Project description:In recent years, the scale culture of Chinese soft-shelled turtle has developed rapidly. However, diseases in aquaculture are the main problems affecting the rapid and healthy cultivation. Strengthening the immunity of Chinese soft-shelled turtles is extremely important to control the infection of pathogenic bacteria. Bacillus has attracted attention as a probiotic supplement in aquatic feeds.In our previous studies, we found that the addition of Bacillus subtilis B10 to diets could increase survival rate, daily weight gain (DG) and feed conversion ratio (FCR) of Chinese soft-shelled turtles, improving the activities of digestive enzyme and optimizing the microbial communities of intestinal in Chinese soft-shelled turtle.However, the study on the mechanism of Bacillus subtilis B10 in Chinese soft-shelled turtle culture remains rare. Therefore, in this study, we used Bacillus subtilis B10 to feed the turtle, and used RNA-seq to explore its mechanism.

Project description:Purpose: The global effort to combat COVID-19 rapidly produced a shortlist of approved drugs with anti-viral activities for clinical repurposing. However, the jump to clinical testing was lethal in some cases since a full understanding of the mechanism of antiviral activity as opposed to inherent toxicity for these drugs was lacking. We used parallel lipidomic and transcriptomic analyses to investigate the effect of Niclosamide (NIC), a poorly soluble anti-helminth drug identified for repurposed treatment of COVID-19, on SARS-CoV-2 infected Vero E6 cells. Methods: A time of addition study was performed on VeroE6 cells to gain mRNA profiles of 16 and 48h SARS-CoV-2 and/or niclosamide infected cells in triplicate, using an Illumina NovaSeq6000 platfrom. Sequence reads that passed quality filters were analyzed by featurecounts and DESeq2. Results: Herein, we identified and characterized autophagic or lipophagic pathways (and wider linked cellular networks) that are targeted by NIC in the presence and absence of a SARS-CoV-2 infection. We used the lipid and gene expression signatures induced by this anti-parasitic compound to detect pathways that are predicted to either lead to direct antiviral effects or to cellular dysregulation and cell death. Conclusions: NIC treatment reduced the abundance of phosphatidylethanolamines (PE), a known activator of autophagy, only after establishment of SARS-CoV-2 infection. Countering SARS-CoV-2 subversion of lipid metabolism by NIC through the activation of PE may lead to a focused screen for approved, more druggable compounds that can suppress viral replication in COVID-19 patients.

Project description:In order to understanding the mechanisms of action of niclosamide, the active ingredient in the molluscicides, we used a Biomphalaria glabrata oligonucleotide microarray to investigate the effect of three sublethal exposures to niclosamide on the transcriptional responses of the snail B. glabrata relative to untreated snails. In addition to inducing biotransformation activity and inducing stress responses, niclosamide exposures also resulted in altered function of vesicle trafficking and may affect the snail’s oxygen binding capacity. In order to reveal how sublethal exposures of niclosamide to the snail B. glabrata affects the snail’s gene expression. The three sublethal doses of niclosamide tested are 0.15 mg/L (dose 1), 0.10 mg/L (dose 2), and 0.05 mg/L (dose 3). The control snails did not exposed to the niclosamid (0%). The snails were exposed to a given dose for 24 hours. Twenty live snails from each dose or control group were collected and randomly divided into four replicate groups, each with five snails, for subsequent RNA extraction and transcriptome analysis

The patterns of gene expression differed markedly among the three experimental groups. Normal embryonic development in normoxia was associated with up-regulation of genes involved in DNA replication, the cell cycle, and mitosis, but these genes were commonly down-regulated after incubation in hypoxia. Many target genes of hypoxia inducible factors, including insulin-like growth factor binding protein 1, were down-regulated by normoxic incubation but upregulated by incubation in hypoxia. Notably, some of the transcriptomic effects of hypoxia in green turtle embryos resembled those reported by others to be associated with hypoxia-induced embryonic arrest in diverse taxa, including budding yeast (Saccharomyces cerevisiae), the annelid Caenorhabditis elegans, and zebrafish (Danio rerio). Thus, while among oviparous species, hypoxia-induced pre-ovipositional embryonic arrest appears to be a unique adaptation of turtles, mechanisms underlying hypoxia-induced embryonic arrest per se may be highly conserved across the diverse taxa in which this mechanism operates. We report the transciptional changes in Chelonia mydas embryos incubated in either normoxia or hypoxia comapred to freshly laid eggs.

According gene expression profiles and mechanistic analysis, all evidences revealed niclosamide disrupted multiple metabolic pathways affecting biogenetics, biogenesis and redox regulation. These studies support niclosamide as a promising therapeutic agent for ovarian cancer. CP70sps cells were treated with niclosamide for 0, 2, 4 and 6 hours respectively, and then cells were harvested and analyzed their gene expression profiles.

Project description:ALS is a complex neurodegenerative disease influenced by genetic, epigenetic, and environmental factors, resulting in dysfunction in cellular and molecular pathways. The limited efficacy of current treatments highlights the need for combination therapies targeting multiple aspects of the disease. Niclosamide, an anthelminthic drug listed as an essential medicine, has been repurposed in clinical trials due to its anti-inflammatory and anti-fibrotic properties. Niclosamide can inhibit various molecular pathways (such as STAT3 and mTOR) that are dysregulated in ALS, suggesting its potential to disrupt these altered mechanisms associated with the pathology. Our recent findings have demonstrated that niclosamide can effectively inhibit inflammatory and fibrotic molecular pathways in ALS models. We administered niclosamide intraperitoneally to two transgenic murine models, FUS and SOD1-G93A mice, which replicate key pathologies and biological processes of ALS. The treatment was initiated at the onset of symptoms, and we assessed the progression of the disease by neurological scores, rotarod and grip tests, as well as monitoring survival. Additionally, we examined the effects of the treatment on spinal cord and muscle degeneration in the treated mice. In both models, the administration of niclosamide resulted in a slowdown of disease progression, an increase in survival rates, and an improvement in tissue pathology. This was characterized by a reduction in gliosis, motor neuron loss, muscle atrophy, and inflammatory markers. Based on these results, our findings demonstrate that niclosamide can impact multiple pathways involved in ALS. This multi-targeted approach leads to a slowdown in the progression of the disease, thereby positioning niclosamide as a promising candidate for repurposing in the treatment of ALS.
