Differentially expressed mRNAs in lungs between hypoxic human umbilical cord mesenchymal stromal cell-derived extracellular vesicles (Hypo-EVs)-INH-treated mice and OVA-induced asthma mice

ABSTRACT: To better understand the molecular processes of nebulized Hypo-Evs on asthma intervention, OVA-induced chronic asthma model were established (OVA group), and a therapeutic regimen was instigated by inhaling Hypo-EVs four times (Hypo-EVs group) . Then a global RNA sequencing of mouse lung from the OVA and Hypo-EVs groups was performed.

We used microarrays to detail the global program of gene expression in asthma models and identify miRNAs that are differentially expressed in this process.

Project description:Atopic asthma is a chronic inflammatory disease of the lungs that is commonly associated with a Th2 response. The role of allergen-specific IgG in the initiation and development of allergic airway inflammation is still poorly understood; however, a receptor of IgG-immune complexes, CD16, has been demonstrated to promote augmentation of Th2 responses. To identify what genes downstream of CD16 signaling may be contributing to development of a Th2 response, we use ovalbumin (OVA) as our model antigen and compared wildtype and CD16-/- BMDCs that were treated overnight with OVA or OVA-immune complex. C57Bl/6 and CD16-/- BMDCs were treated for 24 hours with OVA or OVA-immune complex and then analyzed for gene expression changes.

We found that free feeding of an ODNcap (a CpG-ODN-embedded particle) -containing feed (ODNcap-F) prophylactically attenuates allergic airway inflammation, hyperresponsiveness, and goblet cell hyperplasia in an ovalbumin (OVA) -induced asthma model. To seek the suppressive mechanism of action of ODNcap-F in OVA-induced airway insults, we analyzed the lung transcriptome using DNA microarray analysis.

Project description:Intercellular communication orchestrates effective immune responses against disease-causing agents. Extracellular vesicles (EVs) are potent mediators of cell-cell communication. EVs carry bioactive molecules, including microRNAs, which modulate gene expression and function in the recipient cell. Here, we show that formation of cognate primary OVA-specific T-B lymphocyte immune contacts promotes transfer to the B cell of a very restricted set of OTII-derived CD4+ T cell exosomal microRNAs (mmu-miR20a-5p, mmu-miR-25-3p and mmu-miR-155-3p). To discriminate exogenously transferred microRNAs from microRNAs induced in B cells after immune synaps, small RNAseq of CD19Cre/Ki DICERfl/fl (DICER-KO) B cells, virtually lacking mature microRNAs, alone and cocultured with OTII CD4+T cells in the presence and absence of OVA was performed; (N=3) independent experiments
