adipose tissue turnover are conversly associated with improved insulin signaling and overall health. We previously found the secreted protein CRISPLD2 up-regulated in adipocytes during weight loss; CRISPLD has been suggested regulate inflammation and tissue turnover. As loss of CRISPLD2 is embryonic lethal, we developed mice with adipocyte-specific inducible CRISPLD2 overexpression to explore its function. Single-cell RNAseq analysis of the stromal vascular fraction (SVF) during long-term adipocyte CRISPLD2 overexpression demonstrated broad down-regulation of collagen transcription across multiple cell types; we found similar down-regulation of fibrosis via picosirius red staining of adipose tissue overexpressing CRISPLD2, along with a decrease in adipocyte cell size in chow-fed animals.

Thermogenic differentiation of adipocyte progenitors

Project description:FACS-purified adipocyte progenitors from murine subcutaneous adipose tissue were cultured under conditions promoting general adipogenic differentiation or beige/brite adipocyte differentiation (treatment with cPGI2). Time course expression profiling was performed during differentiation. In addition, some cultures of differentiated adipocytes were stimulated with norepinephrine for 3 hours. In parallel, differentiation and norepinephrine stimulation of progenitors from interscapular brown fat was performed and profiled.

A non-cell autonomous mechanism of beige adipocyte renaissance in subcutaneous adipose tissue

Project description:Two types of UCP1 positive cells-brown and beige adipocytes exist in mammals. Beige adipocytes are very plastic, and can be dynamically regulated by environment.Beige adipocytes formed postnatally in subcutaneous inguinal white adipose tissue (iWAT) lost thermogenic gene expression and multilocular morphology at adult stage, but cold could restore their “beigeing” characteristics, a phenomenon termed as beige adipocyte renaissance. Our results showed that beige cell maintenance and renaissance in adult mice were regulated by cAMP and HDAC4 signaling in white adipocytes non-cell autonomously. Genetic modulations of various components of this cAMP-HDAC4 cascade (e.g. LKB1) led to persistent browning and reduced adiposity independent of thermogenesis. To further study the mechanisms of beige adipocytes maintenance, we performed RNA-seq with samples from inguinal white adipose tissues of WT, AdipoqCre LKB1 F/F, and AdipoqCre LKB1 F/F; HDAC4 F/F mice.Our studies will move the beige adipocyte field forward and attract clinical applications to target beige adipocyte renaissance.

Sodium Alginate Activates Subcutaneous Fat Differentiation via the PPAR Pathway

Project description:To investigate the effect of sodium alginate on adipocyte differentiation, we isolated the stromal vascular fraction (SVF) from wild-type C57BL/6J mice and subjected it to whitening treatment. During the induction of differentiation, sodium alginate reagent at concentrations of 0 μM, 50 μM, and 100 μM was added to the experimental groups, respectively. On the sixth day of culture, cells from both the experimental and control groups were collected for transcriptome analysis.
