Comparison of miRNA profiles between young (6 months) and old (24 months) male mouse vertebral samples

ABSTRACT: To understand the effect of age on miRNA profiles in male mouse vertebrae, we isolated L4-5 vertebral samples and digested with endotoxin-free collagenase (Liberase) twice for 30 minutes. The resulting bone was confirmed to be largely osteocyte-enriched and RNA was isolated using QIAzol reagent.

Gene expression-based, individualized outcome prediction for osteoporosis with vertebral fracture

MiRNAs array analysis was performed on the plasma samples including a pool of 6 miRNA samples from osteoporotic patients with vertebral fractures, a pool of 6 miRNA samples from osteoporotic patients without fracture and another pool of 6 miRNA samples from nonosteoporotic patients to identify regulated miRNAs in the plasma.

VRTN Regulates the Number of Thoracic Vertebrae During Vertebrate Development [ChIP-seq]

Project description:The number of vertebrae is precisely defined in almost all vertebrate species, but varies considerably in pigs, making this animal an excellent model for studying the mechanisms that control vertebral number. Vertnin (VRTN) variants have been associated with thoracic vertebral number (TVN) in pigs. However, the causal relation between VRTN and TVN remains to be established, and the role of VRTN in modulating TVN is not yet known. Here, we demonstrate that VRTN is one of the genes responsible for determining TVN. We show that VRTN is a DNA-binding transcription factor, which is essential for the formation of thoracic vertebrae during early embryogenesis as VRTN-null mice showed embryonic lethality at the later thoracic somite stages and had fewer somites than their wild-type and heterozygous littermates. We also show that VRTN causative variants increase Notch signaling in pig embryos, suggesting that VRTN controls segment number by altering the pace of somatic segmentation. These findings advance our understanding of the role of VRTN in the formation of thoracic vertebrae and reveal new aspects of somite developmental biology.

Scl-Ab: Exploratory 26-Week Subcutaneous Toxicology Study in the Aged Ovariectomized Female Sprague Dawley Rat with an 18-week Recovery

Project description:This study is designed to compare and contrast the temporal and spatial changes in bone formation rates and transcriptional profiles in cortical and cancellous bone cell populations enriched by laser capture microdissection (LCM) in ovariectomized rats administered Scl-Ab by subcutaneous injection for up to 26 consecutive weeks, followed by a recovery period of up to 18 weeks. Six-month-old Sprague-Dawley female rats were OVX, left untreated for 8 weeks. Rats were assigned to three treatment groups using a computerized blocking procedure designed to achieve body weight balance across treatment groups and then divided into three groups and administered by s.c. injection VEH or 3 or 50 mg/kg/wk of a Scl-Ab engineered to be less immunogenic in rats up to d183. Rats were administered either VEH or 3 or 50 mg/kg of Scl-Ab once weekly by s.c. injection. Up to 15 rats per group were euthanized at d8, 29, 85, and 183. To assess effects following treatment withdrawal, additional groups were maintained through a treatment-free period (TFP) and euthanized at d197 and 267 for VEH and 3 mg/kg groups and at d237 and 309 for VEH and 50 mg/kg groups. To label active bone-forming surfaces and facilitate osteoblast (OB) and lining cell (LC) enrichment during laser capture microdissection (LCM), 10 mg/kg calcein green was administered s.c. 13 and 3 days prior to scheduled euthanasia. At necropsy, the first and second lumbar vertebral (L1 and L2) bodies were isolated for histomorphometry and pQCT analyses, respectively. Cryosection preparation of L3 vertebral body, LCM procedure and validation of cell enrichment, RNA extraction and amplification, TaqMan, and initial microarray analysis was performed. LCM was performed on 4â6 ?m cryosections of vertebrae from the first 5 animals per treatment group per timepoint to enrich for OB, LC, and osteocyte (OCy) subpopulations for transcriptional analyses using Affmetrix Rat 230_2 microarrays

Genome-wide DNA methylation analysis of a cohort of 41 patients affected by Oculo-auriculo-vertebral spectrum (OAVS).

Project description:Oculo-auriculo-vertebral spectrum (OAVS; OMIM 164210) is a rare developmental disorder originating from abnormal development of the first and second branchial arch. The clinical phenotype is extremely heterogeneous with ear anomalies, hemifacial microsomia, ocular defects, and vertebral malformations being the main features. MYT1, AMIGO2 and ZYG11B gene variants having been reported in a few OAVS patients, but the etiology of the disease remains largely unknown. A multifactorial origin of the OAVS has been proposed, including the involvement of environmental or epigenetic mechanisms. To identify the epigenetic mechanisms contributing to OAVS, we evaluated the DNA-methylation profile of 41 affected patients by using a BeadChip 450K genome-wide approach. The analysis was first carried out at a group level by comparing the OAVS group methylation profile to the control group (n=48) revealing a slight epigenetic variation in a number of genes implicated in biological processes involving basic chromatin dynamics such as DNA packaging, chromatin assembly, protein-DNA organization, and gene silencing in patients compared to controls . A subsequent analysis focused on searching for Stochastic Epigenetic Variations (SEV) in individual profiles identified an increased number of SEV in OAVS patients compared to controls. Although no recurrent deregulated enriched region was evidenced, isolated patients harboring suggestive epigenetic variations, including changes in imprinted domains, were identified. The recognition of a different DNA methylation pattern in OAVS patients and the identification of isolated patients with suggestive epigenetic variations provide consistent evidences for the contribution of epigenetic mechanisms to the etiology of this complex and heterogeneous disorder.
